Weather Conditions Influence on Lavandin Essential Oil and Hydrolate Quality

AB  - Lavandula sp. essential oil and hydrolate are commercially valuable in various industry branches with the potential for wide-ranging applications. This study aimed to evaluate the quality of these products obtained from L. x intermedia cv. ‘Budrovka’ for the first time cultivated on Fruška Gora Mt. (Serbia) during three successive seasons (2019, 2020, and 2021). Essential oil extraction was obtained by steam distillation, and the composition and influence of weather conditions were also assessed, using flowering tops. The obtained essential oils and hydrolates were analysed by gas chromatography with a flame ionization detector (GC-FID) and gas chromatography coupled to mass spectrometry (GC-MS). A linear regression model was developed to predict L. x intermedia cv. ‘Budrovka’ essential oil volatile compound content and hydrolate composition during three years, according to temperature and precipitation data, and the appropriate regression coefficients were calculated, while the correlation analysis was employed to analyse the correlations in hydrolate and essential oil compounds. To completely describe the structure of the research data that would present a better insight into the similarities and differences among the diverse L. x intermedia cv. ‘Budrovka’ samples, the PCA was used. The most dominant in L. intermedia cv. ‘Budrovka’ essential oil and hydrolate were oxygenated monoterpenes: linalool, 1,8-cineole, borneol, linalyl acetate, and terpinene-4-ol. It is established that the temperature was positively correlated with all essential oil and hydrolate compounds. The precipitations were positively correlated with the main compounds (linalool, 1,8-cineole, and borneol), while the other compounds’ content negatively correlated to precipitation. The results indicated that Fruška Gora Mt. has suitable agro-ecological requirements for cultivating Lavandula sp. and providing satisfactory essential oil and hydrolate.

AB  - In continuation of our study on Cotinus coggygria from Serbia, 10 known flavonoids (1-10) were isolated from the methylene chloride/ methanol extract of the heartwood. They were tested for in vitro protective effect against chromosome aberrations in peripheral human lymphocytes, using the cytokinesis-block micronucleus assay. All tested compounds (in minimal doses of 1 μg/mL) exerted a beneficial effect by decreasing DNA damage of human lymphocytes in the range of 24.2% to 54.5%, better than the radio protectant control, amifostine. Functional groups, such as 3′,4′-dihydroxyphenyl (catechol), 5-OH, 3-OH, and 4-keto in flavonoids (3-keto in aurones), which play a key role in antioxidant activity, are proposed to be responsible for the DNA protective activity of the tested compounds.

Antioxidant Activity of Selected Polyphenolics in Yeast Cells: The Case Study of Montenegrin Merlot Wine

AB  - Screens of antioxidant activity (AA) of various natural products have been a focus of the research community worldwide. This work aimed to differentiate selected samples of Merlot wines originated from Montenegro, with regard to phenolic profile and antioxidant capacity studied by survival rate, total sulfhydryl groups and activities of glutathione peroxidase (GPx), glutathione reductase and catalase in H2O2–stressed Saccharomyces cerevisiae cells. In this study, DPPH assay was also performed. Higher total phenolic content leads to an enhanced AA under both conditions. The same trend was observed for catechin and gallic acid, the most abundant phenolics in the examined wine samples. Finally, the findings of an Artificial Neural Network (ANN) model were in a good agreement (r2 = 0.978) with the experimental data. All tested samples exhibited a protective effect in H2O2–stressed yeast cells. Pre-treatment with examined wines increased survival in H2O2–stressed cells and shifted antioxidative defense towards GPx–mediated defense. Finally, sensitivity analysis of obtained ANN model highlights the complexity of the impact that variations in the concentrations of specific phenolic components have on the antioxidant defense system.

