Hafnium-containing Wells-Dawson nanocluster as a promising contrast agent candidate

AB  - The purpose of this study was to evaluate in vitro contrast properties of a synthesized Wells-Dawson P2W17O61)2]·19H2O (Hf-WD 1:2), as a potential contrast agent for computed tomography (CT). X-ray attenuation was determined in vitro in the presence of increasing tungsten concentrations (3.125-100 mM) of Hf-WD 1:2. In order to compare the contrast properties of the tungsten-containing Hf-WD 1:2 with a commercially available CT contrast agent that has been used in clinical practice, X-ray attenuation was presented for the same iodine concentrations of iodine-based iohexol solution as well. Higher HU values for particular concentrations were obtained for Hf-WD 1:2 in comparison with those determined for the standard iohexol. Thus, the studied Hf-WD 1:2 POM could be considered a promising contrast agent candidate for CT. Nevertheless, further development of Hf-WD 1:2 as a potential CT contrast requires additional CT studies in vivo and relevant toxicity studies in vitro and in vivo as well.

AB  - In this study contrast properties for computed tomography (CT) of two tungstate-containing molecules differing in size, shape, and structure: Keggin type polyoxometalate, 12-tungstosilicic acid, WSiA and sodium tungstate, Na2WO4, were assessed in vitro. In this aim, X-ray attenuation was monitored in vitro at different tungsten concentrations in the range of 3.125-100 mM. A stronger X-ray attenuation was observed for Na2WO4 in the presence of the same tungsten concentrations. In addition, both Na2WO4 and WSiA demonstrated better contrast performance compared to iodine-based iohexol solution, which has been widely applied as a contrast agent for CT. Accordingly, tungsten-containing compounds deserve attention as promising new-generation CT contrast candidates. Thus, future studies should be directed to the synthesis of novel tungsten molecules of different structures resulting in appropriate contrast and low toxicity.

AB  - Polyoxotungstate nanoclusters have recently emerged as promising contrast agents for computed tomography (CT). In order to evaluate their clinical potential, in this study, we evaluated the in vitro CT imaging properties, potential toxic effects in vivo, and tissue distribution of monolacunary Wells–Dawson polyoxometalate, α2-K10P2W17O61.20H2O (mono-WD POM). Mono-WD POM showed superior X-ray attenuation compared to other tungsten-containing nanoclusters (its parent WD-POM and Keggin POM) and the standard iodine-based contrast agent (iohexol). The calculated X-ray attenuation linear slope for mono-WD POM was significantly higher compared to parent WD-POM, Keggin POM, and iohexol (5.97 ± 0.14 vs. 4.84 ± 0.05, 4.55 ± 0.16, and 4.30 ± 0.09, respectively). Acute oral (maximum-administered dose (MAD) = 960 mg/kg) and intravenous administration (1/10, 1/5, and 1/3 MAD) of mono-WD POM did not induce unexpected changes in rats’ general habits or mortality. Results of blood gas analysis, CO-oximetry status, and the levels of electrolytes, glucose, lactate, creatinine, and BUN demonstrated a dose-dependent tendency 14 days after intravenous administration of mono-WD POM. The most significant differences compared to the control were observed for 1/3 MAD, being approximately seventy times higher than the typically used dose (0.015 mmol W/kg) of tungsten-based contrast agents. The highest tungsten deposition was found in the kidney (1/3 MAD—0.67 ± 0.12; 1/5 MAD—0.59 ± 0.07; 1/10 MAD—0.54 ± 0.05), which corresponded to detected morphological irregularities, electrolyte imbalance, and increased BUN levels.

Exploring polyoxometalates as non-destructive staining agents for contrast-enhanced computed tomography

