AB  - Thirty days before expected time of parturition, 20 Holstein cows were divided into −Cr and +Cr groups. From day 25 before parturition (BP) up to day 30 after parturition (AP), +Cr cows received 10 mg of Cr (chromium-enriched yeast) daily. Muscle and adipose tissue samples were taken at days −30, −10, +7 and +10 related to parturition, when body condition score (BCS) was also determined. Hepatic tissue samples were taken at days −10 and +7. Tissue samples were used for determination of the insulin signalling pathway protein expressions. Intravenous glucose tolerance test (IVGTT) was performed at days −28, −7, +10 and +30. Milk yield was recorded during first 14 weeks AP. Milk composition was obtained at days 7 and 28 AP. At day 10 BP, protein content of β-subunit of insulin receptor (IRβ) was significantly higher (p ˂ 0.05) in muscle, and phosphorylation of insulin receptor substrate 1 at serine 307 (pIRS-1 Ser307) was significantly lower (p ˂ 0.05) in hepatic tissue of +Cr group. After parturition, pIRS-1 Ser307 was significantly lower in muscle tissue at days 7 and 28 (p ˂ 0.05 and p ˂ 0.001, respectively), while phosphorylation of Akt at serine 473 (pAkt Ser473) was significantly higher (p ˂ 0.01) in hepatic tissue at day 7 AP in +Cr group. Chromium had opposite effect on insulin kinetics during IVGTTs obtained BP and AP. Insulin secretion was significantly reduced at day 7 BP and significantly enhanced at day 10 AP, when NEFA concentration was also significantly increased. Milk yield and ECM value were depressed in +Cr group. DMI and BCS were significantly enhanced in +Cr group at day 7 BP. In conclusion, chromium modulates insulin signalling pathway in dairy cows, but targeted signalling molecules are different in antepartal then post-partal period, probably due to duration of exposure to chromium and different energy status between those periods.

AB  - Циљ ове докторске дисертације је био да се испита утицај органског комплекса тровалентног хрома на високомлечне краве у периоду касног гравидитета и ране лактације, као и његов утицај на млечност. Тридесет дана пре очекиваног термина тељења је одабрано 20 крава холштајн расе које су подељене у две једнаке групе контролну (–Cr) и огледну (+Cr). У периоду од 25. дана пре очекиваног термина тељења до 30. дана после тељења, +Cr група крава је свакодневно перорално са јутарњим оброком добијала 10 mg, односно 5 g препарата са органски везаним хромом (0,2 % органског препарата тровалентног хрома везаног у квасцу (Co-Factor III Cr Yeast, Alltech).Интравенски тест толеранције глукозе (IVGTT) је урађен на укупно 12 крава (n=6 крава по групи) четири пута током испитиваног периода и то 28. и 7. дана пре очекиваног термина тељења као и 10. и 30. дана после тељења. Крв је узимана 0., 15., 30., 60., 90., 120. минута теста за одређивање концентрација глукозе и инсулина.Коришћењем података добијених током IVGTT урађена је и кинетика глукозе и инсулина. У узорцима крви узетим 0. минута IVGTT одређена је, поред концентрација глукозе и инсулина, концентрација NEFA (non-esterified fatty acids), BHBА (beta hydroxybutyric acid), укупних протеина, албумина, урее, холестерола, триглицерида (TG), укупног билирубина (TB), Mg, Ca, P и кортизола, као и активности AST и GGT. У циљу испитивања инсулинске резистенције периферних ткива обрачунат је RQUICKI (Revised Quantitative Insulin Sensitivity Check Index) који је показатељ осетљивости ткива на инсулин. Узорци мишићног и масног ткива су узимани биопсијом четири пута у току огледног периода и то 30. и 10. дана пре очекиваног термина тељења као и 7. и 28. дана после тељења, када је урађена ипроцена телесне кондиције (BCS-body condition score)...
AB  - The objective of this study was to investigate the effects of peroral chromium administration on the on high-yielding cows metabolism during the period of late gestation and early lactation, as well as its impact on milk yield. Thirty days before the expected time of parturition, 20 Holstein cows were chosen and divided into two equal groups: control (–Cr) and experimental (+Cr). From day 25 before expected time of parturition up to day 30 after parturition, +Cr received 10 mg of Cr or 5 g per day in the form of chromium-enriched yeast (0.2 % Cr3+ Co-factor III chromium yeast, Alltech).Intravenous glucose tolerance test (IVGTT) was performed on a 12 cows (n = 6 cows per group) four times during the examined period, on days 28th and 7th before the expected time of parturition, as well as on the 10th and 30th day after calving. The blood was taken at 0., 15., 30., 60., 90., 120. minutes of the test to determine the glucose and insulin levels.Using the data obtained during IVGTT, kinetics of glucose and insulin were also performed. The concentrations of non-esterified fatty acids (NEFA), BHBA (beta hydroxybutyric acid), total proteins, albumin, urea, cholesterol, triglycerides (TG), total bilirubin (ТВ), Mg, Ca, P and cortisol, as well as activities of AST and GGT were measured in blood samples taken at minute 0. of IVGTT. In order to examine the peripheral tissue insulin resistance, RQUICKI (Revised Quantitative Insulin Sensitivity Check Index) was calculated. Muscle and adipose tissue samples were taken by biopsy four times during the trial period, 30 and 10 days before the expected time of parturition as well as on the day 7th and 28th day after calving, in time when the body condition score (BCS) was also determined. Hepatic tissue samples were taken twice during the trial period, 10 days before expected time of parturition, and 7 days after calving in order to determine the protein expression of the insulin signal pathway molecules. Milk yield wasrecorded during the first 14 weeks of lactation...