An insight into chemical composition and biological activity of Montenegrin Vranac red wine

AB  - This study aimed to describe quality and potential health benefits of three Vranac wines obtained from new grape clones (CI, CII and CIII) recently developed and recognised. In this aim, their phenolic profiles, anti-2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical activities along with metal compositions were analyzed. Compared with the commercial one, CII and CIII Vranac wines were found to contain both higher contents of total phenolics, flavonoids and monomeric anthocyanins as well as anti-DPPH radical activities. The most abundant phenolics were gallic acid (8.88-16.36 mg/L), catechin (12.42-24.35 mg/L) and epicatechin (4.30-7.79 mg/L). Finally, the metal content of all the analyzed samples was within the toxicological safety limits. Taken all together, Montenegrin Vranac wines represent a rich source of both phenolics and minerals exhibiting promising antioxidant potential. CII and CIII wines can be considered for commercialising due to their high amounts of gallic acid, catechin and epicatechin, significant K/Na ratios and favourable contents of essential metals making them products with health added values.

Additionally, antioxidant capacity was assessed by cyclic voltammetry. VCR 1 wine from 2011 stood out for its elemental composition. On the other hand, antioxidant capacity of VCR 101 wines was the highest one for the both vintages. According to the experimental data obtained, all three wines are good source of essential elements and products with a significant antioxidant activity and specific geographical origin.

AB  - The overall aim was to compare the quality of two Montenegrin Merlot wines obtained from new vine clones (VCR1 and VCR 101) along with commercial Merlot wine throughout the consecutive vintages in 2010 and 2011, due to preliminary screening of the potential of novel Merlot vine clones for the wine industry. The content of phenolic compounds was determined by liquid chromatography tandem mass spectrometry (LC-MS/MS). Elemental composition was analysed using inductively coupled plasma optical emission spectrometery (ICP-OES) and inductively coupled plasma mass spectrometery (ICP-MS)]. Additionaly, antioxidant capacity was assessed by cyclic voltammetry. Compared with the commercial one, both wine clones contained higher percent of major phenolic compounds (namely, gallic acid and catechin) with top values recorded for VCR1 both in 2010 (30.46 +/- 0.42 mg/1) and 2011 (28.12 +/- 0.29 mg/1). The same wines were enriched with epicatechin, resveratrol, myricetin and quercetin. Furthermore, VCR 1 wine from 2011 stood out for its elemental composition. On the other hand, antioxidant capacity of VCR 101 wines was the highest one for the both vintages. According to the experimental data obtained, both new Montenegrin Merlot wines represent good candidates for the commercialised wines with an indication of geographical origin.

Surface modification of anatase nanoparticles with substituted catecholate type ligands

AB  - The surface modification of nanocrystalline TiO2 particles (45 Å) with
catecholate-type ligands having electron donating (4-methylcatechol,
4MetCat) and electron withdrawing (4-nitrocatechol, 4NitCat) substituent
groups, was found to alter the optical properties of nanoparticles in a similar
way to catechol (Cat). The formation of the inner-sphere charge-transfer
(CT) complexes results in a red shift of the semiconductor absorption onset
compared to unmodified nanocrystallites, being slightly less pronounced in
the case of electron withdrawing substituent. The investigated ligands have
the optimal geometry for binding to surface Ti atoms, resulting in ring
coordination complexes of catecholate type (binuclear bidentate binding -
bridging) thus restoring six-coordinated octahedral geometry of surface Ti
atoms. The binding structures were investigated by using FTIR
spectroscopy.

