AB  - Structural knowledge of protein assemblies in their physiological environment is paramount to understand cellular functions at the molecular level. Protein interactions from Imaging Complexes after Translocation (PICT) is a live-cell imaging technique for the structural characterization of macromolecular assemblies in living cells. PICT relies on the measurement of the separation between labelled molecules using fluorescence microscopy and cell engineering. Unfortunately, the required computational tools to extract molecular distances involve a variety of sophisticated software programs that challenge reproducibility and limit their implementation to highly specialized researchers. Here we introduce PyF2F, a Python-based software that provides a workflow for measuring molecular distances from PICT data, with minimal user programming expertise. We used a published dataset to validate PyF2F’s performance.

Gold-riboflavin hybrid nanostrucutures as possible photodynamic therapy agents

AB  - This study reports on the optical properties of hybrid nanostructures that comprise gold nanoparticles (Au NPs) and biomolecule riboflavin (vitamin B2, Rb). The riboflavin is photosensitizing molecule and the generation of reactive oxygen species (ROS) by the hybrid nanoparticles was tested in various aqueous and biological environments. It was shown that the gold nanoparticles enhanced the photosensitizing activity of riboflavin. A comparison of EPR spectra obtained for the Au nanoparticle colloid, hybrid nanoparticle colloid and molecular water solution revealed that the strongest ROS signal came from the AuRb nanosystem. Furthermore, the ROS signal stability was much higher than that of the molecular solution, with a time span of several minutes. Photodynamic activity of the AuRb hybrids was tested by using gram-negative bacterium Escherichia coli as a model system. The bacteria were incubated by AuRb nanoparticles and the survival rate of the colonies was investigated by fluorescent bioimaging. The obtain results provide new insights on photosensitizer’s activities and suggest advantages of using metal-based nanoplatforms for developing of novel agents for photodynamic therapies.

AB  - Interest in developing micro/nano motors (MNMs) increased rapidly in the last couple of years. These advanced systems can address critical challenges in manipulation of objects at small-scales. Micro/nano motors are capable to move controllably in fluids under the influence of various stimuli (thermal, magnetic, light, etc.) and perform specific operations, including propelling micromachines. Here we synthesized two types of hybrid Janus particles, Au-TiO2 and AgAg2S-TiO2, and tested them as possible light-driven micromotors. Light, as a versatile power source, allows non-invasive control of the motion of the motors with high spatial and temporal resolutions. Illumination induce asymmetric reactions on the two sides of the Janus particles, which can create the propulsion. To optimize the properties of the MNMs, we performed synchrotron radiation gas-phase photoemission spectroscopy (SR PES) of the free standing hybrid particles at synchrotron SOLEIL, France. SR PES provided an information about the valence level alignment of the components, possible hybrid states and dominant scattering processes.

AB  - This article reports on photodielectric properties of hydrocolloids of TiO2  particles and Au/TiO2 hybrid particles of lateral dimension of ∼200 nm. Illumination of the colloids with visible light did not cause measurable changes in their electrical conductivity, while the application of UV (365 nm) light led to photoinduced increase in conductivity of up to 2%. The photogeneration of ions in water, regardless of the presence of the particles, makes a dominant contribution to the photoinduced increase in conductivity of the colloids

AB  - Gold nanoparticles were functionalized by amino acid tryptophan and vitamin riboflavin - a resonance energy transfer (RET) pair of biomolecules. The presence of the gold nanoparticles resulted in 65% increase in RET efficiency. Because of enhanced RET efficiency, the photobleaching dynamics of the fluorescent molecules at the surface of the nanoparticles is different from that of molecules in solution. The observed effect was used for detection of the functionalized nanoparticles within biological material rich with autofluorescent species. Synchrotron radiation deep-ultraviolet fluorescence microscopy is used to study the photobleaching dynamics of the fluorescence centers within human hepatocellular carcinoma Huh7.5.1 cells incubated with the nanoparticles. The fluorescent centers were classified according to their photobleaching dynamics, which enabled the discrimination of the cell areas where the accumulation of the nanoparticles takes place, even though the particles were smaller than the spatial resolution of the images.

