AB  - Cilj istraživanja bio je ispitivanje antimikrobnog potencijala monoterpena karvakrola i geraniola na bakterijskim izolatima Streptococcus spp. od značaja za etiologiju karijesa (S. mutans, S. sanguinis, S. parasanguinis, S. gordonii). Antimikrobni potencijal karvakrola i geraniola testiran je mikrodilucionim testom, kojim su određene minimalna inhibitorna koncentracija (MIC) i minimalna baktericidna koncentracija (MBC). Testirani monoterpeni ispoljili su značajan antimikrobni potencijal, MIC je bio u opsegu od 0.55 mg/mL do 4.39 mg/mL, a MBC od 2.20 mg/mL do 19.54 mg/mL.
AB  - The aim of our study was to investigate the antimicrobial potential of monoterpenes carvacrol and geraniol towards the Streptococcus spp. isolates relevant to tooth caries (S. mutans, S. sanguinis, S. parasanguinis, S gordonii). Microdilution test was used for the assessment of the antimicrobial potential of carvacrol and geraniol, and the minimal inhibitory concentrations (MICs) and minimal bactericidal concentrations (MBCs) were determined. Tested monoterpenes achieved notable antimicrobial activity, with MICs ranging from 0.55 mg/mL to 4.39 mg/mL, and MBCs from 2.20 mg/mL to 19.54 mg/mL.

EO-based mouthwashes: Is there something that should be known?

AB  - The possibility to utilize the antibacterial and antibiofilm potential of essential oils (EOs) in dentistry is recognized and best seen in the design of EO-based mouthwashes. The up-to-date results, obtained with the most commonly used formulation being consisted of thymol, eucalyptol, menthol, and methyl salicylate (Listerine), proved its efficacy against plaque and gingival inflammation. However, novel alcohol-free products remain to be further investigated. Commercially available mouthwash containing curcumin, clove oil, mentha oil, eucalyptol, thymol and tea tree oil, and the mouthwash based on ginger EO (a-zingiberene, b-bisabolene, b-sesquiphellandrene, curcumene), reduced plaque index and improved periodontal status of the patients. The plaque index was also reduced and overall healing was improved after the surgical removal of the third molar by mouthwash containing John wort oil. In addition, Cinnamomum zeylanicum EO-based mouthwash and mouthwash containing peppermint oil were efficient against stomatitis and xerostomia, respectively. In addition to beneficial effects achieved in the mouth, i.e. at the appropriate site of application, novel investigations pointed-out that EO-based mouthwashes could be recognized as efficient in the reduction of bacterial and viral aerosols. This has been shown for Listerine, and for several mouthwashes containing following active substances of plants' EO origin: a-farnesene, b-farnesene, farnesol, nerolidol, a-bulnesene, eremanthin, b-sesquiphellandrene, spiroether, cinnamylacetate, cinnamaldehyde, eugenol, menthol, and carvacrol. This finding could be of special interest, in order to be included in further clinical studies related to COVID -19 pandemic.

Antibacterial efficiency of adjuvant photodynamic therapy and high-power diode laser in the treatment of young permanent teeth with chronic periapical periodontitis. A prospective clinical study

AB  - Background  The study aimed at determining antibacterial efficiency of adjuvant photodynamic therapy (PDT) and high-power diode laser (DL) in the treatment of chronic periapical periodontitis (CPP) in young permanent teeth. Methods  Forty-four young permanent teeth with CPP were randomly divided into three groups (PDT, DL and control). Each tooth underwent standard chemo-mechanical treatment, while within tested groups was additionally treated by PDT or DL. Bacterial identification and quantification were provided by MALDI–TOF spectrometry and plate counting assay, performed after accessing the canal, following chemo-mechanical preparation, and after PDT or DL procedure where applicable. Results  Thirty-nine young permanent teeth with CCP (patients age 9.77 ± 1.43) completed the study. Before the treatments, 202 isolates belonging to 13 genera/species, including Streptococcus (36), Actinomyces (34), Peptostreptococcus micros (27), Veillonella (25) and Enterococcus faecalis (22), were recovered. Chemo-mechanical treatment reduced CFU count in the all three groups (p < 0.001), but complete eradication was not observed for any of isolated species. Adjuvant PDT and DL completely eradicated isolates of 8 and 6 bacterial genera/species, resulting in complete bacterial elimination from 53.8% and 30.8% of root canals, respectively. In the rest canals, total Δlog CFUs were 4.71 and 4.58. Conclusion  The results indicated that both PDT and DL could be performed as adjuvants to standard endodontic treatment of the young permanent teeth with CPP.

