AB  - Herein, a screen–printed diamond electrode (SPDE) coupled with a “point-of-care” platform (30 µL-drop concepts, single-drop-detection approach) was successfully applied for the electrochemical determination of pterostilbene (PTS). Cyclic voltammetry identified irreversible oxidation of PTS, where oxidation peak was shown to be strongly dependent on the pH of the working environmental. Although the proposition of the detailed electrochemical oxidation mechanism of PTS goes out of the scope of the present research, we have determined the most probable reactive site of our analyte, by utilizing DFT-based reactivity descriptors (Fukui functions). For electrochemical quantification of PTS, oxidation peak at 0.32 V (vs. Ag/AgCl) was followed in presence of 0.5 mol L−1 of Briton–Robinson buffer solution (pH = 9). Coupled with the optimized parameters of differential pulse voltammetry (DPV), SPDE detected PTS in two linear ranges (first range was from 0.011 to 0.912 µmol L−1; second range was from 0.912 to 4.420 µmol L−1), providing the LOD and LOQ on a nanomolar level (3.1 nmol L−1 and 10.0 nmol L−1, respectively). The selectivity of the optimized DPV method was found to be excellent, with the current changes of less than 7%, in the presence of ten times higher concentrations of the certain interferences. The practical applicability of the SPDE and single-drop-detection approach in dietary supplements (with a declared PTS content of 50 mg/tablet), with the recovery values ranging from 95 to 102%, shows that the developed method has high potential for precise and accurate PTS detection, as well as exceptional miniaturization possibilities of relevant equipment for on-site sensing.

Advanced electrochemical platform for simple and rapid quantification of tannic acid in beverages using batch injection analysis with amperometric detection

AB  - In this work, an advanced electrochemical platform was developed for the simple, rapid and sensitive determination of the polyphenolic compound tannic acid in various beverages using the combination of batch injection analysis with amperometric detection on a screen-printed carbon electrode. Several experimental parameters (pH of supporting electrolyte, detection potential, dispensing rate, injected volume, stirring) were consistently evaluated. The most favorable analytical performance in terms of sensitivity, selectivity, repeatability and sampling frequency was obtained in Britton-Robinson buffer pH 5.0 at a detection potential of +0.6 V vs. Ag/AgCl, a dispensing rate of 204 µL/s and an injected volume of 80 µL under stirring condition (1500 r.p.m.). The presented platform has advantages for routine analysis including portable and small-scale experimental setup, low sample consumption (∼100 µL), simple sample preparation (dilution in supporting electrolyte), high sampling frequency (180 injections per hour), low limit of detection (80 nM) and suitable precision (RSD = 4.2% for 10 μM tannic acid, n = 20). The applicability of the method was verified by analyzing several beverage samples (tea, wine) in spike-recovery assay with the recovery values for tannic acid ranging from 94 to 101 %. © 2023 Elsevier B.V.

AB  - This work presents advanced electrochemical platform based on screen printed diamond electrode (SPDE) system for the single drop “point-of-care” testing. Proposed approach was applied for the quantification of doxorubicin hydrochloride (DOX) in biological fluids and pharmaceutical product. Using a single drop (~30 μL) of the tested solution at the electrode surface, DOX showed high electroactivity over a wide range of pHs. In these conditions, single oval shaped, well-defined and pH dependent oxidation peak was observed in the potential range from 0.5 V to 1.3 V. In the reverse scan, two cathodic peaks, were noted – around 0.3 V and – 0.5 V. Similarly, first reduction peak was pH dependent, while second one was independent in the studied range. Experimental conditions for DOX quantification were optimized and natures of the electrode reactions were investigated. Working linear range obtained for DOX detection was from 0.1 to 2.5 μM. Diffusion controlled electrode reaction reveal long life time of the proposed electrode as well high potential for practical application. Developed procedure was successfully applied for the DOX analysis in biological fluids – urine and pharmaceutical formulation. Obtained results clearly indicated that given procedure can be easily implemented for pharmaceutical control and medical analysis, in both, laboratory and field conditions. © 2021 Elsevier B.V.