AB  - On the basis of LC/UV/MS analysis, 35 compounds from the extracts of seeds of black, red, and white currants (2 cultivars of each) were identified. Black currants cultivars contained protocatechuic acid and p-hydroxybenzoic acid, and traces of nitril containing phenolic acids. The presence of synapoyl glucoside was characteristic for cv. Malling Jewel. Sesquiterpenoid glycosides and carboxymethylindol glycosides were present mainly in white and red currant cultivars. Blackcurrant seeds contained higher amounts of flavonoids, especially rutin, isoquercetin, and taxifolin. The currant seed extracts were tested for in vitro protective effect on chromosome aberrations in peripheral human lymphocytes using cytochalasin-B blocked micronucleus (CBMN) assay. The frequency of MN was scored in binucleated cells, and the cultivars of black currants showed the best antioxidant potential. At a concentration of only 0.17 mg/mL, extract of the seed cv. Malling Jewel effected a decrease in the frequency of MN of 60% compared with control cell cultures. Practical Application: Our results provide evidence of protective effects of currant seed extracts and isolated pure compounds on cytogenetic damages in human lymphocytes. Thus, currant seed extracts could exert beneficial effects in quite a few diseases, for many of the biological actions have been attributed to their antioxidant properties.

The structure elucidations were based on high resolution-mass spectrometry and nuclear magnetic resonance studies. The BSEs and 3 isolated pure compounds were tested for in vitro protective effect on chromosome aberrations in peripheral human lymphocytes using cytochalasin-B blocked micronucleus (MN) assay. The frequency of MN was scored in binucleated cells, and nuclear proliferation index was calculated. Among the tested extracts, the seeds of cv. Thornfree at concentration of 1 mu g/mL exhibit the most prominent effect decreasing the frequency of MN by 62.4%, when compared with the controls cell cultures. Antioxidant potential of pure ellagitannins cannot explain the strong effect of BSEs. The assumption was that better antioxidant effect of BSEs result from synergistic effects of individual compounds contained in the extracts and/or some minor components possessed strong activity.

Protective Effect on Human Lymphocytes of Some Flavonoids Isolated from Two Achillea Species

AB  - This study was conducted to elucidate the in vitro protective effect of five flavonoids [apigenin (1), apigenin-7-O-glucoside (2), centaureidin (3), jaceidin (4) and quercetin (5)] against chromosomal damage in mitogen-induced human lymphocytes. Using the Cytochalasin-B blocked micronucleus (CBMN) assay, in which the biomarker of chromosome breakage and/or chromosome loss is the elevated frequency of micronucleus (MN) in binucleated (BN) cells, the presence of flavonoid 2 in minimal concentration (3 mu g/mL) gave a 35.5% decrease M the frequency of MN when compared with control human lymphocytes. The same concentration of flavonoids 1, 3 and 4, reduced the MN frequency by 24.4%, 28.0% and 28.0%, respectively. Higher concentrations (6 mu g/mL and 10 mu g/mL) seemed less effective. Flavonoid 5 (3 mu g/mL.) induced a slight decrease in MN frequency (5%), while higher doses (6 mu g/mL and 10 mu g/mL) provoked an increase of DNA damage. The comparable values for the cytokinesis-block proliferation index (CBPI) of the tested flavonoids and positive control suggested an inhibitory effect on lymphocyte proliferation. In the DPPH scavenging assay, flavonoids 1-4 demonstrated modest activity, in a dose-dependent manner, compared with the synthetic antioxidants BHT and Trolox, while 5 exhibited comparably high antioxidative activity.

abstract = "This study was conducted to elucidate the in vitro protective effect of five flavonoids [apigenin (1), apigenin-7-O-glucoside (2), centaureidin (3), jaceidin (4) and quercetin (5)] against chromosomal damage in mitogen-induced human lymphocytes. Using the Cytochalasin-B blocked micronucleus (CBMN) assay, in which the biomarker of chromosome breakage and/or chromosome loss is the elevated frequency of micronucleus (MN) in binucleated (BN) cells, the presence of flavonoid 2 in minimal concentration (3 mu g/mL) gave a 35.5% decrease M the frequency of MN when compared with control human lymphocytes. The same concentration of flavonoids 1, 3 and 4, reduced the MN frequency by 24.4%, 28.0% and 28.0%, respectively. Higher concentrations (6 mu g/mL and 10 mu g/mL) seemed less effective. Flavonoid 5 (3 mu g/mL.) induced a slight decrease in MN frequency (5%), while higher doses (6 mu g/mL and 10 mu g/mL) provoked an increase of DNA damage. The comparable values for the cytokinesis-block proliferation index (CBPI) of the tested flavonoids and positive control suggested an inhibitory effect on lymphocyte proliferation. In the DPPH scavenging assay, flavonoids 1-4 demonstrated modest activity, in a dose-dependent manner, compared with the synthetic antioxidants BHT and Trolox, while 5 exhibited comparably high antioxidative activity.",