Biocompatibility and Antibacterial Potential of the Cinnamomum camphora cineoliferum (L.) J. Presl. and Melaleuca ericifolia Sm. Essential Oils Against Facultative and Obligate Endodontic Anaerobes

AB  - The aim of the study was to investigate the potency of Cinnamomum camphora cineoliferum and Melaleuca ericifolia essential oils (EOs) against facultative and obligate anaerobes isolated from teeth’ infected root canals. Biocompatibility of the EOs was also in the study focus. Chemical composition of the EOs was determined by GC/MS analysis. Their antibacterial potential and cytotoxicity were evaluated by microdilution and MTT assays, respectively. Statistical analysis was performed by Student’s t-test and one way ANOVA. Two the most abundant constituents of C. camphora cineoliferum EO were 1,8-cineole and sabinene, while in M. ericifolia EO the dominant were linalool and 1,8-cineole. MICs achieved against all tested strains were in the ranges 0.95±0.50 - 22.50±0.00 mg/mL and 0.65±0.00 - 23.35±7.80 mg/mL for the C. camphora cioneoliferum and M. ericifolia EOs, respectively. Statistical analysis showed that response to both oils has not differed between facultative and obligate anaerobes. Sensitivity pattern of the tested strains was different for the EOs and the positive control triple antibiotic paste (TAP): while E. faecalis and certain strains of S. salivarius were among the most sensitive to the oils, they were the least susceptible to the conventional antimicrobial TAP. Although the observed cytotoxicity of the EOs after 30 min of cell exposition was high (50.78-66.35% and up to 70% for C. camphora cioneoliferum and M. ericifolia EOs, respectively) it was lower than that of positive control (1.5% NaOCl, p<0.05). To conclude, both EOs showed promising antibacterial potential and achieved at least the same or even better biocompatibility than the conventional intracanal antimicrobials. Obtained results strongly encourage future research with an aim to formulate C. camphora cioneoliferum and M. ericifolia EO- based irrigants and medicaments, that could be used in endodontic practice.

AB  - Cymbopogon martinii is an aromatic crop cultivated for the production of its essential oil (EO) which has different applications. Bearing in mind that C. martinii EO (CMEO) has already been proved for high antibacterial potential the aim of this study was to formulate stable CMEO nanoemulsions, being capable to combat Enterococcus faecalis biofilm, especially within infected tooth’s root canal. Ten emulsions with variable CMEO content (2.5–10 % v/v) and Tween 80 (T80, 2.5–15 % v/v) were formulated (designated as ECMEO:T80) and screened for volume-weighted mean diameter (D4,3) and stability, using dynamic light scattering measurements. Chemical composition of stable nanoemulsions was monitored by UV–VIS and FTIR, while their antibacterial activity was estimated in microdilution assay. Antibiofilm properties of selected nanoemulsions were tested in vitro (cristal violet assay) and ex vivo (within root canals of the extracted teeth). AB  - To enhance endodontics, the search for new antibacterials that can improve infected tooth root canal treatment is ongoing. As potent antibacterial and antibiofilm agents, essential oils (EOs) have been suggested as novel endodontic materials. Several studies indicate that EO-based irrigants and medicaments show promising reductive potential against the most important intracanal pathogen, Enterococcus faecalis, and notably contribute to intracanal biofilm eradication. In terms of additional benefits that EO-based endodontic materials can provide, their antioxidant and anti-inflammatory potential are also important, but they have only scarcely been explored in research. Investigations into the benefits of EO-based endodontic materials together with their biocompatibility are needed. The results presented in this review strongly encourage further research on this topic.

AB  - Aim: The objective was to formulate and characterize the nanoemulsion based on Cymbopogon citratus oil, intended for use in infected teeth root canal therapy. Antibiofilm efficacy of irrigation procedure including nanoemulsion was screened on extracted teeth (by CFU-counting assay). Results: Notable antibacterial and antibiofilm activity, both against forming and preformed biofilms of oil, was observed. Irrigation involved nanoemulsion showed remarkable antibiofilm potential. Both substances induced some antioxidant activity. Conclusion: Results encourage further research with the aim of application of the nanoemulsion in dental practice.

