Prefrontal Cortex Cytosolic Proteome and Machine Learning-Based Predictors of Resilience toward Chronic Social Isolation in Rats

AB  - Chronic social isolation (CSIS) generates two stress-related phenotypes: resilience and susceptibility. However, the molecular mechanisms underlying CSIS resilience remain unclear. We identified altered proteome components and biochemical pathways and processes in the prefrontal cortex cytosolic fraction in CSIS-resilient rats compared to CSIS-susceptible and control rats using liquid chromatography coupled with tandem mass spectrometry followed by label-free quantification and STRING bioinformatics. A sucrose preference test was performed to distinguish rat phenotypes. Potential predictive proteins discriminating between the CSIS-resilient and CSIS-susceptible groups were identified using machine learning (ML) algorithms: support vector machine-based sequential feature selection and random forest-based feature importance scores. Predominantly, decreased levels of some glycolytic enzymes, G protein-coupled receptor proteins, the Ras subfamily of GTPases proteins, and antioxidant proteins were found in the CSIS-resilient vs. CSIS-susceptible groups. Altered levels of Gapdh, microtubular, cytoskeletal, and calcium-binding proteins were identified between the two phenotypes. Increased levels of proteins involved in GABA synthesis, the proteasome system, nitrogen metabolism, and chaperone-mediated protein folding were identified. Predictive proteins make CSIS-resilient vs. CSIS-susceptible groups linearly separable, whereby a 100% validation accuracy was achieved by ML models. The overall ratio of significantly up- and downregulated cytosolic proteins suggests adaptive cellular alterations as part of the stress-coping process specific for the CSIS-resilient phenotype.

AB  - Magnesium titanate ceramics were prepared by reactive spark plasma sintering (SPS) at 1200 °C for 5 min. Prior to sintering, MgO and TiO2 powders were mixed by high energy ball milling (HEBM) for 15, 30, or 60 min. The effect of milling time on phase composition was analyzed by X-ray diffraction (XRD) for milled powders and sintered specimens. The morphology of the sintered ceramics was investigated by scanning electron microscopy (SEM), while elemental distribution was determined by energy dispersive spectroscopy (EDS). The presence of the MgTi2O5 phase was detected in XRD and was confirmed by EDS analysis. Microcracking was observed for specimens prepared with longer milling times and attributed to the coefficient of thermal expansion (CTE) mismatch dominantly due to the anisotropic crystal structure of MgTiO3 phase. The sample milled for 15 min showed the highest Vickers hardness due to less or no microcracking and nearly full density. The dielectric measurements in the range of 10 Hz–250 MHz demonstrated extremely high dielectric permittivity, as high as 104 at 1 kHz. An increase in loss tangents was observed due to oxygen vacancy formation, which was promoted by the vacuum environment during SPS. Oxygen vacancy-related dielectric relaxation was also detected and explained.

AB  - ZnO-based heterostructures are up-and-coming candidates for terahertz (THz) optoelectronic devices, largely owing to their innate material attributes. The significant ZnO LO-phonon energy plays a pivotal role in mitigating thermally induced LO-phonon scattering, potentially significantly elevating the temperature performance of quantum cascade lasers (QCLs). In this work, we calculate the electronic structure and absorption of ZnO/ZnMgO multiple semiconductor quantum wells (MQWs) and the current density–voltage characteristics of nonpolar m-plane ZnO/ZnMgO double-barrier resonant tunnelling diodes (RTDs). Both MQWs and RTDs are considered here as two building blocks of a QCL. We show how the doping, Mg percentage and layer thickness affect the absorption of MQWs at room temperature. We confirm that in the high doping concentrations regime, a full quantum treatment that includes the depolarisation shift effect must be considered, as it shifts mid-infrared absorption peak energy for several tens of meV. Furthermore, we also focus on the performance of RTDs for various parameter changes and conclude that, to maximise the peak-to-valley ratio (PVR), the optimal doping density of the analysed ZnO/Zn88Mg12O double-barrier RTD should be approximately 1018 cm−3, whilst the optimal barrier thickness should be 1.3 nm, with a Mg mole fraction of ~9%.

AB  - We present our progress on the “Multi-SENSor SysteM and ARTificial intelligence in service of heart failure diagnosis (SensSmart)” project, which was introduced at the last year’s edition of the Workshop [1]. The goal of the SensSmart project is to enable early diagnosis of heart failure, through the development of: 1) a multi-sensor polycardiograph apparatus (PCG) that produces simultaneous acquisition of the subject’s electrocardiogram (ECG), photoplethysmogram (PPG), heart sounds, and heart movements, and 2) AI-assisted analysis of the acquired signals. This presentation is going to focus on the acquisition and processing of PPG signals. PPG is obtained by using a pulse oximeter which illuminates the skin and measures the changes in light absorption, thereby enabling the detection of blood volume changes in the vessels. Our PCG apparatus measures the blood flow through the brachial, radial, and carotid arteries. During each heartbeat, the generated waveform typically exhibits several characteristic points [2]. The magnitudes and time distances between these points are useful indicators of many cardiac conditions, including heart failure [3]. However, the inter-patient variability of the PPG waveform makes it challenging to derive simple rule-based diagnostic procedures. This has led many researchers to turn to statistical or machine learning methods for processing of PPG signals [4].  In this presentation, we give an overview of AI-based signal processing methods for PPG, and present some preliminary results and challenges in extracting features from real-world signals obtained using our PCG.

