Improving gallic acid detection in plant samples: Fabrication and optimization of a sensitive and selective NiO-supported carbon paste electrode

AB  - In this study, we successfully prepared a modified nickel oxide (NiO) carbon paste electrode to detect gallic acid (GA). NiO nanoparticles were synthesized by the simple, organic solvent-free chemical coprecipitation method, and the electrochemical properties of the electrode and GA were thoroughly investigated using CV, SWV, and EIS, while morphological properties were examined using ICP-OES, TEM, SEM, and XRD. Excellent catalytic characteristics are displayed by the developed material which facilitates the interaction of the target with the electrode surface. The obtained electrochemical information showed that the incorporation of NiO nanoparticles to the carbon paste electrode effectively facilitates electron transfer processes and enriches the catalytic response of the carbon paste electrode. The fabricated NiO/CPE sensor showed a satisfactory linear relationship between peak current and GA concentration in the broad range of 0.2–100 μM and 100–200 μM with a low detection limit of 0.04 μM and limit of quantification of 0.12 μM at pH 3 of BRBS as supporting electrolyte. The selectivity of the proposed method was satisfactory, with acceptable stability, considerable repeatability, and accurate reproducibility. Moreover, the good practicability performance could be effectuated at the NiO/CPE sensor for the quantitative analysis of GA in bourtree, walnut, primrose, and chamomile tea samples. The results were compared with the standard DPPH test and statistical processing of the results was performed, which confirmed the excellent agreement between the two methods. The developed method can provide a cost-effective, rapid, selective, and sensitive means for GA monitoring. When compared to other works, the developed technique has a wider linear range and lower LOD and LOQ, which makes this work a very important reference for the highly sensitive analysis of GA in the field of food safety.

The Profile and Antimicrobial Activity of Bacillus Lipopeptide Extracts of Five Potential Biocontrol Strains

AB  - In this study the efficacy of two different methods for extracting lipopeptides produced by five Bacillus strains-ethyl acetate extraction, and acid precipitation followed by methanol extraction-was investigated using mass spectrometry. High performance thin layer chromatography (HPTLC) was also used for the simultaneous separation of complex mixtures of lipopeptide extracts and for the determination of antimicrobial activity of their components. The mass spectra clearly showed well-resolved groups of peaks corresponding to different lipopeptide families (kurstakins, iturins, surfactins, and fengycins). The ethyl acetate extracts produced the most favorable results. The extracts of SS-12.6, SS-13.1, and SS-38.4 showed the highest inhibition zones. An iturin analog is responsible for the inhibition of Xanthomonas arboricola and Pseudomonas syringae phytopathogenic strains. HPTLC bioautography effectively identified the active compounds from a mixture of lipopeptide extracts, proving in situ its potential for use in direct detection and determination of antimicrobials. In the test of potential synergism among individual extracts used in different mixtures, stronger antimicrobial effects were not observed. Biochemical and phylogenetic analysis clustered isolates SS-12.6, SS-13.1, SS-27.2, and SS-38.4 together with Bacillus amyloliquefaciens, while SS-10.7 was more closely related to Bacillus pumilus.

AB  - Propolis, as a beneficial natural product, has found application in the food and pharmaceutical industries as a food preservative, active packaging material as well as a main component of propolis containing products. Quality control of propolis has thus gained in importance. We used simple, fast, and low-cost techniques such as UV/Vis spectrophotometry and cyclic voltammetry applied in combination with principal component analysis to confirm the presence of two botanical sub-types of propolis. Cyclic voltammetry, as a green, sensitive, and convenient technique suitable for investigation of chemical profile, together with multivariate chemometrics technique, was used for the first time for characterization and classification of propolis. UV/Vis spectroscopy was applied in two aspects, absorption at certain wavelength for the quantitative determination of analytes, i.e. determination of quality control parameters, and spectral profiles, for the confirmation of the existence of two botanically different poplar types. All investigated Serbian propolis samples were characterized with specific chemical patterns and were classified in two main botanical varieties, so-called orange and blue. The results confirmed that the applied methods have a potential to be used for the discrimination of complex natural products such as propolis.
