AB  - A binary system composed of carbon dots (CDs) and N-doped CDs (N-CDs) embedded in an organic matrix was used for the analysis of cholesterol by MALDI (matrix-assisted laser desorption and ionization time-of-flight) mass spectrometry, as a model for detection of small, biologically relevant molecules. The results showed that both CDs are sensitive to the cholesterol and can be used either alone or in a binary system with 2,5-dihydroxybenzoic acid (DHB) to enhance the detection process. It was found that both COOH and NH2 groups on CDs surface contributed to the enhancement in the cholesterol detection by MALDI mass spectrometry in the presence of inorganic cations. Nevertheless, in the presence of NaCl, N-CDs led to a better reproducibility of results. It was due to the coexistence of positive and negative charge on N-CDs surface that led to a homogeneous analyte/substrate distribution, which is an important detection parameter. The enhancing effect of carbon dots was linked to a negative Gibbs energy of the complex formation between CDs, Na+, cholesterol and DHB, and it was supported by theoretical calculations. Moreover, upon the addition of CDs/N-CDs, such features as a low ionization potential, vertical excitation, dipole moment and oscillator strength positively affected the cholesterol detection by MALDI in the presence of Na+. © 2021 Elsevier Inc.

AB  - Rationale: Changes in lipid composition might be associated with the onset and progression of various neurodegenerative diseases. Herein, we investigated the changes in the plasma phosphatidylcholine (PC)/lysophosphatidylcholine (LPC) ratios in patients with Parkinson's disease (PD) in comparison with healthy subjects and their correlation with clinico-pathological features. Methods: The study included 10 controls and 25 patients with PD. All patients were assigned to groups based on clinico-pathological characteristics (gender, age at examination, duration of disease and Hoehn and Yahr (H&Y) stage). The analysis of the PC/LPC intensity ratios in plasma lipid extracts was performed using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. Results: PD patients exhibited an increased PC/LPC intensity ratio in comparison with the control group of healthy subjects. Furthermore, the investigated ratio was shown to be correlated with clinico-pathological parameters, in particular with H&Y stage and disease duration. The PC/LPC intensity ratio in plasma samples of PD patients was found to be elevated in all examined H&Y stages and throughout the disease duration. Conclusions: To our knowledge, this is the first study examining the PC/LPC ratios in plasma of patients with PD and illustrating their correlation with clinico-pathological features. Although the presented results may be considered as preliminary due to the limited number of participants, the observed alterations of PC/LPC ratios in plasma might be a first step in the characterization of plasma lipid changes in PD patients and an indicator of lipid reconfiguration. © 2019 John Wiley & Sons, Ltd.

AB  - In this work, the performances of two ionic liquid matrices (ILMs) with the same ammonium counterpart for mass spectrometric analysis of the insoluble and soluble sucralfate were compared. Matrix-assisted laser desorption/ionization mass spectrometry (MALDI MS) was performed assisted by the butylammonium salts of α-cyano-hydroxycinnamic acid (CHCAB) and 2,5-dihydroxybenzoic acid (DHBB). CHCAB has a higher IE than DHBB, but better optical properties. CHCAB is more suitable for the analysis of sucralfate, although molecular ions of both compounds were detectable only with low intensities. Thus, optical properties of ILMs are crucial to enhance the sensitivity of MALDI MS detection of polysulfated oligosaccharides. © 2019, © 2019 Taylor & Francis Group, LLC.

AB  - TiO2 prolatenanospheroids (PNSs) may be photosensitizers (PSs), which act by catalyzation of hydroxyl radical (∙OH) formation upon light illumination. ∙OH might, in turn, contribute to killing of cancer cells. On the other hand, there is great concern about toxicity in the dark of TiO2 nanoparticles in general. In this work, we have investigated the biocompatibility of TiO2 PNSs of the anatase crystal form (length between 100 and 300 nm and width 50 nm) in the dark with immune cells and light-induced cytotoxicity on several cancer cell lines. The effects of the treatment of different cell lines with several concentrations of TiO2 PNSs suspensions showed the specifics of cells’ viability and the intracellular localization. The results of in vitro studies obtained by cytotoxicity assays adjusted to individual cell lines’ metabolism point towards the biocompatibility of TiO2 PNSs at low and moderate concentrations in the dark, which neither kill the cells, nor induce activation of the immune system cells. Laser scanning confocal microscopy revealed that PNSs are taken up by cells, and insight into the intracellular distribution was obtained in this study.