To examine the variation in sensitivity among strains of different origin, isolates from symptomatic and asymptomatic root canal infections, as well as the reference, strains S. sanguinis ATCC 10556 and S. salivarius ATCC 9222 were used in the study. EOs were chemically characterised (GC/MS). Their antibacterial and antibiofilm properties were detected by microdilution and CV assay respectively. Statistical analysis was performed by One way ANOVA. Examined bacterial strains, regardless clinical or ATCC, exhibited the highest sensitivity to L. petersonii and E. citriodora EOs (MIC ranged 0.53±0.00 - 5.88±1.27 mg/ml and 0.08±0.03 - 9.37±1.25 mg/ml respectively), while two other EOs were less efficient. Comparison of MIC values showed that sensitivity of the isolates recovered from symptomatic infections was the least, while the susceptibility of asymptomatic isolates and especially of reference strains were higher. EO inhibition of biofilm formation differed between clinical isolates of S. salivarius and their reference strain. Inhibition was higher in the case of S. salivarius ATCC 9222 (84 %-89 %) in comparison to that of clinical isolates (24 %-75 %). Regardless of the strains tested, E. citriodora exhibited the highest antibiofilm potential. To conclude: L. petersonii induced notable antibacterial potential, while E. citriodora EO proved promising antibacterial and antibiofilm activity. Due to the variable sensitivity of the isolates to EOs, it is suggested to screen activity on a large number of strains, especially from symptomatic infections.

Its uncontrolled overexploitation led to the protection regime and encouraged development of in vitro culture. The aim of this work was to chemically characterize root and leaf methanol extracts of plant grown in plantation and in vitro conditions, and to determine their antigenotoxicity against heterocyclic aromatic amines IQ and PhIP, the food-borne mutagens that are formed during high temperature cooking of protein-reach foods. High yields and growth ratios were determined for in vitro plants. Higher content of polyphenols and flavonoids were determined in aerial parts, while UPLC-MS/MS analysis pointed at the richness of in vitro grown shoot extract (GLvS) in active constituents, and high amounts of gentiopicroside in all tested extracts. Notable amounts of sweroside were detected in plantation root extract (GLR) and in GLvS, and of loganic acid in plantation leaf extract (GLL) and GLvS. Two experimental models for genotoxicity/antigenotoxicity study, i.e. SOS/umuC test with metabolic activation (addition of rat liver S9 fraction) and alkaline comet assay on hepatocellur carcinoma HepG2 cells indicated dual effects: genotoxic of high and antigenotoxic of lower non-genotoxic doses. While in vitro extracts were genotoxic in both models, plantation extracts were active only in SOS/umuC with S9. In contrast, inhibition of genotoxicity was observed in both models for all extracts (up to 83 %). Results of DPPH assay indicated that antioxidativity could be responsible for antigenotoxic properties. In conclusion, genoprotective effect encourages further investigation of antigenotoxicity, while high growth ratio and richness of GLvS in active compounds justify the use of in vitro cultivation method.

AB  - OBJECTIVE The objective was to determine the antibiofilm efficacy of Cymbopogon martinii and Thymus zygis essential oils and to estimate the disruption potential of oil-based endodontic irrigants on the multispecies biofilm formed in the root canals of extracted teeth. DESIGN The essential oils were characterized (GC-MS), while their antibacterial and cytotoxic properties were detected by microdilution and MTT assays. Particles sizes and polydispersity indices were determined for the irrigants. The isolates from root canals of pediatric patients were identified (MALDI-TOF). The multispecies biofilms were formed from Streptococcus mitis, Streptococcus sanguinis and Enterococcus faecalis. Antibiofilm properties of the essential oils and irrigants were determined by crystal violet and plate counting assay. Statistical analysis was performed by one-way ANOVA. RESULTS The screening of biofilm biomass revealed a disruption potential of C. martinii essential oil. The plate counting assay showed the efficacy of both oils in diminishing cell viability: high in biofilms (reduction of log10CFU was 2.75-2.87) and moderate in the planktons formed above. The essential oil-based irrigants showed the same antibiofilm activity as in the control (1.5 % sodium hypochlorite) for C. martini and almost 2-fold higher for T. zygis. Successive irrigations with 1.5 % sodium hypochlorite, saline and an oil-based irrigant was more efficient for C. martini than for the control (reductions of log10CFU was 1.69 and 1.14, respectively). CONCLUSIONS Notable activities achieved by essential oils and oil-based irrigants, particularly regarding C. martini, against the biofilm consisting of S. mitis, S. sanguinis, and E. faecalis, are expected to encourage further research in endodontics.