AB  - In this contribution, we present our recent work on modeling intersubband transitions in the conduction band of semiconductor-based quantum well structures [1], [2]. Particularly interesting are possibilities offered by ZnO/ZnMgO and La-doped BaSnO3/BaO perovskite-oxide for the realization of room temperature oxide-based THz quantum well optoelectronic devices due to their advantageous physical and chemical properties [3], [4]. The electronic structure is calculated self-consistently by solving the Schrödinger–Poisson system of equations. A significant change of the transition energy due to the depolarization shift is also considered in cases when high doping is present. The charge-induced coherence due to the strong dipole-dipole Coulomb interaction between intersubband transitions leads to the formation of multisubband plasmons and a complete quantum model [5] based on the dipole representation must be used to calculate absorption spectra.

AB  - Ceramic/polymer composites can be chemically stable, mechanically strong, and flexible, which make them candidates for electric devices, such as pressure or temperature sensors, energy storage or harvesting devices, actuators, and so forth. Depending on the application, various electrical properties are of importance. Polymers usually have low dielectric permittivity, but increased dielectric permittivity can be achieved by the addition of the ceramic fillers with high dielectric constant. With the aim to enhance dielectric properties of the composite without loss of flexibility, 5 wt% of BaTiO3-Fe2O3 powder was added into a polyvinylidene fluoride matrix. The powder was prepared by different synthesis conditions to produce core/shell structures. The effect of the phase composition and morphology of the BaTiO3-Fe2O3 core/shell filler on the structure and lattice dynamics of the polymer composites was investigated. Based on the results of the thermal analysis, various parameters of ceramic/polymer composites were determined. Differences in the phase composition and morphology of the filler have an influence on the formation of various polyvinylidene fluoride allomorphs and the degree of crystallinity. Furthermore, the dielectric performances of pure polyvinylidene fluoride and the polymer/ceramic composites were measured.

Advantages and limitations of active phase silanization in PVDF composites: Focus on electrical properties and energy harvesting potential

AB  - In order to further improve the performance of 0.94[(Bi0.5Na0.5)TiO3]-0.06BaTiO3/polyvinylidene fluoride (NBT-BT/PVDF) flexible composite films prepared by the hot-pressing method, the effect of surface modification of the NBT-BT particles on the structure and properties of the films was investigated. Two coupling agents, namely, (3-aminopropyl)triethoxysilane (APTES) and dodecyl triethoxysilane (DDTES) were added to enhance dispersion and interfacial adhesion of the active phase powder with the polymer matrix. The highest amount of the electroactive PVDF β-phase was formed in APTES-modified samples while in DDTES samples mainly γ-phase was formed as shown by Fourier-transform infrared spectroscopy analysis. Differential scanning calorimetry measurements indicated that the addition of filler particles reduced the total crystallinity degree of the PVDF. Dielectric permittivity values as well as dielectric losses decreased for silanized samples due to reduced tension at the interface between particles and polymer. Strong intermolecular interaction between the PVDF chains and the APTES-modified particles led to enhanced breakdown strength of these samples. The highest level of agglomeration in the DDTES-modified samples induced the deterioration of ferroelectric properties. The highest voltage output of ~15 V and 225 μW of power was obtained for the APTES-modified harvester, evidencing their potential for energy harvesting applications.

AB  - For the last several decades, energy harvesters based on piezoelectricity from mechanical vibration have emerged as very promising devices that are being explored extensively for their functionality in energy technologies. In this paper, a series of flexible lead-free BaZr0.2Ti0.8O3 (BZT)/PVDF and lead-based PbZr0.52Ti0.48O3 (PZT)/PVDF piezocomposites with variable filler content up to 50 vol% were prepared by a hot pressing method. The structure and morphology of the BZT and PZT powders, as well as the distribution of the piezo-active filler in the obtained flexible films were characterized by XRD and SEM analysis. In addition, the remnant polarization (Pr) and leakage current were also investigated to evaluate the breakdown strength in both types of flexible films. The calculations of storage energies and output voltage obtained for the investigated materials revealed an increasing trend with an increasing amount of BZT and PZT active phases. The maximum storage energy of 0.42 J/cm3 (and energy efficiency of 40.7 %) was obtained for the PZT–PVDF (40–60) films, while the maximum output voltage of about 10 V (~10 µA) was obtained for the PZT–PVDF (50–50) flexible film. In addition, a comparison between the properties of the lead-based and lead-free flexible films, as well as the potential use of these films as energy storage and energy harvesting systems were analyzed.