AB  - The objective of this study was to investigate the effects of peroral administration of chromium-enriched yeast on glucose tolerance in Holstein calves, assessed by insulin signaling pathway molecule determination and intravenous glucose tolerance test (IVGTT). Twenty-four Holstein calves, aged 1 month, were chosen for the study and divided into two groups: the PoCr group (n = 12) that perorally received 0.04 mg of Cr/kg of body mass daily, for 70 days, and the NCr group (n = 12) that received no chromium supplementation. Skeletal tissue samples from each calf were obtained on day 0 and day 70 of the experiment. Chromium supplementation increased protein content of the insulin beta-subunit receptor, phosphorylation of insulin receptor substrate 1 at Tyrosine 632, phosphorylation of Akt at Serine 473, glucose transporter-4, and AMP-activated protein kinase in skeletal muscle tissue, while phosphorylation of insulin receptor substrate 1 at Serine 307 was not affected by chromium treatment. Results obtained during IVGTT, which was conducted on days 0, 30, 50, and 70, suggested an increased insulin sensitivity and, consequently, a better utilization of glucose in the PoCr group. Lower basal concentrations of glucose and insulin in the PoCr group on days 30 and 70 were also obtained. Our results indicate that chromium supplementation improves glucose utilization in calves by enhancing insulin intracellular signaling in the skeletal muscle tissue.

AB  - Cortisol levels were measured in hair, blood and milk in two different cattle breeds, kept under different breeding conditions and with different genetic merit for milk production. Cows and heifers of Holstein and Busha breeds were selected for the study. Cortisol concentration was determined by immunoassays. Cortisol accumulation was determined in proximal (close to the skin) and distal (far from the skin) segments of the hair shaft. The influence of hair colour and washing prior to extraction and analysis was also examined in order to establish additional factors that may have an impact on hair cortisol concentrations. Concentrations of cortisol determined in the proximal and distal segments of the shaft were significantly higher in Holstein than Busha cows and heifers (P LT 0.05 and P LT 0.01, respectively). In Holstein cows, no significant difference was found between concentrations in black and white hair. In hair washed with isopropanol, cortisol concentration was significantly lower compared to unwashed hair (P LT 0.01). Thus, cortisol concentration in hair varies with the technique of hair processing (washing), but not with colour in Holstein cows. Blood serum cortisol concentrations in Holstein cows and heifers were significantly higher than in Busha cows and heifers, (P LT 0.01 and P LT 0.05, respectively). Milk cortisol in Holstein cows was significantly higher than in Busha cows (P LT 0.05). The higher cortisol concentrations in Holstein cows are assumed to be the result of intensive breeding and physiological adaptation to high milk production.