AB  - Abstract: In this study, we have synthesized a series of dinuclear and trinuclear gold(III) complexes of the general formula [Au2(N–N)Cl6] (1–3) for dinuclear and [Au3(N–N)2Cl8]+ (4–6) for trinuclear compounds, respectively, in which N–N is a bidentate ligand (1,4-diaminobutane; 1,6-diaminohexane or 1,8-diaminooctane). These complexes were characterized by elemental analysis, molar conductivity, and spectroscopic techniques (IR, UV–Vis, 1H NMR, ESI–MS). We performed DFT calculations to get insight into the geometry of the studies complexes. DNA-binding studies were performed by UV–Vis spectrophotometry and fluorescence spectroscopy. The results of competitive reactions between gold(III) complexes and ethidium bromide (EB) towards DNA have shown that selected complexes can displace EB from DNA-EB adduct. In addition, these experiments confirm that polynuclear gold(III) complexes interact with DNA covalently or via intercalation. Furthermore, high values of binding constants of gold(III) complexes towards bovine serum albumin (BSA) protein indicate good binding affinity. In addition, redox stability of complexes in the presence of DNA/BSA was confirmed by cyclic voltammetry. Results of the interactions between gold(III) complexes with DNA/BSA were discussed in reference to molecular docking data obtain by Molegro virtual docker. The cytotoxic activity of synthesized gold(III) complexes was evaluated on human breast cancer cell line (MDA-MB-231), human colorectal cancer cell line (HCT-116), and normal human lung fibroblast cell line (MRC-5). All complexes dose-dependently reduced cancer and normal cells viabilities, with significant cytotoxic effects (IC50 < 25 μM) for trinuclear gold(III) complexes (4, 5) on HCT-116 cells. AB  - Binding of three ruthenium(ii) compounds of general formula mer-[Ru(L3)(N-N)X][Y] (where L3 = 4-chloro-2,2:6,2-terpyridine (Cl-tpy); N-N = 1,2-diaminoethane (en), 1,2-diaminocyclohexane (dach) or 2,2-bipyridine (bipy); X = Cl; Y = Cl) to human serum albumin (HSA) has been investigated by nano-LC/nano-ESI MS and docking studies. A bottom-up proteomics approach has been applied for the structural characterization of metallated proteins and the data were analyzed in both the positive and negative ion mode. The negative ion mode was achieved after the post-column addition of an isopropanol solution of formaldehyde that enabled sample ionization at micro-flow rates. The negative ion mode MS has been proved to be beneficial for the analysis of binding sites on ruthenated protein in terms of ion charge reduction and consequent simplification of target sequence identification based on isotopic differences between ruthenated and non-ruthenated peptides. Moreover, the negative ion mode ESI MS shows the advantage of singly charged ion formation and, unlike MALDI MS, it does not cause complete ligand fragmentation, merging the benefits of each method into a single experiment. Six target sequences were identified for the binding of en and dach compounds, and four sequences for the binding of bipy. All compounds have been found to bind histidine and one aspartate residue. Docking studies showed that the identified sequences are the constituents of five distinct binding sites for en and dach, or two sites for the bipy complex. The selection of binding sites seems to be dependent on the chelate ligand and the form of the complex prior or after hydrolysis of the leaving chloride ligand.