AB  - A new prenylated flavanonol named seselinonol (1) was isolated from the roots of Seseli annuum, together with the well-known biologically active polyacetylenes falcarinol (2) and falcarindiol (3), and the prenylated furanocoumarin phellopterin (4). Its structure was elucidated by extensive spectroscopic analysis, including HR-ESI-MS, ID- and 2D-NMR. Seselinonol and phellopterin were tested for in vitro protective effect on chromosome aberrations in peripheral human lymphocytes using cytochalasin-B blocked micronucleus (CBMN) assay. The new compound exerted a beneficial effect by decreasing DNA damage of human lymphocytes.

AB  - In this study, volatile compounds were analyzed in five samples of home-made spirit beverage made by the distillation of fermented fruits of cornelian cherry (Cornus mas L.). The major volatile compounds, besides ethanol, identified and quantified were: methanol, acetaldehyde, I-propanol, ethyl acetate, 2-methyl-1-propanol, 1-butanol, amyl alcohols, 1-hexanol and 2-phenyl-ethanol, The minor volatiles were submitted to liquid liquid extraction with dichloromethane and analyzed by gas chromatography and gas chromatography/ /mass spectrometry (GC/MS). A total of 84 compounds were identified. The most abundant compounds were straight-chain free fatty acids, ethyl esters of C(6)-C(18) acids, limonene, 2-phenylethanol and 4-ethylphenol. Most of the compounds found in the Drenja spirits investigated in this study arc similar to those present in other alcoholic beverages.

We evaluated the effects of three different extracts (50%, 80%, and 100% methanol) in doses of 1.4, 4.2, and 8.4 mu g/mL, per 5 mL culture using cytochalasin-B micronucleus (CBMN) assay in peripheral human lymphocytes. The frequency of MN was scored in binucleated (BN) cells. The nuclear proliferation index was also calculated. The distribution of polyphenolic compounds in RSEs was determined using LC/UV/ESI-TOF MS. The identified 37 compounds comprised flavanol monomers and oligomers, as well as varieties of ellagitannin components. Treatment of lymphocytes with RSEs induced a significant decrease in the frequency of micronuclei by 80%. These results demonstrate that the constituents of RSEs may be important in the prevention of oxidative lymphocyte damage by reactive oxygen species and may also reduce the level of DNA damage. These findings support the potential benefits of polyphenolic compounds from raspberry seeds as efficient antioxidants. (C) 2009 Elsevier Ltd. All rights reserved.

AB  - The distribution of polyphenolic compounds in a grape (Vitis vinifera) seed extract (GSE) was determined using LC/ESI-TOF MS, HPLC/DAD, and (13)C-NMR. The 17 identified compounds comprised gallic and protocatechuic acid, catetchin and epicatechin monomers, procyanidin oligomers, and procyanidin gallates. This study addresses the in vitro effects of grape seed extract (GSE) on the frequency of micronuclei with reference to the antioxidant status in human lymphocytes. To establish the most effective protective support, we used four different concentrations of GSE, in the range 1-6 mu g/mL. Treatment of lymphocytes with GSE at a concentration of 2.5 mu g/mL induced a significant decrease in the frequency of micronuclei by 40%, reduction of malonyldialdehyde production by 30%, while a concentration of 5 mu g/mL increased catalase and glutathione S-transferase activity by 10 % and 15 %, respectively. These results demonstrate that GSE may be effective in the prevention of oxidative lymphocyte damage by ROS.