Olanzapine alleviates oxidative stress in the liver of socially isolated rats

AB  - Olanzapine, an antipsychotic drug, is used to treat depressive disorder, but its effects on the liver, the main site of drug metabolism, still remain elusive. We studied the effects of 3 weeks of olanzapine treatment (7.5 mg/kg per day) on the malondialdehyde (MDA) and protein carbonyl (PCO) contents, protein expression of copper/zinc superoxide dismutase (CuZnSOD), and activity of total superoxide dismutase (SOD), as well as catalase (CAT) protein expression and activity levels in the liver cytosol of rats exposed to 6 weeks of chronic social isolation (CSIS), which causes depressive- and anxiety-like behaviors. Increased cytosolic MDA in CSIS rats (vehicle-or olanzapine-treated) indicated hepatic oxidative stress. Increase in PCO and CAT activity associated with unchanged total SOD activity following CSIS also confirm the presence of oxidative stress. Chronic olanzapine treatment in CSIS prevented increase in PCO without an effect on MDA content. Increased SOD activity in olanzapine-treated (controls and CSIS) groups compared with corresponding vehicle-treated groups and decreased CAT activity in olanzapine-treated CSIS rats compared with vehicle-treated CSIS group was found. The data suggest that chronic olanzapine treatment has a protective effect on hepatic protein oxidation and improves antioxidant defense. The beneficial effects of olanzapine may be due to its free radical scavenging properties and antioxidant activity.

AB  - We investigated the hypothesis that obesity in dairy cows enhanced expression of proteins involved in hepatic fatty acid uptake and metabolism. Sixteen Holstein-Friesian close-up cows were divided into 2 equal groups based on their body condition score (BCS) as optimal (3.25 LT = BCS LT = 3.5) and high (4.0 LT = BCS LT = 425). Intravenous glucose tolerance test (GTT) and liver biopsies were carried out at day 10 before calving. Blood samples were collected before (basal) and after glucose infusion, and glucose, insulin and non-esterified fatty acid (NEFA) levels were determined at each sample point. In addition, beta-hydroxybutyrate and triglycerides levels were measured in the basal samples. The liver biopsies were analyzed for total lipid content and protein expression of insulin receptor beta (IR beta), fatty acid translocase (FAT/CD36) and sterol regulatory element-binding protein-1 (SREBP-1). Basal glucose and insulin were higher in high-BCS cows, which coincided with higher circulating triglycerides and hepatic lipid content. Clearance rate and AUC for NEFA during GTT were higher in optimal-BCS cows. The development of insulin resistance and fatty liver in obese cows was paralleled by increased hepatic expression of the IR beta, CD36 and SREBP-1. These results suggest that increased expression of hepatic CD36 and SREBP-1 is relevant in the obesity-driven lipid accumulation in the liver of dairy cows during late gestation. Low intensity exercise prevents disturbances in rat cardiac insulin signaling and endothelial nitric oxide synthase induced by high fructose diet

AB  - Increase in fructose consumption together with decrease in physical activity contributes to the development of metabolic syndrome and consequently cardiovascular diseases. The current study examined the preventive role of exercise on defects in cardiac insulin signaling and function of endothelial nitric oxide synthase (eNOS) in fructose fed rats. Male Wistar rats were divided into control, sedentary fructose (received 10% fructose for 9 weeks) and exercise fructose (additionally exposed to low intensity exercise) groups. Concentration of triglycerides, glucose, insulin and visceral adipose tissue weight were determined to estimate metabolic syndrome development. Expression and/or phosphorylation of cardiac insulin receptor (IR), insulin receptor substrate 1 (IRS1), tyrosine-specific protein phosphatase 1B (PTP1B), Akt, extracellular signal-regulated protein kinases 1 and 2 (ERK1/2) and eNOS were evaluated. Fructose overload increased visceral adipose tissue, insulin concentration and homeostasis model assessment index. Exercise managed to decrease visceral adiposity and insulin level and to increase insulin sensitivity. Fructose diet increased level of cardiac PTP1B and pIRS1 (Ser307), while levels of IR and ERK1/2, as well as pIRS1 (Tyr 632), pAkt (Ser473, Thr308) and pERK1/2 were decreased. These disturbances were accompanied by reduced phosphorylation of eNOS at Ser1177. Exercise managed to prevent most of the disturbances in insulin signaling caused by fructose diet (except phosphorylation of IRS1 at Tyr 632 and phosphorylation and protein expression of ERK1/2) and consequently restored function of eNOS. Low intensity exercise could be considered as efficient treatment of cardiac insulin resistance induced by fructose diet. (C) 2015 Elsevier Ireland Ltd. All rights reserved.