Toward improved PVDF-BaTiO3 composite dielectrics: mechanical activation of the filler versus filler content

AB  - Barium titanate (BT) perovskite particles were surface modified by means of mechanical treatment and used as inorganic component in polyvinylidene fluoride (PVDF) based composites. The changes in electrical properties of the composite films with increasing in filler content were followed by dielectric spectroscopy, breakdown strength and D-E measurements. A comparison of the properties of the composites prepared with untreated and mechanically activated particles revealed that there is a significant difference in their performances at low filler concentrations (<20 wt%). Introduction of the surface modified ceramic particles into PVDF matrix led to an increase of the dielectric constant without affecting significantly the electrical breakdown strength. In contrast, when as received BT particles were used a filler, both dielectric constants and breakdown strengths of the composite films were lower than the corresponding values observed for the pure PVDF. At higher concentrations, however, the influence of pre-treatment of the filler on the effective electrical properties becomes less significant. The obtained results were discussed in terms of the pronounced crystallization of polar β and γ crystal phases of PVDF in the presence of surface modified BT fillers, which is confirmed by Raman spectroscopy.

AB  - Ceramic/polymer composites can be chemically stable, mechanically strong, and flexible, which make them candidates for electric devices, such as pressure or temperature sensors, energy storage or harvesting devices, actuators, and so forth. Depending on the application, various electrical properties are of importance. Polymers usually have low dielectric permittivity, but increased dielectric permittivity can be achieved by the addition of the ceramic fillers with high dielectric constant. With the aim to enhance dielectric properties of the composite without loss of flexibility, 5 wt% of BaTiO3-Fe2O3 powder was added into a polyvinylidene fluoride matrix. The powder was prepared by different synthesis conditions to produce core/shell structures. The effect of the phase composition and morphology of the BaTiO3-Fe2O3 core/shell filler on the structure and lattice dynamics of the polymer composites was investigated. Based on the results of the thermal analysis, various parameters of ceramic/polymer composites were determined. Differences in the phase composition and morphology of the filler have an influence on the formation of various polyvinylidene fluoride allomorphs and the degree of crystallinity. Furthermore, the dielectric performances of pure polyvinylidene fluoride and the polymer/ceramic composites were measured.

AB  - Silver–bismuth iodide (Ag–Bi–I) rudorffites are chemically stable and non-toxic materials that can act as a possible lead-free replacement for methylammonium lead halides in optoelectronic applications. We report on a simple route for fabricating Ag–Bi–I colloidal nanoplatelets approximately 160 nm in lateral dimensions and 1–8 nm in thickness via exfoliation of Ag–Bi–I rudorffite powders in acetonitrile. The valence band electronic structure of isolated Ag–Bi–I nanoplatelets was investigated using synchrotron radiation to perform X-ray aerosol photoelectron spectroscopy (XAPS). The ionization energy of the material was found to be 6.1 ± 0.2 eV with respect to the vacuum level. UV–vis absorption and photoluminescence spectroscopies of the Ag–Bi–I colloids showed that the optical properties of the nanoplatelets originate from I 5p to Bi 6p and I 5p to I 5p transitions, which is further confirmed by density functional theory (DFT) calculations. Finally, calculations based on the DFT and k · p theoretical methods showed that the quantum confinement effect is very weak in the system studied.

In this approach the aerosol particles can be produced directly from a solution or a colloidal dispersion, which opens a possibility for investigation of a variety of nanosystems that can be produced by wet chemistry methods. This technique relies on the interaction of focused beam of isolated particles with ionizing radiation under high vacuum conditions. In addition, by using tunable synchrotron radiation as an excitation source it is possible to obtain highresolution photoelectron spectra in the investigated photoelectron energy range.

AB  - We report on hydrothermal synthesis of zinc oxide (ZnO) hexagonal microrods and microtubes. Zinc oxide seeds were deposited onto glass slides by dip coating and used as templates for hydrothermal growth of ZnO microcrystals. The process was carried out in an alkaline reaction medium with acetate and nitrate zinc precursors, using hexamethylene tetramine (HMT) as surfactant. The obtained ZnO microrods were successfully decorated with silver particles via dipping ZnO-containing glass slides in silver nitrate solution and subsequent UV irradiation (5 min). Zinc oxide and Ag/ZnO microparticles were characterized by SEM, EDS and XRD. The presented results are part of the larger concept of developing of anisotropic Ag/ZnO microrods for conversion of chemical to mechanical energy.