AB  - Due to the high complexity and heterogeneous structure of biological tissues, imaging techniques that allow for precise and quantitative structural analyses of such materials are of high importance. These techniques could also advance clinical translation of regenerative medicine by providing better insights in tissue development and disease. The standard techniques for evaluating biological tissues such as histological sectioning and staining, have a high discriminative power, but allow assessment of the tissue distribution only in two dimensions. Due to restricted sectioning orientation and limited depth resolution, this leads to loss of information in three dimensions (3D), and therefore, more precise imaging of the 3D microstructure and spatial interrelationships of the different tissues within organs is crucial. MicroCT can provide full 3D structural information of mineralized tissues and dense biomaterials. However, the intrinsic low X-ray absorption of soft tissues requires contrast-enhancing staining agents (CESAs) to be used. We have shown that a range of polyoxometalate clusters (POMs) can be excellent non-destructive staining agents for high-resolution contrast-enhanced microCT (CE-CT) visualization of various tissues, of bone and its marrow vascularization and adiposity. A range of Wells-Dawson POMs, differing in structure and overall charge, has been synthesized and evaluated for their potential as soft tissue CESAs. We have shown that hafnium-substituted POM (Hf-POM) allows for simultaneous contrast-enhanced microCT (CE-CT) visualization of bone and its marrow vascularization and adiposity. Monolacunary Wells-Dawson POM (Mono-WD POM) showed similar soft tissue enhancement as Hf-WD POM and phosphotungstic acid (PTA), a frequently used but destructive CESA. However, compared to PTA, the POMs are much less destructive and show a better diffusion. The solubility of Mono-WD POM can be improved by simple addition of lithium chloride to the staining solution, leading to further enhancement of the soft tissue contrast. In vivo toxicity of Wells-Dawson POM has been also evaluated according to standard toxicological protocols using Wistar albino rats, which is the first and important step in evaluating side effects of these polyoxometalate nanoclusters that show large potential as therapeutics and contrast agents.

Toksikološka ispitivanja tokom razvijanja novih lekova i medicinskih sredstava

AB  - Toksikološka ispitivanja predstavljaju osnovu prekliničkih ispitivanja novih lekova i medicinskih sredstava. Ovaj tip studija može predstavljati presudni korak u procesu procene daljeg razvoja lekova i medicinskih sredstava. Svedoci smo sve većeg interesovanja industrije i šire akademske zajednice za ovaj tip istraživanja, pa je upoznavanje sa vodičima za sprovođenje ovih studija, potpomognuto primerima iz prakse od ključnog značaja za njihovu edukaciju. Uzimajući sve ovo u obzir neophodno je da šira akademska zajednica bude bolje upoznata sa značajem ovih studija i pravilnim načinom njihovog sprovođenja. Od velikog je značaja upoznati zainteresovane strane o načinu sprovođenja toksikoloških studija, pre svega sa zakonskim okvirima koji se moraju poštovati u toku njihovog sprovođenja. Neophodno je zainteresovanim stranama iz šire akademske zajednice i industrije ukazati na etičke izazove na koje se nailazi tokom sprovođenja jedne toksikološke studije, pogotovu ako se u obzir uzme koliko su ovakva ispitivanja sa etičkog aspekta osetljiva. Postoji i potreba da se razjasni kako ispitivanje različitih toksikoloških aspekata zahteva i različite pristupe u sprovođenju ovih studija. Tako se npr. međusobno se razlikuju studije koje ispituju akutnu ili hroničnu toksičnost, studije u kojima se ispituju različiti načini primene (per os, intravenski,...) lekova i medicinskih sredstva i sl. Sve ove informacije bi trebalo da razjasne zainteresovanim stranama kako pravilno sprovesti toksikološke studije i kako iz njih dobiti relevantne rezultate, a da se pri tome ne krše zakonske odredbe niti etičke norme.

Ovaj tip studija može predstavljati presudni korak u procesu procene daljeg razvoja lekova i medicinskih sredstava. Svedoci smo sve većeg interesovanja industrije i šire akademske zajednice za ovaj tip istraživanja, pa je upoznavanje sa vodičima za sprovođenje ovih studija, potpomognuto primerima iz prakse od ključnog značaja za njihovu edukaciju. Uzimajući sve ovo u obzir neophodno je da šira akademska zajednica bude bolje upoznata sa značajem ovih studija i pravilnim načinom njihovog sprovođenja. Od velikog je značaja upoznati zainteresovane strane o načinu sprovođenja toksikoloških studija, pre svega sa zakonskim okvirima koji se moraju poštovati u toku njihovog sprovođenja. Neophodno je zainteresovanim stranama iz šire akademske zajednice i industrije ukazati na etičke izazove na koje se nailazi tokom sprovođenja jedne toksikološke studije, pogotovu ako se u obzir uzme koliko su ovakva ispitivanja sa etičkog aspekta osetljiva. Postoji i potreba da se razjasni kako ispitivanje različitih toksikoloških aspekata zahteva i različite pristupe u sprovođenju ovih studija. Tako se npr. međusobno se razlikuju studije koje ispituju akutnu ili hroničnu toksičnost, studije u kojima se ispituju različiti načini primene (per os, intravenski,...) lekova i medicinskih sredstva i sl. Sve ove informacije bi trebalo da razjasne zainteresovanim stranama kako pravilno sprovesti toksikološke studije i kako iz njih dobiti relevantne rezultate, a da se pri tome ne krše zakonske odredbe niti etičke norme.",

