Fruit Wine and Its Biologically Active Compounds’ Ability in Health Prevention

AB  - Background and objectives: Blackberry is one of the most important berry fruits growing in Serbia. Crops and derived products are rich sources of many natural active compounds, which possess beneficial health effects. In particular, it is possible to point out the role of blackberry and its derived products in hyperglycemia prevention since it is a very common problem among the general population today. Blackberry wine is a product which could have significant beneficial health effects during moderate consumption. The aim of this study was to investigate the α-glucosidase inhibitory activity of blackberry wine and the contribution of some natural compounds to this activity. Methods: Fruit wines were produced through microvinification during controlled fermentation by using pure yeast strain culture. Samples were lyophilized and dissolved in DMSO. The inhibition of α-glucosidase was evaluated by using α-glucosidase and substrate solution, p-nitrophenyl α-Dglucopyranoside. The identification and quantification of some natural compounds was conducted by using UPLC TQ-MS/MS. Results: After lyophilization of the produced blackberry wine, all of the determinations were conducted in four samples. The results for the α-glucosidase inhibitory activity were in the range of 31.5–55.7 µg/mL. The control showed acarbose whit inhibitory activity of 75.3 µg/mL. Moreover, the estimated amount of epicatehin, catehin, chlorogenic, ellagic, and gallic acids and their contribution to the α-glucosidase inhibitory activity of the sample was from 1.7% to 7.7%. Discussion: The obtained results show that blackberry wine is a good inhibitor of α-glucosidase compared to acarbose. The presence of phenolic compounds in wine is due to its extraction from the skin and seeds of blackberry during alcoholic fermentation. Ethanol, which naturally occurs in wine, has a key role in this process. Fruit wine can be considered as a potential functional food. α-glucosidase activity depends on the synergistic and antagonistic effect of natural active compounds in fruit wine.

AB  - The fruit wines from blueberry were made by microvinification procedure. Wines were produced in the absence or presence of sugar and/or enzymatic preparation glycosidase (EPG). Selected phenolic acids were quantified using UPLC/MS-MS analysis. Total phenolic content (TPC) was determined by the Folin-Ciocalteu method. Also, 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing ability of plasma (FRAP) methods were applied. The α-glucosidase inhibitory activity of blueberry wines was also investigated. Wines made with addition of sugar and EPG showed the best results. Phenolic profile and biological activity of blueberry wine depended from microvinification procedure which was used in the production.

The Influence of Cabernet Sauvignon Ripeness, Healthy State and Maceration Time on Wine and Fermented Pomace Phenolic Profile

AB  - The phenolic composition and antioxidant activity of wine and fermented pomace (FP) from Cabernet Sauvignon grapes harvested at three ripening stages were evaluated using LC-MS/MS and spectrophotometric analyses. An investigation of grey mold’s (Botrytis cinerea) influence on wine phenolic content modulation was conducted as well. Finally, the influence of the plant’s ripening stage on the dynamics of the phenolic compounds extracted from wine and FP obtained from fully ripe grapes was evaluated. In this study, the content of catechin, epicatechin, quercetin, and p-coumaric, gallic, and syringic acids was analyzed. Wine and FP were obtained after extended maceration during the spontaneous and inoculated fermentation of fully ripe grapes. When comparing the wine and FP obtained from véraison, fully ripe, and overripe grapes, catechin was the most abundant in wine (40.13 ± 3.25 mg/L) and quercetin in FP (10.96 ± 0.14 mg/kg). A decrease in analyzed phenolic compounds was noticed in wine produced from grapes affected by Botrytis cinerea, and the highest depletion was found for quercetin. The use of a winemaking technique that involved differing maceration periods and inoculation using yeasts as well as spontaneous fermentation significantly modulated the phenolic content of derived wines and FP. The dynamics of the phenolic compounds extracted into wine, evaluated using a principal component analysis (PCA), highlighted contents of catechin and epicatechin. After a decrease in maceration, the PCA revealed a notable content of gallic and syringic acids, as well as quercetin, in samples of FP. This study offers a perspective for future research and the development of functional food with a high content of phenolic compounds originating from red grape products, such as wine and fermented pomace.