A new voltammetric platform for reliable determination of the sport performance-enhancing stimulant synephrine in dietary supplements using a boron-doped diamond electrode

AB  - For the first time, we propose a new modification-free voltammetric platform for simple, fast and reliable quantification of the sport performance-enhancing stimulant synephrine (SYN) based on the usage of a commercial screen-printed boron-doped diamond (BDD) electrochemical sensor. Applying cyclic voltammetry, SYN exhibited one well-shaped, irreversible and diffusion-driven oxidation peak at a peak potential of +1.45 V (vs. the silver pseudoreference electrode) in the presence of 2 M HClO4. Under the selected experimental conditions, the following analytical parameters for determination of SYN were obtained: linear concentration ranges from 19.6 up to 1000 μM and from 9.9 up to 1000 μM for differential pulse (DPV) and square-wave voltammetry (SWV), respectively, detection limits at micromolar concentration levels and intra-day repeatability with a relative standard deviation below 4% for both pulse techniques. The interference study revealed fair selectivity when considering the target dietary supplements. The feasibility of the developed voltammetric platform was verified in the analysis of commercially available dietary supplements and reasonable recovery values were achieved by DPV and SWV, respectively. In addition, the method can be performed directly without higher consumption of chemicals, waste generation, complex sample extraction and higher instrumentation cost. The advanced BDD electrochemical sensor has appeared to be a suitable competitor for efficient applications in food quality control analysis.

AB  - In present work boron doped diamond (BDD) electrode was applied for studying electrochemical behavior, as well as for quantitative determination of natural alkaloid, tubocurarine (arrow poison/ TUB). Electrochemical behavior was investigated using several organic solvents and aqueous buffers, at different pHs, as supporting electrolytes. After selection of the most appropriate supporting electrolyte and investigation of its electrochemical behavior, analytical procedures for quantitative analysis of TUB were developed in an acetonitrile/methanol (80:20) mixture and in 1 mol L−1 nitric acid, which had linear quantification ranges from 4 × 10−6 to 1 × 10−4 mol L−1 (LOD = 3.07 × 10−6 mol L−1) and 4 × 10−6 to 9 × 10−5 mol L−1 (LOD = 2.45 × 10−6 mol L−1 and 5.28 × 10−7 mol L−1, depending on the selected peak), respectively. The oxidation mechanism of TUB was proposed from the theoretical aspect. Remarkable selectivity and good sensitivity were obtained after the TUB quantification method was optimized, allowing application of the developed method for TUB quantification in biological fluids, with excellent reproducibility, accuracy and precision.

AB  - In present work we report application of bismuth (III) oxide (Bi2O3) decorated graphene nanoribbons (GNR) composite for modification of screen printed electrode, and use of resulting modified screen-printed electrode (SPCE/GNR/Bi2O3) as a disposable glucose biosensor. The composite was synthesized by use of co-precipitation method followed by ultra-sonication. After characterization, the material was applied on the surface of the screen-printed electrode in a ‘drop casting’ fashion. Glucose oxidase (GOx) was added to the modified surface and finally covered by Nafion film. Resulting biosensor electrode (SPCE/GNR/Bi2O3/GOx/Naf) shows excellent performance toward glucose detection, with working linear range from 0.28 to 1.70 mM and detection limit of 0.07 mM. Prepared biosensor was applied for the estimation of glucose level in standard honey samples with declared glucose level, in order to validate the methods (PT scheme). Obtained results are indicating excellent stability, reproducibility, accuracy and precision of the method for the determination of glucose in such samples with negligible matrix effect. Developed biosensor showed high precision, accuracy and selectivity even when employed for determination of glucose content in blood serum and urine samples. Proposed biosensor construction shows promising results and can be considered as stabile and reproducible tool for detection and quantification of glucose in investigated samples. © 2019 Elsevier B.V.

AB  - Ergosterol (Erg) is the major sterol component in the plasma membranes of fungi. The main goal of this work was to develop rapid, reliable, and cost-effective method toward Erg detection based on application of unmodified boron-doped diamond electrode. Several organic solvents were tested (acetonitrile, dimethyformamide, dimethylsulfoxide, dichloromethane), and it was found that best analytical characteristics were achieved using acetonitrile containing 0.1 M tetrabutylammonium hexa-fluorophosphate as supporting electrolyte. Oval shaped and well-defined oxidation peak was recorded at potential of around 1.17 V versus Ag/Ag+ non-aqueous reference electrode. Under optimized experimental conditions, linear working range from 2 to 200 mu M was estimated, with limit of detection of 0.7 mu M, using square wave voltammetry. Negligible effect of possible interfering compound was observed. Proposed methodology was successfully applied for estimation of Erg content in mushrooms extract with satisfactory results in comparison with spectrophotometric procedure. Obtained results clearly show that developed analytical methodology can be adequate replacement for the, up to date, used methods and open-novel approach for ergosterol detection.