The aim of the study was to determine differences in cardiorespiratory parameters of male cyclists according to their specialty: flat riders (N.=21), hill riders (N.=35), or sprinters (N.=20) and obtain the multivariate model for further cyclists classification by specialties, based on selected variables. METHODS: Seventeen variables were measured at submaximal and maximum load on the cycle ergometer Cosmed E 400HK (Cosmed, Rome, Italy) (initial 100 W with 25-W increase, 90-100 rpm). Multivariate discriminant analysis was used to determine which variables group cyclists within their specialty, and to predict which variables can direct cyclists to a particular specialty. RESULTS: Among nine variables that statistically contribute to the discriminant power of the model, achieved power on the anaerobic threshold and the produced CO 2 had the biggest impact. The obtained discriminatory model correctly classified 91.43% of flat riders, 85.71% of hill riders, while sprinters were classified completely correct (100%), i.e. 92.10% of examinees were correctly classified, which point out the strength of the discriminatory model. CONCLUSIONS: Respiratory indicators mostly contribute to the discriminant power of the model, which may significantly contribute to training practice and laboratory tests in future. © 2018 EDIZIONI MINERVA MEDICA.

AB  - During high-temperature cooking of protein rich foods, especially meat and fish, heterocyclic aromatic amines can be formed. These amines are a class of potent mutagens that can cause alterations in the structure of DNA and chromosomes. In recent decades, research has been focused on investigating plants and their phytochemicals as potential antimutagens. The aim of this study was to examine the anti-genotoxic effect of methanolic root and leaf extracts of Gentiana lutea against the food mutagens 2-amino-3-methylimidazo[4,5-f]quinoline (IQ) and 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP) produced in thermally processed meat. To determine the protective potential of extracts, the alkaline comet assay was applied. The results obtained indicated strong anti-genotoxic effect of both extracts against the tested mutagens. The highest inhibition of IQ-induced genotoxicity was recorded for leaf extract (72%). Regarding PhiP, root extract achieved inhibition of 80% of DNA damage, so was more successful than leaf extract. The data obtained in this study stimulates further research of G. lutea extracts and its constituents as potential dietary supplements in improving human health. © Published under licence by IOP Publishing Ltd.

To compare cytotoxicity and genotoxicity of novel urethane-based monomer FIT-852 and monoacylphosphine oxide photoinitiator (Lucirin TPO) with conventional Bisphenol A-glycidyl-methacrylate (BisGMA) and triethylene glycol dimethacrylate (TEGDMA) monomers and camphorquinone (CQ)/amine photoinitiator system, respectively. Moreover, we quantified and analyzed the combinatorial effects of individual substances in resin-based mixtures concerning the nature of the combinatorial effects. Methods. Cytotoxic and genotoxic effects of BisGMA, FIT, TEGDMA, CQ DMAEMA and TPO and their combined toxicity in four clinically relevant mixtures (FIT/TPO, FIT/CQ BisGMA/TPO, BisGMA/CQ) were tested on human fetal lung fibroblasts MRC-5 using MTT and Comet assays. We assessed combination effects of monomers and photoinitiators on overall toxicity from the measured concentration-effect relationships. Combination index (CI) was calculated on the basis of the median-effect equation derived from the mass-action law principle. Results. Individual substances showed decreasing cytotoxic effects in the following order: BisGMA GT TPO GT FIT GT CQ GT DMAEMA GT TEGDMA. Experimental mixtures showed decreasing cytotoxic effects in the order BisGMA/TPO GT BisGMA/CQ GT FIT/CQ GT FIT/TPO. FIT-based mixtures exhibited antagonistic cytotoxic effects between components while BisGMA-based mixtures demonstrated synergistic effects at ED50 TPO amplified both antagonistic and synergistic cytotoxic effects in mixtures. Pure substances showed genotoxicity in the following order: TPO GT BisGMA GT FIT GT CQ GT TEGDMA. We did not detect the genotoxic potential of DMAEMA. The rank of genotoxic concentrations of the mixtures was: BisGMA/TPO GT BisGMA/CQ GT FIT/CQ GT FIT/TPO. Significance. Lower cytotoxicity and genotoxicity of FIT than BisGMA suggests its greater bio-co mpatibility. Conversely, photoinitiator TPO was significantly more cytotoxic and genotoxic than both CQ and DMAEMA. CI values showed that components of FIT-based mixtures exhibit an antagonistic cytotoxic effect, while compontents of BisGMA-based mixtures show synergism.