AB  - Fluorescent nanocomposite films were prepared by solution mixing of polyvinyl alcohol (PVA) and tryptophan functionalized silver (AgTrp) nanoparticles. Synchrotron excitation deep-ultraviolet (DUV) fluorescence imaging was used to follow the distribution of AgTrp nanoparticles within the polymer matrix. Pronounced growth of dendrite-like structures in the presence of AgTrp nanoparticles was confirmed by bright field and fluorescence imaging as well as with atomic force microscopies. Both tryptophan and AgTrp nanoparticles influence thermal properties of the matrix. Differential scanning calorimetry measurements showed that they reduce crystallinity and significantly increase the glass transition temperature of PVA (in the case of PVA-AgTrp film the Tg increases by ~20 °C). Dielectric properties of the films were studied under two different conditions (dark and illumination). The illumination with 250 nm light induces a significant increase in conductance of the PVA-AgTrp nanocomposite. The relative changes, with respect to the conductance measured in the dark, were almost ~200%.

AB  - Na0.25K0.25Bi0.5TiO3 (NKBT) perovskite particles are synthesized by solid-state method and used as a filler for polyvinylidene fluoride-co-hexafluoropropylene (PVDF-HFP) co-polymer. X-ray diffraction analysis of NKBT powders shows that the particles have a rhombohedral perovskite crystal structure (R3c symmetry). Raman spectroscopy reveals that the co-polymer crystallizes predominantly into the mixture of polar β- and γ-crystals, while there is also a contribution of the non-polar α-crystal phase. The introduction of the NKBT into the PVDF-HFP results with an increase in effective dielectric permittivity and this effect depends on the inorganic content in the composite. The most interesting result of the present study is that the introduction of NKBT particles induces the appearance of an additional transition peak in the dielectric spectra of the co-polymer matrix. At the fixed frequency of ~2 kHz, the observed process appears at ~10 °C (about 45° above the glass transition temperature) and its magnitude strongly depends on the amount of the NKBT in the composite. Dielectric spectroscopy measurements of the composites are carried out in the wide range of frequencies (from 0.1 Hz to 1 MHz) and temperatures (from −100 to 100 °C). They reveal that the novel process can be clearly distinguished in the frequency range between 160 Hz and ~50 kHz. © 2021 The Authors

Fabricating AgBi-I in nanocrystal form couldfacilitate further their integration in the photovoltaic devices and enhance device performance due to size confinement effects. In our previous study, we successfully fabricated ligand-free Ag3BiI6nanoparticles in the form of aerosols [3]. Here, we report on thefabricationprocedure of 2D Ag-Bi-I nanomaterials in the colloidal form. The results of the structural and morphological investigation of the nanosheets will be presented, as well as reconstruction of the electronic levels of the Ag3BiI6nanoparticles from the combined UV-vis absorption andX-ray photoelectron spectroscopy data.In addition, an analysis that shows the relation between the positions of the bands in the Ag3BiI6nanosheet absorption spectra and the thickness of the nanosheet will be discussed.

