abstract = "In spite of numerous advantages, there are serious drawbacks ofthe application of matrix assisted laser desorption/ionizationtime-of-flight mass spectrometry (MALDI TOF MS) for small-molecule analyses (below 500 Da) and quantification. The mainproblem is the background interference from commonly usedMALDI matrix materials. The aim of this work is to evaluateMALDI TOF mass spectra of physiologically relevant smallmolecules: steroid hormones, vitamins, amino acids and carbohy-drates, acquired with several organic, traditional matrices.Small volume, 0.5 lL, of each sample solution (testosterone,progesterone, estradiol, L-cysteine, L-alanine, DL-methionine,glutathione, D-(+)-glucose, D-(+)-maltose, vitamin A, vitamin E)was mixed on the sample plate with the same volume of organicmatrix solutions (DHB, THAP, CHCA, 9-AA). For each mole-cule/matrix pair, we determined quantitative and qualitativeparameters of MS analysis. To calculate within day and day-to-day variation we used Excel tools (ANOVA tests). In addition,homogeneity of the sample/matrix distribution on the target wasalso calculated and expressed as the coefficient of variation of aseries of measurements.Our results show selectivity of the detection of individualmolecules related with the matrix applied. The statistical analysisof certain molecule/matrix pairs gave within and day-to-day vari-ations less than 15%. Additionally, homogeneity of the sample/matrix mixture distribution on the target plate was with somematrices, also less than 15%.Some of the used matrices have a great potential for the anal-ysis of small molecules with good analytical parameters, with lowvariations and high homogeneity of samples on the MALDI tar-get plate.These results hold potential for quantification of metaboli-cally-significant small molecules and are very promising forfuture applications of MALDI TOF MS analyses.",
