AB - An efficient encapsulation of the fullerene into two hydrophobized and one native polysaccharide provided water soluble supramolecular hybrids. After covalent modification of polysaccharides by cholesterol, noncovalent hybrids were prepared by a three-step procedure, including mixing of individual aqueous solutions of hydrophobized, as well as native sugar with solution of the fullerene in pyridine, dialysis and lyophilization. Although the degree of the fullerene incorporation into hydrophobized substrates, cholesterol-levan and cholesterol-pullulan, was lower in comparison to the native polysaccharide levan, hydrophobization provided nanoparticles with improved properties. The particle size distribution, studied by dynamic light scattering and scanning electron microscopy revealed formation of moderately polydisperse aggregates, with the diameter contraction in comparison to the corresponding free polysaccharide, especially in the case of hydrophobized substrates. The morphological examination, done by scanning electron microscopy indicated the self-organization of the fullerene-native polysaccharide to round individual structures, while fullerene-hydrophobized polysaccharide hybrids tend to form networks. The antioxidant activity of the synthesized polysaccharide-C60 noncovalent hybrids versus starting polysaccharides was investigated by the DPPH radical scavenging and the β-carotene-linoleic acid bleaching methods. In all three complexes, the radical scavenging ability of the fullerene remained preserved, and a positive effect of levan hydrophobization was observed.

Optimized reaction conditions, spectroscopic and electrochemical characteristics of the reaction products are provided. The X-ray structural analyses of three cycloadducts were done and obtained results were compared with properties of reported N,N'-bicyclic systems those displayed conformational similarity. Antibacterial activities of all products against two Gram-positive and two Gram-negative bacterial strains were evaluated by the experimental methods. (c) 2018 Elsevier B.V. All rights reserved.

AB - A series of ferrocenes which contain dinitrogen-fused pyrazolidinone ring were synthesized from acryloylferrocene (4) and N,N′‑cyclic azomethine imines (3). Novel 5‑aryl‑6‑ferrocenoyltetrahydropyrazolo[1,2‑a]pyrazol‑1(5H)-ones were obtained as mixtures of two diastereoisomers (trans and cis) which were separated and isolated as pure substances. Ortho-substituted N,N′‑cyclic azomethine imines 5-oxo-2-(2,4,6-trimethylbenzylidene)pyrazolidin-2-ium-1-ide (3e) and 2-(2-methoxybenzylidene)-5-oxopyrazolidin-2-ium-1-ide (3f) reacted stereoselectively affording only trans-6-ferrocenoyl-5-mesityltetrahydropyrazolo[1,2-a]pyrazol-1(5H)-one (5e) and 6-ferrocenoyl-5-(2-methoxyphenyl)tetrahydropyrazolo[1,2-a]pyrazol-1(5H)-one (5f). Ferrocenyl derivatives were screened for in vitro antioxidant and antifungal activities and excellent DPPH and ABTS radicals scavenging activity was observed with majority of tested 5‑aryl‑6‑ferrocenoyltetrahydropyrazolo[1,2‑a]pyrazol‑1(5H)-ones. Several tested compounds showed selective scavenging properties neutralizing ABTS•+ radical cations in contrast to inactivity toward DPPH[rad] radicals. Trans-5-aryl-6-ferrocenoyltetrahydropyrazolo[1,2-a]pyrazol-1(5H)-ones 5b, 5c, 5d, 5j as well as cis-6-ferrocenoyl-5-(p-tolyl)tetrahydropyrazolo[1,2-a]pyrazol-1(5H)-one (6d) displayed fungal growth inhibition at low concentration against C. albicans and/or A. brasiliensis. Molecular docking studies revealed that the cis-6-ferrocenoyl-5-(4-nitrophenyl)tetrahydropyrazolo[1,2-a]pyrazol-1(5H)-one (6l) and cis-6-ferrocenoyl-5-(naphthalen-2-yl)tetrahydropyrazolo[1,2-a]pyrazol-1(5H)-one (6n) have potential to become lead molecules in drug discovery process. © 2018 Elsevier Inc.