Gold chloride cluster ions generated by vacuum laser ablation

AB  - In this work, a simple way for study the possibility of formation a vapor cluster species of tetrachloroauric acid (HAuCl4), using the laser ablation in the absence of a buffer or reactive atmosphere, and without a postablation supersonic expansion on a commercial matrix assisted laser desorption/ionization time-of-flight mass spectrometer, is reported. Tetrachloroauric acid is known as precursor for the synthesis of gold nanostructures and the complex salts; therefore it is an important task to discover and quantify the species arising from HAuCl4, in order to understand their role in the gold assisted reactions. Mass spectrum of HAuCl4in a reflector negative-ion mode contains the hydrated mono- and dinuclear gold clusters in the m/z range 286–436, and gold chloride clusters in the m/z range 447–795. In the first part of spectrum, m/z range 286–436, the hydrated gold cluster species of type Aun−(H2O)m(n = 1–2; m = 1, 2, 5, 7, 8) and [Aun(OH)k]−(H2O)m(n = 1–2; k = 1–2; m = 1, 4–8) were found. Besides that, there are gold chloride clusters with general formula [AuHr(HCl)2]−(H2O)m(m = 1–5; 8–9; r = 0–2) in this part of spectrum. In the second part of spectrum, the m/z range 447–795, only gold chloride clusters were obtained. Their general formulae can be written as [AuClt(HCl)v]−(H2O)m(t = 1–4; v = 5–8; m = 2–4, 6–8) and [Aun(HCl)v]−(H2O)m(n = 1–2, v = 4–5, m = 1–2, 5, 7). The analysis of concentration effects on the LDI mass spectra of gold clusters reveals that the relative intensities of signals for the mono- and dinuclear Au clusters increase with decreasing the concentration of water HAuCl4solutions.

As potential medicament, we use Ru(II)(dcbpy)2Cl2complex, and in order to achieve better photosensitization properties, the Ru complex was attached to the nano carrier—TiO2nanoparticles. Additionally, this nanocomposite system was encapsulated in the phospholipid vesicles, which could be classified as small unilamellar vesicles, based on the technique of production. The complex-release tests were performed under light illumination, at pH 5, characteristic for tumor cells` interior and compared with the release pattern at pH 7, characteristic for the serum, i.e. physiological solution.

AB  - Background: Biomarkers of oxidative stress are relevant in the evaluation of the disease status and prooxidant-antioxidant balance, advanced oxidation protein products and lipid peroxidation products (malondialdehyde and 4-hydroxynonenal) are being extensively evaluated regarding their relationship with clinical presentation and disease severity.Aim of the study: The aim of this study was to evaluate the levels of the above-mentioned parameters in plasma of 39 men and 17 women with Parkinson's disease, originated from the Republic of Serbia and their relation to clinicopathological characteristics (gender, age at examination, duration of the disease, and Hoehn and Yahr score) and oxidative status.Results: The incidence of disease was 2:1 towards males. The investigated oxidative parameters were gender and Hoehn and Yahr related. Significant association of higher Hoehn and Yahr scores was observed for malondialdehyde (p = 0.01) and prooxidant-antioxidant balance (p = 0.02). Relation between oxidant-antioxidant status was further supported by observed positive correlation between 4-hydroxynonenal (p = 0.04) and prooxidant-antioxidant balance (p = 0.03). Finally, the multivariate analysis indicated that prooxidant-antioxidant balance and malondialdehyde were partially determined by gender (10.6% and 7.6%) and Hoehn and Yahr scores (13.6% and 18.8%), while Hoehn and Yahr scores contributed to the variance of advanced oxidation protein products with 13.2%.Conclusion: Our results indicate the higher level of oxidative stress (oxidant-antioxidant imbalance) and possible relation of several markers with gender and disease stage in patients with Parkinson's disease. The analyzed markers could be used to specify the severity of oxidative stress; however, their potential value should be analyzed in further studies.