Blueberry wine biologically active compounds protect against oxidative stress

AB  - Fruit and derived product represent a rich source of biologically active compounds which exhibit beneficial health effect on human organism. Among fruit especially berries it is important to highlight blueberries. One of derived products with added value from this fruit is wine. The aim of this study was to investigate in vitro activity of blueberry wine by monitoring activities of antioxidant protection enzymes and lipid peroxidation (malondialdehyde level) in isolated rat synaptosomes. Fruit wines were produced in controlled conditions of different microvinifications in which pure culture of selected wine yeast was used. Synaptosomes were isolated from the brain of Wistar albino rats. Analyzed wine samples influenced on the activity of antioxidant protection enzymes. Wine samples also showed ability to decrease malondialdehyde level. Activity for superoxide dismutase in synaptosomes was in range (6.47–7.21 U/mg) while catalase activity was (0.045–0.061 U/mg). Glutathione peroxidase activity was in range (0.0212– 0.0232 U/mg), as well as malondialdehyde level (2.17–2.35 nmol/mg). Obtained results indicate that blueberry wines possess antioxidant properties and abilities to protect against free radicals generated during oxidative stress.

AB  - Significant amount of natural active compounds are present in the fruit. Those compounds exhibit beneficial effect on the human health. Antioxidant properties are very important for health prevention. The aim of this study was to investigate natural active compounds from fruit wines and its activity on enzymes of antioxidant protection in vitro. Fruit wines were produced in controlled conditions during microvinifications. Phenolic profile of fruit wines were obtained by UPLC MS/MS, while enzymatic activity determined by spectrophotometric methods. Fruit wines showed significant content of phenolic compounds among them it is possible to emphasize phenolic acids. Also fruit wines influenced on the activity of enzymes of antioxidant protection which could be used in the prevention of the oxidative stress.

Analyzed wine samples showed significant activity on antioxidant protection enzymes and decreased malondialdehyde level. Results indicate that peach wines showed protective activity against free radicals.

AB  - One of the important tasks of evaluating the integrity of mechanical process elements and structures is to determine the local mechanical properties. In this paper, experimental and numerical analyses of the mechanical behavior of an output inter-heater tube, made of 12H1MF heat-resistant steel, was performed after 200,000 h of exploitation. During exploitation, the tube was exposed to various mechanisms of damage including gas corrosion. The tube was cut from a pipe system during reparations of a thermal power plant, and then cold-deformed by bending to obtain a U-bend tube, which was then used in the experiment. For this purpose, a specimen holder made of structural steel S235 was specifically designed to test such a sample. The U-bend tube was then exposed to the external compressive load during the experiment. Experimental research was based on the application of the 3D digital image correlation (DIC), while a finite element method (FEM) was applied in numerical simulation performed by using the Abaqus software package. The 3D DIC is an optical and contactless experimental method that allows measurements of displacement fields and deformations of geometrically complex structures. The Aramis system was used for the experimental analysis as well as for verification of the numerical model. During the experiment, the von Mises strain field was measured at the top of the U-bend tube, in the tightening zone, as it represents a critical place for crack initiation and propagation during the work of an inter-heater. Based on the obtained results and a comparative analysis of experimental and numerical values of the von Mises strain field at the U-bend tube, deviation of the model predictions of about 18 % was determined. The FEM predicted smaller values of the von Mises strain field compared to the DIC method. This is the result of an incomplete geometry applied in the model due to deformation that occurred in the bend zone of the U-bend tube, loss of material and the tube surface damage due to the influence of gas corrosion during 200,000 h of exploitation. Experimental analysis has confirmed that the U-bend tube, after 200,000 h of exploitation, can remain in service even if it is damaged due to the effect of gas corrosion.
AB  - Jedan od važnih zadataka procene integriteta mašinskih elemenata i konstrukcija je određivanje lokalnih mehaničkih svojstava. U ovom radu izvršeno je eksperimentalno i numeričko ispitivanje mehaničkih svojstava cevi, izlaznog međupregrejača pare, izrađenog od toplotnopostojanog čelika 12H1MF, nakon 200.000 h rada. Cev izlaznog međupregrejača pare je isečena iz cevnog sistema tokom remonta termoelektrane, a potom hladno deformisana savijanjem, u cilju dobijanja cevnog luka, za potrebe eksperimenta. Namenski je konstruisan držač za ispitivanje takvog uzorka. Eksperimentalno istraživanje je bazirano na primeni 3D metode korelacije digitalnih slika, dok je metoda konačnih elemenata primenjena u numeričkoj simulaciji. Metoda 3D korelacije digitalnih slika predstavlja optičku i beskontaktnu eksperimentalnu metodu koja omogućava merenje polja pomeranja i deformacija geometrijski kompleksnih struktura. Za eksperimentalnu analizu kao i verifikaciju numeričkog modela korišćen je sistem Aramis, baziran na metodi korelacije digitalnih slika. Na osnovu dobijenih rezultata i uporedne analize eksperimentalnih i numeričkih vrednosti, koje se odnose na fon Mizesove deformacije na temenu cevnog luka, javlja se odstupanje od oko 18%. Ovo odstupanje nastaje kao rezultat korišćenih metoda koje imaju bitne razlike u njihovim početnim uslovima ispitivanja.