AB - Cerium oxide (CeO2) nanoparticles (CONPs) are interesting biomaterials with various applications in biomedicine, cosmetics and the pharmaceutical industry, but with limited practical application because of their low stability in aqueous media. The aim of this study was to obtain CONPs with increased stability by coating the particles. Microbial exopolysaccharides (levan, pullulan) and glucose were used to prepare CONPs under different synthesis conditions. Coating was attempted by adding the carbohydrates during (direct coating) or after (subsequent coating) the synthesis of CONPs. The obtained nanoparticles were characterized by X-Ray diffraction analysis, Fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM) and transmission electron microscopy (TEM). The suspension stability of the uncoated and coated CONPs in aqueous media was evaluated by measuring the hydrodynamic size, zeta potential and turbidity. The FT-IR spectra revealed the differences between coated CONPs and showed the success of subsequent coating with carbohydrates. Coating with carbohydrates improved the stability the CONP suspension by decreasing the size of aggregated particles. The suspensions of levan- and glucose-coated CONPs had the best stability. In this study, CONPs were prepared using non-toxic materials, which were completely environmentally friendly. The obtained results open new horizons for CONP synthesis, improving their biological applications.

AB - Commercially available pesticides were examined as Mus musculus and Homo sapiens acetylcholinesterase (mAChE and hAChE) inhibitors by means of ligand-based (LB) and structure-based (SB) in silico approaches. Initially, the crystal structures of simazine, monocrotophos, dimethoate, and acetamiprid were reproduced using various force fields. Subsequently, LB alignment rules were assessed and applied to determine the inter synaptic conformations of atrazine, propazine, carbofuran, carbaryl, tebufenozide, imidacloprid, diuron, monuron, and linuron. Afterwards, molecular docking and dynamics SB studies were performed on either mAChE or hAChE, to predict the listed pesticides’ binding modes. Calculated energies of global minima (Eglob_min) and free energies of binding (∆Gbinding) were correlated with the pesticides’ acute toxicities (i.e., the LD50 values) against mice, as well to generate the model that could predict the LD50s against humans. Although for most of the pesticides the low Eglob_min correlates with the high acute toxicity, it is the ∆Gbinding that conditions the LD50 values for all the evaluated pesticides. Derived pLD50 = f(∆Gbinding) mAChE model may predict the pLD50 against hAChE, too. The hAChE inhibition by atrazine, propazine, and simazine (the most toxic pesticides) was elucidated by SB quantum mechanics (QM) DFT mechanistic and concentration-dependent kinetic studies, enriching the knowledge for design of less toxic pesticides.

AB - The electrochemical techniques were used to determine the total antioxidant capacity of breast milks and the results were compared with a commonly used spectrophotometric (2,2-diphenyl-1-picrylhydrazyl, DPPH) method. Breast milk from mothers of preterm infants was monitored in three lactation phases and after storage of expressed milk by monitoring changes in the total antioxidant capacity over a two year period. Statistical analysis showed there was no significant difference between the ability of the three methods to detect changes in breast milk after storage. Either of the electrochemical techniques studied could be successfully used to replace the time-consuming spectrophotometric method and can be applied to clinical trials.

AB - The aim of this paper was to investigate whether changes in the quantity and structure of humic acids (HA) occur during bioremediation of waste mazute (heavy residual fuel oil). The effects of humification on the structure of HA were evaluated by Fourier Transform Infrared Spectroscopy (FT-IR), potentiometric titrations, the ratio of absorbances at 465 and 665 nm (E4/E6 ratio) and the ratio. Also, the quality of humification was monitored by determining parameters which are important for measuring the evolution of humic substances (HS), such as CHA/CFA (the humic acidifulvic acidratio), the rate of extraction (TE) and the index of polymerization (IP). An increase of the content of HA from 0.23% to 0.70% was observed during the bioremediation process. The FT-IR spectra and 04 ratio of HA extracted at the beginning and the end of the process indicate structural changes during the bioremediation process. The groups containing aromatic and carboxylic carbon increased, resulting in HA structures of higher aromaticity. An increase of the B4/E6 ratio during bioremediation treatment implied the enrichment of HA with functional oxygen groups. Potentiometric titrations of HA solution showed increases in the buffering and the redox capacities of HA during the bioremediation process. The results of parameters of humification showed an increase of the compositional carbon in HA at the end, compared to at the start, of bioremediation, while the carbon that belonged to fulvic acids (FA) reduced by 44%. Transformation into HA is the likely cause of this observed reduction. The TE and the IP demonstrated that humification occured during the bioremediation process. According to the available data, this is the first report on synthesis of HS during bioremediation of petroleum hydrocarbons. (C) 2017 Published by Elsevier Ltd.