AB  - In the present paper, a miniaturized boron-doped diamond electrode (BDDE) is proposed as a progressive electrochemical sensor for simple, fast and reliable quantification of dimethylxanthine alkaloid, theobromine (TB). Using cyclic voltammetry, the studied xanthine provided one well-shaped, irreversible and diffusion-controlled oxidation peak at relatively high potentials (+1.2 V vs. Ag/AgCl/3 M KCl reference electrode) in the presence of 0.1 M H2SO4. After selection of suitable experimental conditions, the linear calibration curves for TB were obtained in the concentration range from 0.99 up to 54.5 μM with the sensitivity of 0.07 μA/μM providing both differential pulse (DPV) and square-wave voltammetric (SWV) techniques, respectively. The elaborated voltammetric protocol yielded low detection limits of 0.42 and 0.51 μM accompanied by adequate intra-day repeatability (relative standard deviation of 2.5 and 1.7%) using DPV and SWV procedure, respectively. The interference study revealed the reasonable selectivity when taking the target food samples into account. The practical applicability of the voltammetric protocol using a miniaturized BDDE was verified in the analysis of six commercially available brands of chocolate products with the determined mass percentages of TB ranging from 0.75 to 2.24% and from 0.69 to 2.15% using DPV and SWV procedure, respectively. The obtained results were also in a good agreement with those achieved by reference titration method with potentiometric indication. The progressive electrochemical sensor based on a miniaturized BDDE has appeared to be an attractive candidate for practical applications in food quality control. Besides, the proposed voltammetric protocol presents advantages when compared to others techniques (e.g. chromatography), concerning simplicity, cost, speed of analysis, waste generation (environmentally friendly) and samples pretreatment (only dilution in electrolyte solution prior to analysis). © 2018 Elsevier B.V.

The elaborated electroanalytical protocol rendered low detection limits of 4.1 × 10−7and 9.3 × 10−7mol L−1in association with favourable intra-day repeatability (relative standard deviation of 3.6 and 4.6%) using DPV and SWV procedures, respectively. The effect of interfering compounds such as ascorbic acid, dopamine, caffeine, uric acid and glucose on the current response of ibuprofen was explored in details. The usefulness of the proposed approach was verified in the analysis of a variety of commercial brands of pharmaceuticals and spiked human urine samples with the significant range of recovery percentages (for pharmaceuticals: 99.8–107.5% and 99.8–105.0% by DPV and SWV, for urine: 95–107% and 97–103% by DPV and SWV). Taking these features into account, the developed protocol may be exploited as a novel, simple and efficient tool in drug quality control analysis and analysis of biological samples. In addition, a bare and electrochemically untreated boron-doped diamond electrode may be applied as a progressive electrochemical sensor and helpful alternative to previously utilized electrochemical platforms in this field.

AB  - In this work, the electrochemical oxidation and subsequent determination of uric acid was investigated using boron-doped diamond electrodes with various B/C ratios (0-2000 ppm). The cyclic voltammetric study showed one irreversible oxidation peak at +(1.1-1.25) V (vs. Ag/AgCl/3 M KCl) in the presence of Britton-Robinson buffer (pH 2.25) depending on the boron content. Employing differential pulse voltammetry using the 2000 ppm boron-doped diamond electrode the acquired analytical parameters were as follows: a limit of detection of 7.7 M, a limit of quantification of 26 M and intra-day repeatability (relative standard deviation of 2.9% for n = 15). After performing an interference study, the method was applied to the determination of uric acid in biological samples (human urine). The uric acid concentrations determined in the urine samples were compared with the reference values stated in the literature. The proposed methodology using boron-doped diamond electrodes could find applications in uric acid sensing within clinical, pharmaceutical and environmental analysis.

AB  - The boron-doped diamond electrodes (BDDEs) with different dopant levels were applied as advanced, mercury-free and modification-free electroanalytical platforms for simple and reliable quantification of benzodiazepines, bromazepam (BZ) and alprazolam (ALZ). AB  - In the present paper, the electrochemical behavior and the properties of two modified glassy carbon (GC) electrodes used for quantification of gallic acid in sweet wines were compared. A comparative study was conducted between titanium nitride- or wolfram carbide-doped reduced graphene oxide, labeled as TNrGO and WCrGO, respectively, modified GC electrodes, which are promising composite nanomaterials for electroanalytical applications. For the first time, WCrGO was synthesized and its electroanalytical properties compared with those of TNrGO. Results showed that the proposed materials exhibited enhanced characteristics, e.g., low limits of detection (1.1 mu M and 3.1 mu M for TNrGO and WCrGO, respectively), wide linear ranges (for TNrGO 4.5-76 mu M and for WCrGO 10-100 mu M), low adsorption, and low background current, which make them promising candidates for electrochemical sensing applications.

