AB  - The aim of this study is to evaluate the size and stability of radiolytically produced silver clusters. The clusters obtained by the gamma irradiation of a mixture of a silver salt and polyvinyl alcohol (PVA) were studied both theoretically and by mass spectrometry. MALDI TOF mass spectrometry of the Ag-n clusters showed the presence of clusters containing between 5 and 29 atoms. Both the theoretical and experimental results of this paper show that the most stable clusters are small clusters with an odd number of atoms. The stability of these clusters is correlated with the clusters electronic configuration. (C) 2012 Elsevier Ltd. All rights reserved.

AB  - In 2011, small mass grave with completely skeletonized remains was discovered in Belgrade suburb. An eyewitness claimed that skeletons belonged to German soldiers killed in WWII. Anthropologists were engaged to investigate whether the skeletal remains correspond to the indicated German group or represent more recent case requiring court trial. Numerous dental restorations were noticed. Owing to the fact that different dental materials were used in dental practice at certain times, the aim of this study was to explore whether analysis of dental restorations could help in identification and estimation of time since death. Inductively coupled plasma optical emission spectrometry revealed that dental fillings corresponded to copper amalgam, conventional silver amalgam, silicophosphate cement, and zinc phosphate cement. Chemical results combined with anthropological and historical facts suggest that the individuals lived before the 1960s in country with well-developed dental service at that time. Therefore, chemical analysis of dental fillings was useful to distinguish between skeletal remains that are too old to be of forensic interest and the remains relevant to legal investigations.

Atomic Force Microscopy Characterization of the External Cortical Bone Surface in Young and Elderly Women: Potential Nanostructural Traces of Periosteal Bone Apposition During Aging

AB  - On the basis of the suggestion that bone nanostructure bears tissue age information and may reflect surface deposition/modification processes, we performed nanoscale characterization of the external cortical bone surface at the femoral neck in women using atomic force microscopy (AFM). The specific aims were to assess age-related differences in bone nanostructure and explore the existence of nanostructural traces of potential bone apposition at this surface. Our findings revealed that the external cortical surface represents a continuous phase composed of densely packed mineral grains. Although the grains varied in size and shape, there was a domination of small grains indicative of freshly deposited bone (mean grain size: young, 35 nm; old, 37 nm; p. 0.05). Advanced quantitative analysis of surface morphological patterns revealed comparable roughness and complexity of the surface, suggesting a similar rate of mineral particle deposition at the surface in both groups. Calcium/phosphorus ratio, a measure of bone tissue age, was within the same range in both groups. In summary, our AFM analyses showed consistent nanostructural and compositional bone features, suggesting existence of new bone at the periosteal bone surface in both young and elderly women. Considering observed age-related increase in the neck diameter, AFM findings may support the theory of continuous bone apposition at the periosteal surface.

AB  - A primer is a strand of nucleic acid that serves as a starting point for DNA
synthesis. DNA, as a large molecule, is very difficult for analysis by means of mass
spectrometry. However, primer, as a short sequence of nucleic acid, is much more
convinient for e.g. MALDI TOF MS analysis. Mass spectrometric experiment
resulted in quality MALDI TOF spectra of 3 primers. Compound 3-HPA (3-
hydroxypicolinic acid) showed the best results as matrix. AFM studies were also
conducted, for both pure primer samples and hybrids of primer molecule with
single wall carbon nanotubes (SWNT). Thus, obtained functionalisation of SWNT
with DNA primers was confirmed by AFM imaging. AFM images clearly showed
wrapping of DNA structures around nanotube „template“. Functionalisation of
SWNT is very important for potential applications of nanotubes in biomedical
field. MALDI TOF mass spectrometry, in combination with AFM imaging, proved
its great potential in analysis of short DNA sequences, and indicated the possibility
of investigating more complex DNA structures.