AB  - Among various nano-scaled materials composed from a spectrum of chemical compounds, inorganic nanoparticles are very attractive due to their physico-chemical properties, as well as their availability, simplicity, possibility of modifications, stability and biocompatibility. They are, on the one hand, an useful tool in advanced analytical chemistry, in particular for studying of biologically-relevant processes, but also important as functional parts of the systems designed for controlled and targeted delivery of medicaments for treatment of a variety of diseases and for imaging. So far, thousands of compounds and systems have been developed for the above-mentioned purposes, but there are only a few reviews dealing with these topics. The aim of this review is, thus, to summarize recent applications of nano-structured inorganic materials in the field of drug delivery, bioimaging and bioanalytics, and to give a prospective from the standpoint of biology-related applications.

AB  - In this study the efficacy of two different methods for extracting lipopeptides produced by five Bacillus strains-ethyl acetate extraction, and acid precipitation followed by methanol extraction-was investigated using mass spectrometry. High performance thin layer chromatography (HPTLC) was also used for the simultaneous separation of complex mixtures of lipopeptide extracts and for the determination of antimicrobial activity of their components. The mass spectra clearly showed well-resolved groups of peaks corresponding to different lipopeptide families (kurstakins, iturins, surfactins, and fengycins). The ethyl acetate extracts produced the most favorable results. The extracts of SS-12.6, SS-13.1, and SS-38.4 showed the highest inhibition zones. An iturin analog is responsible for the inhibition of Xanthomonas arboricola and Pseudomonas syringae phytopathogenic strains. HPTLC bioautography effectively identified the active compounds from a mixture of lipopeptide extracts, proving in situ its potential for use in direct detection and determination of antimicrobials. In the test of potential synergism among individual extracts used in different mixtures, stronger antimicrobial effects were not observed. Biochemical and phylogenetic analysis clustered isolates SS-12.6, SS-13.1, SS-27.2, and SS-38.4 together with Bacillus amyloliquefaciens, while SS-10.7 was more closely related to Bacillus pumilus.

AB  - We studied the colloidal TiO2 nanoparticles as a carrier for controlled delivery of the ruthenium complex to the melanoma cell line. The system demonstrated slower complex release upon visible and increased release rate upon UV light illumination. Accordingly, the light-dependent cytotoxicity of the system was demonstrated on amelanotic melanoma cancer line. The cell death is enhanced by UV and reduced by red light in the presence of investigated nanocomposite system. Both components of the system may act as photosensitizers, by generating reactive oxygen species, which promote cell death. Thus, the system might act dually, as photodynamic therapeutic agent and as the light tunable system for metallo-drug delivery and it might be of interest for development of new more efficient drug delivery approaches by using a light as external stimulus. AB  - In this work it has been shown that both the laser desorption/ionization mass spectrometry (LDI MS) and the matrix assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI TOF MS) are the simple and quick methods for determination of relative natural isotopic distribution of lead. The analysis of metal salts with these approaches does not require any time-consuming preparation of samples: a single run can take only a minute, and numerous information can be obtained. Results obtained in this work show that chosen matrix has no negative effect on quantitative determination of lead isotopes and support once more the applicability of MALDI TOF MS for lead isotope distribution determination in the sample and accurate data are obtained. Additionally, the generation of PbnOn and PbnOn-1 (n: 2-6) clusters have been successfully achieved in the positive mode, using the both LDI and MALDI methods. All stoichiometries were confirmed using isotopic pattern modelling.

AB  - The paper presents chemical and structural analysis of geopolymer materials which are obtained by alkali-activated calcined clay (metakaolin) originated from Serbia under strictly defined conditions. Characterization of the metakaolin and geopolymers molecular structure has been done using X-ray diffraction, scanning electron microscope, Fourier transform infrared spectroscopy and matrix-assisted laser desorption ionization time-of-flight mass spectrometry. The paper presents the possibility of obtaining geopolymer structure and differences in chemical and structural characterization of these materials taking into account the concentration of NaOH as a variable parameter. The results of MALDI analysis of metakaolin and synthesized geopolymer structures using various matrix system: 2,4,6 trihydroxyacetophenone (THAP), alpha-cyano-4-hydroxycinnamic, 2,6 dihydroxyacetophenone and laser desorption/ionization, have shown that THAP matrix is the most appropriate for analysing these aluminosilicate materials.