AB  - Molecular resonance energy transfer (RET) is a basis of a half-century-old tool known as the molecular ruler, used for inference of mutual distances of two spectrally-coupled fluorophores usually relevant in biological contexts [1]. RET influences not only the absolute intensities of fluorescence signals of the molecular pair, but also their other optical characteristics, e.g. the valence electrons’ excited states. On the other hand, metal-based hybrid nanostructures show great promise as future multifunctional nanoplatforms, due to their superior sensitivity to external perturbations [2]. The metal components of the nanostructures influence the optical properties of fluorescent molecules in the vicinity, similarly as bulk metals do, which often presents a drawback. Here we want to demonstrate how this particular feature in metal-based nanosystems can be used as an advantage point in conventional biophysical imaging if the beneficial underlying physical mechanisms are identified [3]. In this talk, we will first explain the influence of gold nanoparticles on resonance energy transfer between tryptophan, essential amino acid, and riboflavin, vitamin B2, in an aqueous environment. We will show that the relative change of the signal in the tryptophan spectral channel is more pronounced when the biomolecules are attached to the nanoparticles. Afterward, we will demonstrate how timelapse fluorescence imaging of nanoparticle-incubated liver cancer cells can be used for inferring subcellular areas of nanoparticle accumulation. By exposing the incubated cell to continuous UV radiation, the photobleaching of the fluorescent molecules in the sample occurs. Being sensitive to the electronic structure of the fluorophore, the photobleaching rates of different subareas in the cell show distinctive trends, depending on the chemical composition. As the nanoparticles promote RET between tryptophan and riboflavin, the nanostructures exhibit a particular photobleaching trend compared to the cells' endogenous fluorescent species [4]. Thus, by separating areas of fluorescence images into different classes based on their photobleaching rates, i.e. grouping the pixels of the same signal trends, we are able to infer the preferential accumulation sites of bifunctionalized gold nanoparticles by using fluorescence microscopy of lower lateral resolution than the size of the nanoparticles. In this way, we demonstrate a potent auxiliary detection modality of metal-based hybrid nanostructures in optical imaging.

As the nanoparticles promote RET between tryptophan and riboflavin, the nanostructures exhibit a particular photobleaching trend compared to the cells' endogenous fluorescent species [4]. Thus, by separating areas of fluorescence images into different classes based on their photobleaching rates, i.e. grouping the pixels of the same signal trends, we are able to infer the preferential accumulation sites of bifunctionalized gold nanoparticles by using fluorescence microscopy of lower lateral resolution than the size of the nanoparticles. In this way, we demonstrate a potent auxiliary detection modality of metal-based hybrid nanostructures in optical imaging.",

AB  - The increase in the utilization of ceramic/polymer composites as components for electric devices is mainly based on their high chemical stability, mechanical strength, and flexibility. The polymers usually have poor dielectric performances. In order to increase the dielectric permittivity, ceramic fillers with high dielectric constant have been used as active fillers, and usually in large amounts, which have a detrimental effect on the mechanical properties of the composite. With the aim to enhance the dielectric properties of the composite without loss of polymer’s flexibility, 5 wt.% of BaTiO3/Fe2O3 core/shell powders, prepared by different synthesis conditions, was added into the PVDF matrix. The effect of the phase composition and morphology of the starting BaTiO3/Fe2O3 core/shell filler on the crystal structure and lattice dynamics was investigated. Based on the results of the thermal analysis, TG/DSC, various parameters of ceramic/polymer composites were determined. We were able to corroborate that differences in the phase composition and morphology of BaTiO3/Fe2O3 core/shell filler have an influence on the formation of various PVDF allomorph modifications, as well as the level of crystallinity. The dielectric performances of the pure PVDF and polymer/ceramic composites were measured. An enhancement in the dielectric permittivity and decrease in the loss tangent were revealed.

AB  - We report on the aerosol generation of ligand-free silver iodobismuthate (Ag-Bi-I) nanoparticles (NPs) and on in situ investigation of their electronic structure using synchrotron radiation soft X-ray aerosol photoelectron spectroscopy (XAPS). The structural and morphological characterizations revealed the aerosol to be composed of spherical rudorffite Ag3BiI6 particles, approximately 100 nm in size. The XAPS showed well-resolved signals from all expected elements (Ag, Bi, and I) and allowed estimation of the NP work function to be about 4.5 eV. The ionization energy of Ag3BiI6 NPs was determined to be 6.1 eV that is in good agreement with our calculations based on a hybrid functional approach. The presented method of production of Ag3BiI6 aerosol can prove beneficial for the future development of Ag-Bi-I-based photovoltaic materials, since it allows the deposition of Ag-Bi-I particles on large surface areas of arbitrary shape and roughness.