AB  - Collagens, one of the most abundant on the Earth, are family of proteins which constitute the basis of connective tissue (extracellular matrix) in multicellular organisms. We used collagen type II from bovine Achilles tendon. In further work, collagen has been studied by transmission electron microscopy (TEM) and atomic force microscopy (AFM). Using TEM we successfully obtained images of collagen and whole collagen fibrils. Using AFM we captured images of whole collagen as well as images of fragments from collagenase-treated collagen (it was digested with collagenase from Clostridium histolycium). AFM images of collagenase-treated collagen showed many fibrils grouping into large bundles – collagen fiber. Based on obtained images we studied surface morphology, shape and length of fragments from collagenase-treated collagen.

AB  - Collagen is the most frequently occuring fibrillar protein in mammals, which can be found in cartilage, tendon, bone, ligament, skin, etc. The structure of collagen is based on a complicated helix structure of single chains of amino acids (most abundant is glicine-Gly, proline-Pro and hydroxyproline-Hyp) which are connected by hydrogen bonds. We tried to establish MALDI method to get better resolution of collagen’s mass spectra. Digested collagen (type II from bovine Achilles tendon which was digested with collagenase from Clostridium histolycium) has been analysed on MALDI-TOF MS in order to find peptide’s fragments that are characteristic for collagen. In mass spetra we found peaks of peptides, which is highly indicative for collagen (GlyPro-Hyp, Gly-Pro-Asp, Gly-Pro-Glu ect.) We hope that it should be possible to obtain MS analysis and structure characterization of collagen by MALDI-TOF/TOF in future.

AB  - Bone is composed of two phases. The organic phase is made of collagen fibrils assembled in broad fibers acting as a template for mineralization. The mineral phase comprises hydroxyapatite (HAP) crystals grown between and inside the collagen fibers. We have developed a material using functionalized carbon nanotubes, carbon fibers and glasy carbon as scaffold to initiate in vitro mineralization. Mentioned carbon materials are functionalized with carboxylic groups prior to decorating. All samples were dispersed in ultra-pure water and incubated for 2 weeks in a synthetic body fluid with dispersed HAP, in order to induce the calcification of the functionalized carbon materials. Atomic force microscopy (AFM) and transmision electron microscopy (TEM) showed that Ca2+ and PO4 3- ions were deposited as round-shaped nodules. Raman spectroscopic studies confirmed the HAP formation, and image analysis made on TEM pictures showed that HAP were packed around and inside the carbon materials together. All three types of investigated carbon materials leading to successful calcification of collagen. This suggests that it takes the presence of carbon materials to prevent it from calcification of collagen, independent of the structures of the material.

AB  - In this study we performed the dynamic and static scintigraphy of the domestic dog, using the newly synthesized radiopharmaceutical, [Tc-99m(CO)(3)(H2O)(3)]-C-60(OH)(22-24). In the current study, an advanced one-step method for the functionalization of fullerenol by Tc-99m is described. Optical properties of as-prepared samples and the mechanism responsible for the functionalization were investigated using UV-VIS and FTIR spectroscopy, respectively. Also, the presence of the Tc complex on fullerenol was confirmed by using the energy dispersive X-ray spectroscopy, HPLC and MALDI TOF techniques. This simple and effective method of producing a new radiopharmaceutical is of interest not only for its application in various areas of technology and biology, but also for investigating its potential use in radiation technology for nanoengineering of materials. With dynamic scintigraphy, performed during 30 minutes (120 frames, 15 sec per frame), we obtained ratios of heart, liver and spleen counts: 222/249/168; 178/320/217; 120/348/239 respectively. By static scintigraphy after 1 hour, we detected the activity in heart, liver, spleen and intestines. After 4 hours, the radiopharmaceutical activity was detected in salivary glands. The detection after 21 hours showed the activity in kidneys and urinary bladder, while the activity in intestines was absent. After 24 hours, we detected the activity in liver, spleen, kidneys and urinary bladder. Pharmacokinetic investigations performed in this study are of key interest for the further fullerenol in vivo research.