AB  - This paper presents optimization of matrix-assisted laser desorption/ionization (MALDI) time-of-flight (TOF) mass spectrometer (MS) instrumental parameters for the analysis of chloro(2,2 , 2 -terpyridine) palladium(II) chloride dihydrate complex applying design of experiments methodology (DoE). This complex is of interest for potential use in the cancer therapy. DoE methodology was proved to succeed in optimization of many complex analytical problems. However, it has been poorly used for MALDI-TOF-MS optimization up to now. The theoretical mathematical relationships which explain the influence of important experimental factors (laser energy, grid voltage and number of laser shots) on the selected responses (signal to noise - S/N ratio and the resolution - R of the leading peak) is established. The optimal instrumental settings providing maximal S/N and R are identified and experimentally verified.

AB  - Photo-responsive drug release systems are promising for drug delivery applications due to many benefits compared to conventional chemotherapy such as targeted, controlled release of a drug and reduced toxicity to healthy tissues. In this work, we report synthesis of the nanocomposite system based on carrier TiO2 nanoparticles and potential anticancer ruthenium complex, with light controllable release properties. Nanocomposite system showed biological activity and induced the generation of free radicals, which are implied in the efficient cell killing. The drug release tests demonstrated sustained release of the transition metal complex from the surface implying the potency for the controlled drug delivery system. Taking into account photoactivity of the Ru-complex, in the next step we have investigated the influence of green light on the rate of the complex release, and the results showed dependence of the Ru-complex release from the surface of TiO2 nanoparticles on the applied laser energy. Therefore, these characteristics make this nanocomposite system promising for the photo-responsive chemotherapy.

AB  - Ruthenium compounds are highly regarded as metallo-drug candidates. Many studies have focused their attention on the interaction between ruthenium complexes with their possible biological targets. The interaction of ruthenium complexes with transport proteins, enzymes and peptides is of great importance for understanding their biodistribution and mechanism of action, therefore, the development of an anti-cancer therapy involving ruthenium complexes has recently shifted from DNA targeting towards protein targeting. With the aim of gaining insight into possible interactions between ruthenium complexes with biologically relevant proteins, we have studied the interaction of cis-dichlorobis(2,2-bipyridyl-4,4-dicarboxylic acid)ruthenium(II) complex [Ru(II)(dcbpy)(2)Cl-2], which previously showed good potency in photo-dynamic chemotherapy, with bovine serum albumin (BSA), phospholipase A(2) (PLA(2)) and glutathione (GSH). Binding constants and possible number of binding sites to mentioned proteins and peptide are investigated by ultraviolet-visible spectroscopy and Matrix-Assisted Laser Desorption Ionization Mass Spectrometry (MALDI TOF MS). The complex binding affinities were in the following order: PLA(2) GT BSA GT GSH. Moreover, genotoxic profile of the complex, tested on peripheral blood lymphocytes as a model system, was also promising.

AB  - In the present study, the possibility for the application of TiO2 nanocrystals of various shapes and sizes, for substrate-assisted laser desorption and ionization time-of-flight mass spectrometric (SALDI TOF MS) quantitative analysis of small molecules (steroid hormones, amino acids and saccharides) was investigated. Parameters, such as homogeneity of the substrate/analyte distribution, reproducibility of the measurements, within-day, and day-to-day repeatability, were determined. The homogeneity of different nanocrystal/analyte combinations on the target plate were compared based on the signal-to-noise values of several analyte signals. Obtained results show that all TiO2 nanocrystals, regardless of their shape, have great potential for the detection and determination of steroid hormones, amino acids and saccharides with good analytical parameters and detection limits. On the other hand, the reproducibility of the S/N ratio and detectability of the analytes recorded in various modes differ depending on the substrate. All examined molecules were detectable in negative ion mode with TiO2 NTs, in contrast to all other organic matrices and substrates, and the best reproducibility was obtained with the larger nanocrystals, TiO2 PNSs and TiO2 NTs, making them good candidates for the quantitative determination of small molecules.

