AB - In this study, graphene samples prepared by mechanical exfoliation were examined by Raman spectroscopy. The Au electrical contacts, fabricated using photolithography, allowed the application of a gate voltage between graphene and the Si substrate. In the Raman spectra of the sample, we observed shifts of position, changes of intensity and the width variations of the G and 2D peaks with the change of the gate voltage. Spatial Raman mapping of the samples was performed showing variations in intensities of the Raman peaks in different flake regions.

AB - A prototype of the safety system for the electromagnetic calorimeter (ECAL) of the CMS detector at LHC has been implemented and tested in Super Modules SM0 and SM1 during the ECAL test-beam campaign in summer 2003. The aim was to examine the system performance thoroughly and to resolve several open issues regarding the final system design. The performed tests mainly focused on the readout System performance, its calibration and noise problems. Furthermore, irradiation tests of the readout electronics components with a 64 MeV proton beam Lip to an equivalent cumulative dose of 200 kGy and 3.2 x 10(14) neutron cm(-2) have been performed at the Paul Scherrer Institute in spring 2004. The results of all these tests are very promising, leading to the conclusion that the tested prototype offers an excellent basis for the development of the safety system for the entire CMS ECAL. AB - In the present study a polystyrene microtiter plate was tested as a support material for synaptic plasma membrane (SPM) immobilization by adsorption. The adsorption was carried out by an 18-h incubation at +4degreesC of SPM with a polystyrene matrix, at pH 7.4. Evaluation of the efficiency of the applied immobilization method revealed that 10% protein fraction of initially applied SPM was bound to the support and that two SPM enzymes, Na+/K+-ATPase and Mg2+-ATPase, retained 70-80% activity after the adsorption. In addition, adsorption stabilizes Na+/K+-ATPase and Mg2+-ATPase, since the activities are substantial 3 weeks after the adsorption. Parallel kinetic analysis showed that adsorption does not alter significantly the kinetic properties of Na+/K+-ATPase and Mg2+-ATPase and their sensitivity to and mechanism of Cd2+- or Hg2+- induced inhibition. The only exception is the high affinity Mg2+-ATPase moiety, whose affinity for ATP and sensitivity toward Cd2+ were increased by the adsorption. The results show that such system may be used as a practical and comfortable model for the in vitro toxicological investigations. (C) 2001 Elsevier Science.

AB - Enzymatic activities of Na+/K+-ATPase and Mg2+-ATPase from rat brain synaptic plasma membrane were studied in the absence and presence of EDTA. The aim of the study was to examine the ability of this strong chelator to prevent and recover the CuSO4-induced inhibition. The influence of experimentally added CuSO4 and EDTA on MgATP(2-) complex and free Cu2+ concentrations in the reaction mixture was calculated and discussed. CuSO4 induced dose-dependent inhibition of both enzymes in the absence and presence of 1 mM EDTA. In the absence of EDTA, the IC50 values of Cu2+, as calculated from the experimental curves, were 5.9 x 10(-7) M for Na+/K+- ATPase and 3.6 x 10(-6) M for Mg2+-ATPase. One millimolar EDTA prevented the enzyme inhibition induced by CuSO4, but also reversed the inhibited activity, in a concentration-dependent manner, following exposure of the enzymes to the metal ion, by lowering free Cu2+ concentration. Kinetic analysis showed that CuSO4, inhibits both the Na+/K+-ATPase and Mg2+-ATPase, by reducing their maximum enzymatic velocities (V-max), rather than apparent affinity for substrate MgATP(2-) (K-0.5), implying the noncompetitive nature of enzyme inhibition induced by the metal. The kinetic analysis also confirmed two distinct Mg2+-ATPase subtypes activated in the presence of low and high MgATP(2-) concentrations. K-0.5 and V-max were calculated using a computer-based program. The results of calculation showed that MgATP(2-) concentration in the kinetic experiments exceeded three times the apparent K-0.5 value for the enzyme activation.