Nanostructure and mineral composition of trabecular bone in the lateral femoral neck: Implications for bone fragility in elderly women

AB  - Despite interest in investigating age-related hip fractures, the determinants of decreased bone strength in advanced age are not clear enough. Hitherto it has been obscure how the aging process affects the femoral neck nanostructure and composition, particularly in the lateral subregion of the femoral neck, which is considered as a fracture-initiating site. The femoral bone samples used in this study were obtained at autopsy in 10 women without skeletal disease (five younger: aged 20-40 years, and five elderly: aged 73-94 years). Atomic force microscopy (AFM) was applied to explore the mineral grain size in situ in young vs. old trabecular bone samples from the lateral femoral neck. The chemical compositions of the samples were determined using inductively coupled plasma optical emission spectroscopy and direct current argon arc plasma optical emission spectrometry. Our AFM study revealed differences in trabecular bone nanostructure between young and elderly women. The mineral grain size in the trabeculae of the old women was larger than that in the young (median: 95 vs. 59 nm), with a particular bimodal distribution: 45% were small grains (similar to the young) and the rest were larger. Since chemical analyses showed that levels of calcium and phosphorus were unchanged with age, our study suggests that during aging the existing bone mineral is reorganized and forms larger aggregates. Given the mechanical disadvantage of large-grained structures (decreased material strength), the observed nanostructural differences contribute to our understanding of the increased fragility of the lateral femoral neck in aged females. Moreover, increasing data on mineral grains in natural bone is essential for advancing calcium-phosphate ceramics for bone tissue replacement. (C) 2011 Acts Materialia Inc. Published by Elsevier Ltd. AB  - In this study, we analysed pellets from long-eared owls (Asio otus) collected from four localities in Belgrade (Serbia). The pellets contained the remains of prey, namely voles (Arvicola terrestris) and field mice (Apodemus agrarius). The concentrations of 14 elements (Ca, P, Mg, Na, K, Fe, Zn, Sr, Ba, Mn, Ti, Cu, Si, B) were evaluated in whole pellets and in samples containing only bone tissue, which were dissected from the whole pellet. The increased levels of certain elements, including Mn, Zn, Ba, Cu and radioactive K-40, indicate contamination of the soil by various sources, such as industrial plants and agricultural practices. From the results presented in this article, we suggest that the analysis of owl pellets may indicate the quality of the local environment.

AB  - In this paper, an advanced method of one-step functionalization of single and multi walled carbon nanotubes (SWCNTs and MWCNTs) using gamma-irradiation was described. Two synthesis procedures, related with different reduction species, were employed. For the first time, poly(vinyl alcohol) PVA is successfully utilized as a source to reduce silver (Ag) metal ions without having any additional reducing agents to obtain Ag nanoparticles on CNTs. The decoration of carbon nanotubes with Ag nanoparticles takes place through anchoring of (PVA) on nanotubes surface. Optical properties of as-prepared samples and mechanism responsible for the functionalization of carbon nanotubes were investigated using UV-vis and FTIR spectroscopy, respectively. Decorated carbon nanotubes were visualized using microscopic techniques: transmission electron microscopy and scanning tunneling microscopy. Also, the presence of Ag on the nanotubes was confirmed using energy dispersive X-ray spectroscopy. This simple and effective method of making a carbon nanotube type of composites is of interest not only for an application in various areas of technology and biology, but for investigation of the potential of radiation technology for nanoengineering of materials. AB  - Carbon nanotubes are unique one-dimensional macromolecules with promising applications in biology and medicine. Since their toxicity is still under debate, here we present a study investigating the genotoxic properties of purified single wall carbon nanotubes (SWCNTs), multiwall carbon nanotubes (MWCNTs), and amide functionalized purified SWCNTs on cultured human lymphocytes employing cytokinesis block micronucleus assay and enumeration of gamma H2AX foci as a measure of double strand breaks (DSBs) of the DNA in normal human fibroblasts. SWCNTs induce micronuclei (MN) formation in lymphocytes and decrease the proliferation potential (CBPI) of cells. In a fibroblast cell line the same dose of SWCNTs induces gamma H2AX foci 2.7-fold higher than in a control. Amide functionalized purified SWCNTs behave differently: they do not disturb the cell proliferation potential of harvested lymphocytes, but induce micronuclei to a higher extent than SWCNTs. When applied on fibroblasts, amide functionalized SWCNTs also induce gamma H2AX foci, 3.18-fold higher than the control. The cellular effects of MWCNTs display the broad spectrum of clastogenic properties seen as the highest incidence of induced lymphocyte micronuclei and anaphase bridges among nuclei in binucleated cells. Surprisingly, the incidence of induced gamma H2AX foci was not as high as was expected by the micronucleus test, which indicates that MWCNTs act as clastogen and aneugen agents simultaneously. Biological endpoints investigated in this study indicate a close relationship between the electrochemical properties of carbon nanotubes and observed genotoxicity.