Cytotoxic and genotoxic effects of BisGMA, FIT, TEGDMA, CQ DMAEMA and TPO and their combined toxicity in four clinically relevant mixtures (FIT/TPO, FIT/CQ BisGMA/TPO, BisGMA/CQ) were tested on human fetal lung fibroblasts MRC-5 using MTT and Comet assays. We assessed combination effects of monomers and photoinitiators on overall toxicity from the measured concentration-effect relationships. Combination index (CI) was calculated on the basis of the median-effect equation derived from the mass-action law principle. Results. Individual substances showed decreasing cytotoxic effects in the following order: BisGMA GT TPO GT FIT GT CQ GT DMAEMA GT TEGDMA. Experimental mixtures showed decreasing cytotoxic effects in the order BisGMA/TPO GT BisGMA/CQ GT FIT/CQ GT FIT/TPO. FIT-based mixtures exhibited antagonistic cytotoxic effects between components while BisGMA-based mixtures demonstrated synergistic effects at ED50 TPO amplified both antagonistic and synergistic cytotoxic effects in mixtures. Pure substances showed genotoxicity in the following order: TPO GT BisGMA GT FIT GT CQ GT TEGDMA. We did not detect the genotoxic potential of DMAEMA. The rank of genotoxic concentrations of the mixtures was: BisGMA/TPO GT BisGMA/CQ GT FIT/CQ GT FIT/TPO. Significance. Lower cytotoxicity and genotoxicity of FIT than BisGMA suggests its greater bio-co mpatibility. Conversely, photoinitiator TPO was significantly more cytotoxic and genotoxic than both CQ and DMAEMA. CI values showed that components of FIT-based mixtures exhibit an antagonistic cytotoxic effect, while compontents of BisGMA-based mixtures show synergism.",

MethodologyCytotoxicity of eluates of new nanostructural noncommercial materials CS and HA-CS, and MTA (White MTA, Angelus((R)) Solucoes Odontologicas, Londrina, Brazil) as a control, were tested using the MTT assay on MRC-5 cells. Eluates of set materials were tested in 100% and 50% concentrations, 24h, 7days and 21days post-elution. The pH values were determined for undiluted eluates of set materials. Polyethylene tubes containing the test materials (CS, HA-CS, MTA) were implanted in subcutaneous tissue of Wistar rats. Histopathological examinations were conducted at 7, 15, 30 and 60days after the implantation. Data were statistically analyzed using three-way and one-way anova Tukeys post hoc test as well as Kruskall-Wallis test with Dunns post hoc test at =0.05. ResultsAll materials significantly reduced cell viability; especially when undiluted eluates were used (P LT 0.001). After 24h elution, cell viability was 101.8%, 49.5 +/- 4.2% and 61 +/- 7.4%, for MTA, and HA-CS, respectively. However, CS and HA-CS were significantly less toxic than the control material MTA (P LT 0.05). Cytotoxicity could be at least partially attributed to pH kinetics over time. Dilution of eluates of all tested materials resulted in better cell survival. Histopathological examination indicated similar inflammatory reaction, vascular congestion and connective tissue integrity associated with CS, HA-CS and MTA at each observation period (P GT 0.05). The only significant difference was found for capsule thickness, that is thicker capsule was associated with HA-CS compared to MTA at 60days (P=0.0039). HA-CS induced moderately thick capsules (median score 3, score range 2-3), whereas MTA resulted in thin capsule formation (median score 2, score range 1-3). Methodology Hydrothermal treatment was applied for synthesis of CS and HA. The final mixture HA-CS, with potential to be used in endodontic practice, is composed of CS (34%) and HA (66%). Human lymphocytes were incubated with HA, HA-CS and CS for 1h, at 37 degrees C and 5% CO2. Cell viability was determined using the trypan blue exclusion assay. To evaluate the level of DNA damage comet assay (single cell gel electrophoresis) was performed. For the statistical analysis anova and Duncans Post Hoc Test were used. Results The SEM analysis indicated that CS consisted mostly of agglomerates of several micrometers in size, built up from smaller particles, with dimensions between 117 and 477nm. This is promising because dimensions of agglomerates are not comparable with channels inside the cell membranes, whereas their nano-elements provide evident activity, important for faster setting of these mixtures compared to MTA. Values of DNA damage obtained in the comet assay indicated low genotoxic risk of the new endodontic materials. Conclusions The significantly improved setting characteristics and low genotoxic risk of the new material support further research.