AB  - Complex structure experimental analysis has always been a huge challenge for researchers. Conventional experimental methods (e.g., strain gauges) give only limited data sets regarding measurement on critical areas with high geometrical discontinuities. A 3D Digital Image Correlation method is an optical method that overcomes the limitations of conventional methods and enables full-field displacement and strain measurement of geometrically complex structures. System Aramis, based on Digital Image Correlation method, is used for experimental analysis and numerical model verification in this paper. Investigated complex structure is sphere/cylinder junction on globe valve housing subjected to axial loading. The highest experimentally measured von Mises strain values around 0.15% are recorded on cylinder/sphere intersection. Von Mises strain values on cylindrical and spherical part are several times smaller than on intersection itself. It is important to emphasize that, to the authors best knowledge, this is the first paper showing experimental results of 3D full and strain field of geometrically complex structure (sphere/cylinder intersection) on the intersection itself on pressure equipment. It is proven that 3D Digital Image Correlation method is fast and versatile method for recording strain during loading of complex structures.

abstract = "Complex structure experimental analysis has always been a huge challenge for researchers. Conventional experimental methods (e.g., strain gauges) give only limited data sets regarding measurement on critical areas with high geometrical discontinuities. A 3D Digital Image Correlation method is an optical method that overcomes the limitations of conventional methods and enables full-field displacement and strain measurement of geometrically complex structures. System Aramis, based on Digital Image Correlation method, is used for experimental analysis and numerical model verification in this paper. Investigated complex structure is sphere/cylinder junction on globe valve housing subjected to axial loading. The highest experimentally measured von Mises strain values around 0.15% are recorded on cylinder/sphere intersection. Von Mises strain values on cylindrical and spherical part are several times smaller than on intersection itself. It is important to emphasize that, to the authors best knowledge, this is the first paper showing experimental results of 3D full and strain field of geometrically complex structure (sphere/cylinder intersection) on the intersection itself on pressure equipment. It is proven that 3D Digital Image Correlation method is fast and versatile method for recording strain during loading of complex structures.",

Granular flow in static mixers by coupled DEM/CFD approach

AB  - The mixing process greatly influences the mixing efficiency, as well as the quality and the price of the intermediate and/or the final product. Static mixer is used for premixing action before the main mixing process, for significant reduction of mixing time and energy consumption. This type of premixing action is not investigated in detail in the open literature. In this article, the novel numerical approach called Discrete Element Method is used for modelling of granular flow in multiple static mixer applications (1-3 Komax or Ross mixing elements were utilized), while the Computational Fluid Dynamic Method was chosen for fluid flow modelling, using the Eulerian multiphase model. The main aim of this article is to predict the behavior of granules being gravitationally transported in different mixer configuration and to choose the best configuration of the mixer taking into account the total particle path, the number of mixing elements and the quality of the obtained mixture. The results of the numerical simulations in the static mixers were compared to experimental results, the mixing quality is examined by RSD (relative standard deviation) criterion, and the effects on the mixer type and the number of mixing elements on mixing process were studied. The effects of the mixer type and the number of mixing elements on mixing process were studied using analysis of variance (ANOVA). Mathematical modelling is used for optimization of number of Ross and Komax segments in mixer in order to gain desirable mixing results.

AB  - Globe valves have widespread application in various industrial sectors. Due to this variety, valve is subjected to different working loads (static, dynamic, thermal etc.) that can cause valve failures. In this paper, the analysis is performed on globe valve subjected to external axial loading caused by pipeline dilatations. Experimental analysis was focused on determining local mechanical properties of critical areas, i.e. on local areas of globe valve housing with high geometrical discontinuities. Experimental analysis and numerical model verification were performed using system Aramis based on Digital Image Correlation method. Using full-field experimental results, numerical model behavior was improved and enabled acquiring more accurate results in transition areas that cannot be found in currently available literature. Determining local mechanical properties of critical areas enables better understanding of complex structures such as valve housing and provides an opportunity for further development and improvement for practical industrial application.
