Cadmium and Fullerenes in Liver Diseases

AB  - Oxidative stress (OS) represents an imbalance of free radicals production and antioxidant defense in which the extensive pro-oxidant production may cause damage to cell structures. OS is the major event occurring in pathogenesis of liver disorders, ranging from metabolic to proliferative disorders. Cadmium is a highly toxic and widespread toxicant. Cadmium itself is unable to generate free radicals directly, as cadmium does not undergo redox cycling. However, indirect generation of various radicals involving the superoxide radical, hydroxyl radical, and nitric oxide has been reported. One of the biologically most relevant features of C60 fullerene is the ability to quench various free radicals, behaving as a "free radical sponge." Conversely, photosensitization of C60 yields highly reactive singlet oxygen or superoxide anion. Therefore, the dual property of fullerene to either quench or generate cell-damaging reactive oxygen species could be potentially exploited for their development as cytoprotective or cytotoxic agents. © 2019 Elsevier Inc. AB  - We investigated the biochemical and ultrastructural changes in the liver of the freshwater fish, European perch (Perca fluviatilis), in response to Aphanizomenon flos-aquae bloom in the Gruza Reservoir, Serbia. The activities of total manganese- and copper zinc-containing superoxide dismutase (Tot SOD, Mn-SOD, Cu/Zn-SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione reductase (GR) and biotransformation phase II enzyme glutathione-S-transferase (GST), as well as concentrations of total glutathione (GSH) and sulfhydryl (-SH) groups were examined before and during the bloom period. Mn-SOD activity was significantly higher, while the activities of Cu/Zn-SOD, CAT and GSH-Px and the concentration of the -SH groups were significantly lower during the bloom. The ultrastructure of the liver revealed necrotic and apoptotic damage to the hepatocytes during the bloom period. Our work represents the first study to report the influences of an Aphanizomenon flos-aquae bloom in the Gruza Reservoir on antioxidant biomarkers and on histopathological alterations in the liver of the freshwater fish European perch (Perca fluviatilis).

AB  - We investigated activities of glutathione peroxidase (GSH-Px), glutathione reductase (GR), and the phase II biotransfomation enzyme glutathione-S-transferase (GST) in the foot of three freshwater mussel species: Unio pictorum (Up), Unio tumidus (Ut), and Sinanodonta woodiana (Sw) from the Sava River. Specific and total GSH-Px activity was lower in Sw than in Up and Ut. Total GR activity was higher in Up than in Sw. Specific GST activity was higher in Up than in Ut. Total GST activity was higher in Up than in Ut and Sw. Electrophoretic analysis of proteins shows species specifities between the investigated mussel species. Our study represents the first comprehensive report of the investigated glutathione-dependent enzyme activities in the foot of three freshwater mussel species from the Sava River, Serbia.