The endohedral fullerenes were generated using a mass spectrometer with a triple rhenium filament as an ion source. The ionisation energies (IE) determined were: 8.52 +/- 0.25 eV for Tc-99m"C-60 and 9.57 +/- 0.25 eV for Tc-99m"C-70.

Molar-mass distribution of urea-formaldehyde resins of different degrees of polymerisation by MALDI-TOF mass spectrometry

AB  - This paper describes some results obtained in an investigation of urea-formaldehyde (UF) resins of different degrees of polymerisation by matrix-assisted laser desorption/ionisation time-of-flight (MALDI-TOF) mass spectrometry (MS). MALDI-TOF MS proved to be an appropriate technique for analyzing these types of polymers, bearing in mind that the results of the analysis correspond with previous physical and chemical measurements. This technique enables a relatively swift determination of the degree of polymerrisation through the monitoring of key changes in the structure of a polymer. Thus, in the analysis of UF resins, it may be possible to monitor a decrease in the intensity of the monohydroxymethyl urea (MMU) signal, which corresponds to an increase of the mass spectra values in the mass range of higher homologues, above 1000 g mol(-1). A noticeable difference concerns the signal intensities in the higher mass ranges (up to 1400 g mol(-1)), which corresponds to more branched and longer homologues of the polymers. Especially, a significantly more intensive signal of MMU was registered. The average molecular weight (MW) of the examined samples was between 936 and 1324 g mol(-1), with a maximal deviation of 20 %, depending on the ratios of the reactants.

AB  - Using the ion implantation technique (introducing negatively charged fullerene into a low temperature lithium plasma column by a strong axial magnetic field) endohedral fullerenes Li"C-70, Li-2"C-70 and Li-3"C-70 were produced. Mass spectral studies (both TIMS-thermal ionization mass spectrometry and MALDI TOF MS - matrix assisted laser desorption ionization time-of-flight mass spectrometry) of these endohedral fullerenes provided detailed structural and reactivity information about these unusual species. The fragmentation of the obtained ions is shown to be by a multiple C-2 loss (shrink-wrap mechanism). According to the fact that more atoms in the fullerene make the produced endohedral less stable, Li-3"C-70 shows a big degree of instability. The presence of Li-3"C-70 was observed on MALDI TOF MS.