AB  - Titanium dioxide is widely used substrate for laser desorption and ionization [(SA)LDI] mass spectrometry. As the semiconductor with high UV absorbance and rapid energy transfer to analytes, TiO2 provides soft and efficient desorption/ionization of analytes with relatively low degrees of fragmentation in the gas phase. TiO2 nanocrystals are convenient for small molecules (molecular mass LT 1000 Da) analysis due to low number of background signals. Desorption/ionization processes on TiO2 nanocrystals are related to the physical properties of the substrate, such as the ability to absorb and dissipate energy from the irradiating laser light source. In addition, size and shape of nanocrystals also affect these abilities. The aim of this study is to investigate the influence of shape and size dependant TiO2 nanocrystals/UV laser interaction on the quality of the mass spectra, detectability of analyzed molecules, intensities of the signals and homogeneity of co-crystals onto the MALDI plate, of carbohydrates. It is shown that the size and shape of nanocrystals influenced the way of packing carbohydrates onto plate, and thus affects homogeneity and reproducibility of mass spectrometry analysis.

AB  - Surface-assisted laser desorption/ionisation time-of-flight mass spectrometry (SALDI-TOF-MS) might be the method of choice for the analysis of low mass molecules (less than m/z 500). Titanium dioxide (TiO2) nanocrystals as a substrate for SALDI-TOF-MS improve the reproducibility of the signal intensities and prevent the fragmentation of some molecules upon laser irradiation, as we have previously shown. In addition, variously shaped and sized TiO2 nanocrystals/substrates for SALDI-MS could be used for quantification of small molecules, which are otherwise difficult to detect with the assistance of organic matrices. TiO2-assisted LDI-MS spectra could be acquired with excellent reproducibility and repeatability and with low detection limit. In the current study, we analysed the spectra of dexasone, citric acid, vitamin E and vitamin A acquired with TiO2 nanocrystals of various shapes and dimensions, i.e. the colloidal TiO2 nanoparticles (TiO2 NPs), TiO2 prolate nanospheroids (TiO2 PNSs) and TiO2 nanotubes (TiO2 NTs). Various shapes and dimensions of substrates were used since these factors determine desorption and ionisation processes. The homogeneity on the target plate was compared based on signal-to-noise values of peaks of interest of analysed molecules as well as the within-day and day-to-day repeatability. AB  - Hyphenated mass spectrometry (MS) techniques have attained an important position in analysis of covalent and non-covalent interactions of metal complexes with peptides and proteins. The aim of the present study was to qualitatively and quantitatively determine ruthenium binding sites on a protein using tandem mass spectrometry and allied techniques, i.e. liquid chromatography (LC) and inductively coupled plasma optical emission spectrometry (ICP-OES). For that purpose, two newly synthesized Ru(II) complexes of a meridional geometry, namely mer-[Ru(4 Cl-tpy)(en)Cl](+) (1) and mer-[Ru(4 Cl-tpy)(dach)Cl](+) (2) (where 4 Cl-tpy = 4-chloro-2,2:6,2 -terpyridine, en = 1,2-diaminoethane and dach = 1,2-diaminocyclohexane), and bovine serum albumin were used. The binding of the complexes to the protein was investigated by means of size exclusion- and reversed phase-LC, ICP OES, matrix-assisted laser desorption ionization MS and MS/MS. Ruthenated peptide sequence and a binding target amino acid were revealed through accurate elucidation of MS/MS spectra. The results obtained in this study suggest a high binding capacity of the protein towards both complexes, with up to 5.77 +/- 0.14 and 6.95 +/- 0.43 mol of 1 and 2 bound per mol of protein, respectively. The proposed binding mechanism for the selected complexes includes the release of Cl ligand, its replacement with water molecule and further coordination to electron donor histidine residue.