AB - Iron and sulfur oxidizing chemolithoautotrophic acidophilic bacteria, such as Acidithiobacillus species, hold the dominant role in mine environments characterized by low pH values and high concentrations of reduced sulfur and iron compounds, such as ores, rocks and acid drainage waters from mines. On the other hand, heterotrophic microorganisms, especially their biofilms, from these specific niches are receiving increased attention, but their potential eco-physiological roles have not been fully understood. Biofilms are considered a threat to human health, but biofilms also have beneficial properties as they are deployed in waste recycling and bioremediation systems. We have analyzed interactions of the metal tolerant heterotrophic microorganisms in biofilms with iron oxidizing autotrophic bacteria both from the sulphidic mine environment (copper mine Bor, Serbia). High tolerance to Cu2+, Cd2+ and Cr6+ and the presence of genetic determinants for the respective metal tolerance and biofilm-forming ability was shown for indigenous heterotrophic bacteria that included strains of Staphylococcus and Rhodococcus. Two well characterized bacteria-Pseudomonas aeruginosa PAO1 (known biofilm former) and Cupriavidus metallidurans CH34 (known metal resistant representative) were also included in the study. The interaction and survivability of autotrophic iron oxidizing Acidithiobacillus bacteria and biofilms of heterotrophic bacteria during co-cultivation was revealed. Finally, the effect of heterotrophic biofilms on bioleaching process with indigenous iron oxidizing Acidithiobacillus species was shown not to be inhibitory under in vitro conditions. AB - The impact of the surface charge of alumina supports on the adsorption of Candida rugosa lipase was investigated in terms of the zeta potentials of the adsorption partners. The lipase adhered onto alumina with similar efficiency under both repulsive and attractive electrostatic conditions, shifting the zeta potential of the support towards that of the enzyme. The behavior was explained by a heterogeneous distribution of the surface charge of the lipase molecule. Special emphasis in this study was placed on the effect of immobilization on the enzyme kinetics and principal reasons for enzyme immobilization: improvement in stability and potential for reuse. The enzyme affinity was not altered by its adsorption onto alumina, while the Vmax value of the lipase decreased. The thermostability of the adsorbed lipase was improved. A significant potential for reuse was found.

AB - Lipase from Candida rugosa was immobilized by adsorption onto laboratory prepared supports, silica SBA-15 and zirconia. The adsorption process was studied as a function of pH in terms of percent of adsorbed lipase, enzyme activity and zeta potential of support and enzyme. Several analytical approaches such as laser Doppler electrophoresis, Fourier transform infrared spectroscopy (FTIR) and field emission scanning electron microscopy (FESEM) showed that the lipase was successfully immobilized onto both supports. The zeta-potential data suggest that the adsorption efficiency does not depends on the sign but on the magnitude of the surface charge of adsorption partners, and therefore underline the importance of their dispersion stability. Adsorption to material surface altered enzyme characteristics. nu(max), for the lipase immobilized onto silica and zirconia were 4.8-fold and 3.6-fold lower than that of the free lipase, respectively. The Km showed no alteration of enzyme-substrate affinity on zirconia support, whereas the enzyme immobilized on silica had 3.6 times lower affinity. Thermostability at 60 degrees C of the lipase was improved 12-fold on zirconia and 4-fold on silica. Finally, in examining reusability, the immobilized lipase retained more than 90% of initial activity after eight reuses on both supports. (C) 2014 Elsevier B.V. All rights reserved.

AB - Perfluoroalkyl sulfonates (PFSAs) and perfluoroalkyl carboxylates (PFCAs) were analyzed in sediment samples from the wastewater canal draining the industrial complex of Pancevo, Serbia (oil refinery, petrochemical plant, and fertilizer factory). The canal is directly connected to Europes second largest river, the Danube, which drains its water into the Black Sea. Perfluorooctane sulfonate (PFOS) up to 5.7 ng d(-1) weight (dw) and total Perfluorinated compounds (PFCs) up to 6.3 ng g(-1) dw were detected. Compared to other reports, high levels of PFOS were found, even though PFCs are not used in the industrial production associated with this canal. The PFOS concentration in water was recalculated using the adsorption coefficient, K-oc from literature. Using the average output of wastewater from the canal, a mass load of 1.38 kg PFOS per year discharged in the Danube River has been calculated, which undoubtedly points to the contribution to global persistent organic pollution of surface waters originating from this industrial place.