AB  - Ionization energies of non-stoichiometric LinFn-1 (n =3,4, 6) clusters determined by a thermal ionization mass spectrometry (TIMS) were 4.2 +/- 0.2 eV for Li3F2, 4.3 +/- 0.2 eV for Li4F3 and 4.1 +/- 0.2 eV for Li6F5. The ionization energy of Li6F5 cluster was obtained experimentally for the first time. The ionization energies of Li3F2 and Li4F3 are in correlation with the results obtained by photoionization time-of-flight mass spectrometry. The determined ionization energies are comparable with theoretical ionization energies calculated by ab initio method. The theoretical predictions supported that the most stable isomers of a non-stoichiometric cluster LinFn-1 (n = 3 and n = 4) in which the excess electron localizes on a specific site have a segregated electronic structure composed of the metallic part and ionic part. AB  - Experimental investigations of the influence of excess oxygen coefficient, H2O/coal mass ratio using high-temperature steam, mean mass diameter of pulverized coal and coal size fraction on basic characteristics of coal gasification were performed. Experiments were carried out on a laboratory scale (0.09 m i.d. x 1.5 m high) coal gasification apparatus with lignite type of coal. Influence of steam was realized through comparison of results obtained from experiments with (H2O/Coal = 0.287 kg kg(-1)) and without steam addition (H2O/coal = 0.024 kg kg(-1)). High values of carbon conversion, obtained both for finely ground and for coarse pulverized coal points to the easiness of lignite gasification, i.e. to its high suitability for gasification. AB  - The biological behavior of fullerene derivatives shows their considerable potential for medical application. Polyhidroxilated, water soluble fullerenol C-60(OH)(22). used in these studies, was synthesized by procedure complete substitution of bromine atoms from polybromine derivatives C60Br24 in alkaline media. In order to investigate the pharmacological behaviour and metabolism of the fullerenol, quickly and conveniently, we present the way and possibilities for its labelling with Tc-99m (T-1/2=6.02h, E-n=141keV), which bind to the outside, leaving the cage intact. Two different labelling approaches with technetium-99m were performed: direct labelling by tin (II) chloride method and labelling with Tc-99m(I) using [Tc-99m(CO)(3)(H2O)(3)](+) precursor. The radiochemical studies results have shown that content of free (TcO4-)-Tc-99m in the samples was depend on fullerenol-stannous chloride ratio and increased if ratio was higher. HPLC quality control results of Tc-99m(CO)(3)-[C-60(OH)(22)] have shown that the labelling yield, in alkaly media, was 95.35%. Protein binding to HA was species-dependent process. The biological behaviour of labelled fullerenol were different in dependence of labelling method and the time of sacrificed of animals. From recent investigations it can be concluded that oxidation state in technetium coordination complexes has a great impact on in vitro and in vivo behaviour of the these complexes.

Surface characterization of silver and palladium modified glassy carbon

AB  - In this work, the influence of silver and palladium on the surface of undoped, boron doped and phosphorus doped glassy carbon has been studied. The silver and palladium concentrations in solution, after metal deposition, were measured by atomic absorption spectrophotometer. The morphology of metal coatings was characterized by scanning electron microscopy. In order to investigate the nature and thermal stability of surface oxygen groups, temperature-programmed desorption method combined with mass spectrometric analyses, was performed. The results obtained have shown that silver and palladium spontaneously deposit from their salt solutions at the surface of glassy carbon samples. Silver deposits have dendrite structure, whilst palladium forms separate clusters. The highest amount of both silver and palladium deposits at the surface of sample containing the highest quantity of surface oxide complexes. It has been concluded that carboxyl groups and structure defects are responsible for metal reduction. Calculated desorption energies have shown that the surface modification by metal deposition leads to the formation of more stable surface of undoped and doped glassy carbon samples.

AB  - In an attempt to purify MWNT in mild conditions, we have applied two different methods of purification. The first method of purifying the material was a combination of relatively often used methods, such as etching and acid treatment, and the second method was a relatively new one, using an organic polymer PmPV. AB  - Molecules of LinX (n = 2,3; X = Cl, Br, I) were examined with a magnetic sector mass spectrometer by surface ionization using a triple rhenium filament impregnated with fullerene (C-60). The ionization energies obtained for Li2Cl, Li2Br and Li2I Molecules are 3.8 +/- 0.1, 3.9 +/- 0.1 and 4.0 +/- 0.1 eV. respectively. The first ionization energy of Li2Cl is documented, while there are no literature data for the ionization energies of Li2Br and Li2I. The molecules of Li3Cl, Li3Br and Li3I were detected experimentally for the first time with ionization energies of 4.0 +/- 0.1, 4.1 +/- 0.1 and 4.1 +/- 0.1 eV, respectively. The ionization energies of LinX (n = 2, 3; X = Cl, Br, I) are in correlation with the theoretical prediction of their hyperlithiated configurations. Copyright (c) 2006 John Wiley and Sons, Ltd.