AB  - An innovative, rapid and simple electrochemical approach for the reliable quantification of cytostatic drug flutamide (FLU) in various matrices is herein proposed. This platform involves coupling of differential pulse (DPV) and square-wave voltammetry (SWV) with a boron-doped diamond (BDD) electrode as the working electrode and 0.1 M sulphuric acid as the supporting electrolyte. For the first time, the voltammetric profile of FLU was manifested by three irreversible and diffusion-controlled oxidation peaks at + 1.1 (P1), + 1.4 (P2) and + 1.9 V (P3). The analytical performance evaluation was assessed for all three peaks, using both pulse voltammetric techniques with the optimized operating parameters and the highest sensitivity of 1.76 nA/mu M was accomplished for P2 using DPV and 3.54 nA/mu M for P3 using SWV. The corresponding linear concentration ranges were found to be 0.99-42.9 and 4.8-35.5 mu M with the detection limits of 0.42 and 0.18 mu M, respectively. The repeatability varied, depending on the oxidation peaks of FLU, with the relative standard deviations in the range of 3.3-8.8% and 2.9-9.3% for DPV and SWV, respectively. The proposed electrochemical platform was successfully applied in the analysis of pharmaceutical formulations, spiked human urine and water samples with the significant mean recoveries. Using BDD electrode, the current work establishes an advanced, simple and rapid alternative platform to so far used toxic mercury-based electrodes and time demanding chemically modified electrodes in cytostatic sensing. AB  - Colchicine (COLC) is a natural toxic product and secondary metabolite most commonly used to treat gout. In this study, its electrochemical behavior and determination was investigated by employing modification-free boron-doped diamond electrode (BDDE). Besides, its interaction with DNA was monitored using electrochemical methods. It was found that oxidation of this compound proceeds in two steps, where first sharp and well defined oxidation peak occurs at potential of around 1.19V, and second one at around 1.37V, in Britton-Robinson buffer solution at pH7.5. Wide dynamic range from 1 to 100 mu M was obtained with a detection limit (3 sigma(intercept)/slope) of a 0.26 mu M, based on the evaluation of first oxidation peak using differential pulse voltammetry. The proposed method was also found to be suitable for monitoring interaction of this drug with DNA as important segment for medical use. Concerning the validation, the analytical procedure shows excellent selectivity and sensitivity toward COLC detection and after method development it was successfully used for its quantification in pharmaceutical preparation and human serum sample, with satisfactory recovery. Obviously, this approach can be promising replacement for time-consuming and expensive separation methods.

AB  - The boron-doped diamond electrodes (BDDEs) with different dopant levels were applied as advanced, mercury-free and modification-free electroanalytical platforms for simple and reliable quantification of benzodiazepines, bromazepam (BZ) and alprazolam (ALZ). Cyclic voltammetric measurements revealed that the electrode reactions of both analytes were manifested by one irreversible and diffusion-controlled reduction peak at -1.10V in Britton-Robinson buffer of pH 11 for BZ using lab-made BDDE with 1000 ppm B/C ratio and -0.84V in BR buffer of pH 5 for ALZ using commercial BDDE with 1000 ppm B/C ratio. Differential pulse voltammetry was used for construction of calibration curves for BZ and ALZ, respectively, with the analytical parameters as follows: the linear dynamic ranges of 1 x 10(-6)-1 x 10(-4) and 8 x 10(-7)-1 x 10(-4) mol/L, the detection limit of 3.1 x 10(-7) and 6.4 x 10(-7) mol/L and the excellent repeatability with the relative standard deviation below 3% for both drugs. The developed methods were applied to analysis of the pharmaceutical tablets with the recoveries from 97.33 to 100.85% for BZ and from 94.14 to 101.32% for ALZ. The usage of BDDEs as advanced electrochemical sensors in drug analysis represents a facile and effective analytical approach which may replace mercury-based sensors and chemically modified electrodes in previous benzodiazepines sensing.