waste warehouses or in radiation sterilization units. In the analysis of costs and benefits, several criteria are
used to evaluate the characteristics of concrete, so that, in addition to the mechanical ones, the radiation
characteristics of the building material are also taken into account. One of the most important characteristics
of radiation shielding concrete is its overall mass attenuation coefficient. In this paper, the XCOM computer
code was used to calculate the total mass attenuation coefficients in the energy range from 0.01 MeV to 100
MeV for three types of concrete: ultrahight properties concrete (UHPC), concrete with barite and concrete
with magnetite and steel. Based on the comparison of the calculation results, it was concluded that concrete
with magnetite and steel has a greater protective power than the other two types of concrete for gamma and X
radiation energy lower than 30 keV. Another important conclusion is that concrete with barite has a higher
protective power than the other two types of concrete in the photon energy range from 30 keV to 300 keV,
and for photon energy greater than 6 MeV. A detailed analysis of the calculation results revealed that in the
energy range from 400 keV to 6 MeV the values for the total mass attenuation coefficients are approximately
the same for three different types of concrete.

Based on engineering practice, it is necessary to compare different types of concrete used for the construction of facilities where there are sources of gamma and X radiation, such as in cases where radiation sources appear in medical facilities with linear accelerators, cyclotron installations for accelerating nuclear particles, in the environment of nuclear reactors, in radioactive waste storages, or in radiation sterilization unit s. In the cost-benefit analysis, several criteria are used to evaluate the characteristics of the concrete, so that in addition to the mechanical characteristics, the radiation characteristics of the construction material are also considered. One of the most important characteristics of concrete for radiation protection is its total mass attenuation coefficient. In this paper, computer code XCOM was used for the calculation of the total mass attenuation coefficients in energy range from 0.01MeV to 100 MeV for three types of concrete: ordinary concrete, concrete with barite and concrete with limonite and steel. Based on the comparison of calculation results, it was concluded that concrete with limonite and steel has greater protective power than other two concrete types for the gamma and X radiation energy less than 30 keV. The second important conclusion is that the concrete with barite has greater protective power than other two concrete types in the range of photon energy from 30 keV to 300 keV, and for photon energy greater than 6 MeV. A detailed analysis of the calculation results found that in the energy range of 400 keV to 6 MeV values for the total mass attenuation coefficients are approximately the same for three different types of concrete.

AB  - The paper reports on the composition of thirty-eight Late Roman glass fragments (3rd–4th century CE) from Viminacium, the capital of Moesia Province, and Egeta, the fort controlling Iron Gates Gorge on the Roman Danube Limes. The glasses are measured using simultaneous particle-induced X-ray emission (PIXE) and particle-induced gamma-ray emission (PIGE). The analysis shows that sixteen glasses belong to the Roman glass decolourized by antimony, nine to the Roman glass decolourized by manganese, and one is recycled using these two types. Five glasses belong to the Foy série 3.2, two to HIMT, one to Jalame type with manganese and one to the rare plant-ash type P-1, produced in Egypt, and for the first time reported from the continental Europe. The comparison of the finds with the contemporary glass from Serbia and from the wider Balkans shows a marked shift in glass trading patterns between the epochs of the High Empire and the Late Antiquity. During the High Empire, glass seems to be imported to the central and eastern Balkans mainly from the west via Roman road Aquileia–Emona–Siscia–Sirmium–Viminacium and perhaps via Adriatic ports, while in the Late Antiquity it was predominantly from east to west, over the Danube, Via Militaris or Aegean ports. Another find is that the richness of the glass market in Viminacium indicates that the capital of Moesia province on the Danube limes was a cosmopolitan city.

AB  - A comparative study of performances of self compacting concrete (SCC) with fly ash and limestone filler was conducted. The experimental program included the production of reference SCC designed with Portland cement, river sand and limestone filler and four experimental self compacting concretes with different types of fine aggregates, fillers and special additive to increase freeze-thaw resistance. The effects that different sand and additions have on the properties of fresh SCC (slump-flow test, V-funnel test, and L-box test) and compressive strengths in early ages (3 and 7 days) were investigated and discussed.

AB  - The aim of this investigation is to develop ultra high performance concrete (UHPC) for multipurpose - higher mechanical and ionizing radiation protection. The effect of nano-silica replacement (2 or 5%) on the properties of UHPC was compared with the referent concrete by testing compressive and flexural strength. As nano-silica influences cement hydration and modifies the pore structure, qualitative and quantitative analysis of pores was done using a device RapidAir 457. The second objective of this paper is to evaluate the influence of different aggregates on concrete properties. Two types of aggregate were used: quartz and barite. One of the most important characteristics of the concrete for protection against gamma and X radiation is its Total Attenuation Coefficient (µ/ρ)tot.
