Antimicrobial potential of irrigants based on essential oils of Cymbopogon martinii and Thymus zygis towards in vitro multispecies biofilm cultured in ex vivo root canals.

AB  - OBJECTIVE The objective was to determine the antibiofilm efficacy of Cymbopogon martinii and Thymus zygis essential oils and to estimate the disruption potential of oil-based endodontic irrigants on the multispecies biofilm formed in the root canals of extracted teeth. DESIGN The essential oils were characterized (GC-MS), while their antibacterial and cytotoxic properties were detected by microdilution and MTT assays. Particles sizes and polydispersity indices were determined for the irrigants. The isolates from root canals of pediatric patients were identified (MALDI-TOF). The multispecies biofilms were formed from Streptococcus mitis, Streptococcus sanguinis and Enterococcus faecalis. Antibiofilm properties of the essential oils and irrigants were determined by crystal violet and plate counting assay. Statistical analysis was performed by one-way ANOVA. RESULTS The screening of biofilm biomass revealed a disruption potential of C. martinii essential oil. The plate counting assay showed the efficacy of both oils in diminishing cell viability: high in biofilms (reduction of log10CFU was 2.75-2.87) and moderate in the planktons formed above. The essential oil-based irrigants showed the same antibiofilm activity as in the control (1.5 % sodium hypochlorite) for C. martini and almost 2-fold higher for T. zygis. Successive irrigations with 1.5 % sodium hypochlorite, saline and an oil-based irrigant was more efficient for C. martini than for the control (reductions of log10CFU was 1.69 and 1.14, respectively). CONCLUSIONS Notable activities achieved by essential oils and oil-based irrigants, particularly regarding C. martini, against the biofilm consisting of S. mitis, S. sanguinis, and E. faecalis, are expected to encourage further research in endodontics.

Antibacterial effects of new endodontic materials based on calcium silicates

AB  - Background/Aim. The main task of endodontic treatment is to eliminate pathologically altered tissue, to disinfect root canal space and to obtain its three-dimensional hermetic obturation. The main purpose of this study was to evaluate antimicrobial activity of new endodontic nano-structured highly active calcium silicates based materials albo-mineral plyoxide carbonate aggregate (ALBO-MPCA) and calcium silicates (CS) in comparison to mineral trioxide aggregate (MTA+) and UltraCal XS (CH). Methods. The antimicrobial activity of materials was tested against Staphylococcus aureus (ATCC 25923) and Enterococcus faecalis (ATCC 14506) strains, and following clinical isolates: Rothia dentocariosa, Enterococcus faecalis, Staphylococcus aureus, Streptococcus anginosus and Streptococcus vestibularis using a double layer agar diffusion test. The pH measurements were performed using the pH meter. Total amount of released ions was determined by inductively coupled plasma optical emission spectrometry (ICP-OES). Results. All tested materials showed the best antibacterial potential after 1 h of incubation. After 3h and 24h of the incubation period, the antibacterial potential of all tested materials were similar. The Agar diffusion test showed that ALBO-MPCA, CS and MTA+ had similar inhibition zones (p > 0.05), except in the activity against Staphylococcus aureus where ALBO-MPCA showed better antimicrobial properties than MTA+ in 3h and 24h of the incubation period (p < 0.05). Following 24h of the incubation, the inhibition zones were the strongest with CH against Staphylococcus aureus (16.67 ± 2.34 mm) followed by ALBO-MPCA (14.67 ± 1.21 mm) and the weakest with CS against Enterococcus faecalis (6.50 ± 1.76 mm). CH showed the highest pH, followed by ALBO-MPCA, CS and MTA+. Conclusion. The expressed antibacterial effects indicate that materials based on nano-structured highly active calcium silicates represent effective therapeutic agents for root canal obturation in one-visit apexification treatment, therefore they are recommend for further examination and clinical trials as they are proposed for MTA substitution. © 2019, Inst. Yield, quality and safety of yellow gentian roots produced under dry-farming conditions in various single basal fertilization and planting density models

AB  - Yellow gentian (Gentiana lutea ssp. symphyandra (Murb.) Hayek) is a high-mountainous perennial spontaneously growing on meadows and open slopes of the Eastern part of the Alps and the Balkan Peninsula. While the excessive exploitation endangered its survival in the nature in many European countries, the orientation to its large-scale production enables its protection at natural stands and satisfaction of increasing market demands for its roots, a highly-valuable herbal drug (Gentianae radix). During the six-year field trial established with Yellow gentian nursery plants on black water-air permeable and biodegradable film, in dry farming conditions of Mountain Tara (Serbia), the influence of a sigle basal application of farm yard manure and mineral fertilizer, at different planting densities (11.1, 13.3 and 16 plants m −2 ) on produced yield (roots), has been investigated. Single dose fertilization positively influenced the yields but did not provide optimal supplies for cultivated crop in the second part of experiment. It depended on crop age, planting density but also the climatic conditions. In the second part of plant production period, particulary in denser establishemnts, additional fertilization should be applied. Providing the appropriate amounts of fertilizers, Yellow gentian can be successfully grown even in the densest planting model (16 plants m −2 ), which is quite important in respect to the economic feasibility of cultivation. Similar yields achieved in mineral and organic treatments (4.51 kg m −2 and 4.85 kg m -2 of fresh root, respectively), suggest the roots might be successfully produced in both, conventional and organic cultivation model. In attempt to estimate impact of fertilization treatments on chemical quality of produced raw material (Gentianae radix) the content of several pharmacological constituents (loganic acid, swertiamarin, gentiopicroside, sweroside, amarogentin and isogentisin) was evaluated; gentiopicroside and loganic acid were the most dominant ones, regadless the fertilization, whereas the content of isogentisin was significantly increased in both, organic and mineral fertilization model, and content of sweroside, only in mineral model. With regard to the safety of produced raw material, the contents of biogenic (Fe, Mn, Zn, Cu) and non-biogenic (Pb) trace elements were analyzed; all trace elements were within the safty limits except the Cd content which slightly exceeded the limit. © 2019 Elsevier B.V.

AB  - U radu je prikazano poređenje rezultata ispitivanja sadržaja radionuklida u uzorcima
građevinskog materijala merenjem istog uzorka koji je pripremljen za merenje na četiri
načina. Najpre je izvršeno poređenje rezultata ispitivanja uzorka koji je grubo usitnjen
(ručno razbijanje i usitnjavanje), sa rezultatima fino usitnjenog granita (mašinskim
mlevenjem u drobilici). Fino usitnjen granit je, potom, raspoređen u tri posude različite
veličine i geometrije: Marineli posuda zapremine 1 l, Marineli posuda zapremine 0,5 l i
cilindrična plastična posuda zapremine 0,2 l, sa ciljem da se proveri minimalna
količina uzorka neophodna za dobijanje prihvatljivih rezultata ispitivanja.
AB  - Laboratory for the radioactivity measurement performed analysis of the content of the
radionuclides in the sample of the building material – Granite Balmoral Rosso. The
sample (control), was measured in three different size containers (in order to verify the
possibility of using a small amount of the sample if the test results are acceptable). In
this paper, comparison of examination results obtained from the sample which is
roughly triturated (manual technique of breaking) and finely grounded (grinding in
mill), was demonstrated.