AB  - We will describe a novel experimental technique that combines surface XPS and gas-phase synchrotron radiation-based XPS/VUV PES, for the investigation of the electronic structure of different nanocrystals that could be low-cost and high-efficient solar cell absorbers [1]. Surface XPS is performed in Notre Dame University (US) using SPECS Surface Nano Analysis GmbH, with a PHOIBOS 150 hemispherical energy analyzer. The synchrotron based XPS/PES is performed at the SOLEIL synchrotron (France). For gas-phase PES, the nanoparticle beam is produced by an atomizer and focused by the aerodynamic lens into the interaction region. On DESIRS beamline, the angle-resolved photoemission spectra were measured by double imaging DELICIOUS3 spectrometer (VMI for electrons and VMI-TOF for ions). On PLEIADES, the ±30o aperture wide-angle lens VG-Scienta R4000 electron analyzer was used to record the XPS. The nanoparticles are synthetized both at Vinca Institute of Nuclear Sciences (Serbia) and in Notre Dame. The experimental results are supported by DFT calculations performed at the Institute of Physics Belgrade (Serbia). Recent results for lead halide perovskites [1] will be presented, as well as preliminary results for lead-free materials such as Ag-Bi-I rudorffite nanosystems. The procedure to synthetize nanoparticles of this material for aerosol generation has been most recently developed at Vinca Institute of Nuclear Sciences. We were able to obtain a complete band alignment to the vacuum of the nanocrystals and investigate their surface properties with high sensitivity.

AB  - Functional materials based on graphene oxide (GO) and reduced graphene oxide (rGO) have a high potential for application in the fields of biophysics, material science, and biomedical engineering [1]. It is due to their tunable physical properties, high surface area, remarkable photoluminescence, as well as their controllable chemical functionalization [2]. Beyond their applications in nanomedicine for drug/gene delivery, phototherapy and bioimaging, they have shown significant interaction and adhesive properties with proteins, mammalian cells and microorganisms, which makes them potential candidates for multifunctional biological applications. In this lecture, we will present a study of the interaction of partially reduced graphene oxide (prGO) with Huh7.5.1 liver cancer cells. The study was conducted by means of synchrotron excitation DUV fluorescence bioimaging (performed on DISCO beamline of synchrotron SOLEIL) [3]. The prGO sample was obtained by the reduction (to a certain extent) of the initially prepared GO nanosheets. The fluorescence of the GO nanosheets increases with time of the reduction due to a change in the ratio of the sp2 and sp3 carbon sites, and the prGO sample was extracted from the dispersion when the intensity of the fluorescence reached its maximum. After that, Huh7.5.1 cells were incubated with GO, prGO and rGO nanosheets and used in bioimaging studies. The presence of graphene materials influenced the fluorescence properties of the cells, and by analyzing fluorescence photobleaching dynamics, we were able to localize graphene nanosheets inside the liver cancer cells.

AB  - Aerosol photoemission spectroscopy became an important method for studying electronic structure of submicrometer particles without the influence of substrate [1]. It comprises irradiation of the focused particle beam by either vacuum-ultraviolet (VUV) or soft x-ray radiation under high vacuum conditions and subsequent detection and discrimination of the photoelectrons according to their kinetic energies and momenta [2-4]. As intermediates between atoms or molecules and macroscopic matter, nanometer-sized objects exhibit specific electronic and transport properties that strongly depend on their size, morphology and surface chemistry. Modification of nanoparticle surfaces by conjugation with molecules presents a convenient method of altering a wide range of physicochemical characteristics of the nanomaterials, which does not require development of new synthetic procedures. Furthermore, by a proper choice of molecules used in surface modification, additional properties of the hybrid nanostructures can be achieved, which could not be found in the starting materials. In this lecture, we will present the selected results of our investigations on hybrid nanostructures comprised of noble metal and metal oxide nanomaterials functionalized by biologically relevant molecules. Particular attention will be given to the vacuum-ultraviolet angle-resolved photoelectron spectroscopy (VUV ARPES) studies on isolated functionalized nanosystems performed at the DESIRS beamline. The emergence of the photoelectron circular dichroism in hybrid nanoparticles will be briefly discussed.

