The role of transferrin receptor 1 in HCV infection

Transferrin receptor 1 (TfR1) is key molecule involved in the accumu-lation of hepatic iron in chronic hepatitis C (HCV). TfR1 is constitutivelyclustered in coated pits and can be rapidly internalized, transported to theacidic endosome, and recycled back to the cell surface. It was alsoproposed that TfR1 is involved in infection of hepatocytes by participa-tion in internalization of E2 protein – major receptor binding protein ofthe HCV. We investigated the role of TfR1 in HCV cell entry, using theinformational spectrum method (ISM), a virtual spectroscopy method forstructure/function analysis of nucleotide and protein sequences. Resultsof the ISM analysis indicate (i) that TfR1 is involved in viral entry viaclatrin associated endocytosis, and (ii) the direct interaction betweenextracellular domain of TfR1 and E2 protein. These results suggestpossible additional role of TfR1, beyond the role in internalization, inHCV receptor cell entry.