AB  - The interaction of partially reduced graphene oxide (prGO) and Huh7.5.1 liver cancer cells was investigated by means of DUV fluorescence bioimaging. The prGO sample was obtained by the reduction (to a certain extent) of the initially prepared graphene oxide (GO) nanosheets with hydrazine. The fluorescence of the GO nanosheets increases with time of the reduction due to a change in ratio of the sp2 and sp3 carbon sites and the prGO sample was extracted from the dispersion after 6 min, when the intensity of the fluorescence reached its maximum. The reduction process was left to proceed further to saturation until highly reduced graphene oxide (denoted here as rGO) was obtained. GO, prGO and rGO samples were investigated by structural (scanning electron microscopy (SEM), scanning transmission electron microscopy coupled with energy dispersive spectrometry (STEM-EDS)) and spectroscopic (UV-vis, photoluminescence (PL), Raman) methods. After that, Huh7.5.1 cells were incubated with GO, prGO and rGO nanosheets and used in bioimaging studies, which were performed on DISCO beamline of synchrotron SOLEIL. It was found that the prGO significantly enhanced the fluorescence of the cells and increased the intensity of the signal by ~2.5 times. Time-lapse fluorescence microscopy experiments showed that fluorescence dynamics strongly depends on the type of nanosheets used. The obtained prGO nanostructure can be easily conjugated with aromatic ring containing drugs, which opens a possibility for its applications in fluorescence microscopy monitored drug delivery. © 2018 IOP Publishing Ltd.

AB  - We report an investigation of lead halide perovskite CH3NH3PbBr3 nanocrystals and associated ligand molecules by combining several different state-of-the-art experimental techniques, including synchrotron radiation-based XPS and VUV PES of free-standing nanocrystals isolated in vacuum. By using this novel approach for perovskite materials, we could directly obtain complete band alignment to vacuum of both CH3NH3PbBr3 nanocrystals and the ligands widely used in their preparation. We discuss the possible influence of the ligand molecules to apparent perovskite properties, and we compare the electronic properties of nanocrystals to those of bulk material. The experimental results were supported by DFT calculations.

Structural and electrical properties of ferroelectric poly(vinylidene fluoride) and mechanically activated ZnO nanoparticle composite films

AB  - The influence of the mechanical activation of ZnO nanoparticle fillers on the structural and electrical properties of the matrix of poly(vinylidenefluoride)-ZnO (PVDF-ZnO) films was investigated. Transmission electron microscopy and scanning electron microscopy analyses showed that mechanical activation in a high energy planetary ball mill reduces the size of ZnO particles. X-ray diffraction and Raman spectroscopy revealed that PVDF crystallized predominantly as the γ-phase. Non-activated ZnO filler reduces the degree of the crystallinity of the matrix and promotes crystallization of α-phase of PVDF in the film, while the fillers activated for 5 and 10 min induce crystallization of β-phase, indicating that mechanical activation of the filler can be used as a general method for fabrication of PVDF composites with increased content of piezoelectric β-phase crystals. Dielectric spectroscopy measurements show that polymer composite with the high content of β-phase (with ZnO filler activated for 5 min) exhibits the highest value of dielectric permittivity in 150-400 K range of temperatures. Kinetic analysis shows combined effects of increased surface area and increased concentration of surface defects on the interactions between polymer chains and activated nanoparticles. © 2018 IOP Publishing Ltd.

Interaction of amino acid-functionalized silver nanoparticles and Candida albicans polymorphs: A deep-UV fluorescence imaging study

AB  - The interaction of the tryptophan functionalized Ag nanoparticles and live Candida albicans cells was studied by synchrotron excitation deep-ultraviolet (DUV) fluorescence imaging at the DISCO beamline of Synchrotron SOLEIL. DUV imaging showed that incubation of the fungus with functionalized nanoparticles results in significant increase in the fluorescence signal. The analysis of the images revealed that the interaction of the nanoparticles with (pseudo)hyphae polymorphs of the diploid fungus was less pronounced than in the case of yeast cells or budding spores. The changes in the intensity of the fluorescence signals of the cells after incubation were followed in [327-353 nm] and [370-410 nm] spectral ranges that correspond to the fluorescence of tryptophan in non-polar and polar environment, respectively. As a consequence of the environmental sensitivity of the silver-tryptophan fluorescent nanoprobe, we were able to determine the possible accumulation sites of the nanoparticles. The analysis of the intensity decay kinetics showed that the photobleaching effects were more pronounced in the case of the functionalized nanoparticle treated cells. The results of time-integrated emission in the mentioned spectral ranges suggested that the nanoparticles penetrate the cells, but that the majority of the nanoparticles attach to the cells surfaces.
