Acid-base equilibria of 2,4-diiodo-6-methylphenylearbamoylmethyl iminodiacetic acid and its labeling with technetium-99m

AB  - The acid-base equilibria of a novel hepatobiliary imaging agent, 2,4-diiodo-6-methylphenylcarbamoylmethyl iminodiacetic acid (DIIODIDA) were studied. The potentiometrically determined acidity constants of the second carboxylic group, amino and amide groups were pK(2) = 2.52 +/- 0.02; pK(3) = 5.86 +/- 0.06 and pK(4) = 10.9 +/- 0.1. The determinations were performed at 25 degrees C and an ionic strength of 0.1 mol/dm(3) (NaCl). The acidity constants (pK(1) = 1.3 +/- 0.4) corresponding to the first carboxylic group was determined indirectly, on the basis of equilibrium constants obtained in a heterogeneous system, at 25 degrees C and an ionic strength 1 mol/dm(3) (HCl, NaCl). DIIODIDA was labeled with teclinctiurn-99m, and the influence of pH on the yield of labeling was investigated. It was found that labeling within the pH range from 5.5 to 6.5 provided a radiopharmaceutical of high radiochemical purity ( GT 98%).

AB  - In order to develop a radiopharmaceutical for hepatobiliary scintigraphy with better hepatobiliary properties new ligand for complexation of Tc-99m, 4-iodo-2,6-dimethylphenylcarbamoylmethyl iminodiacetic acid (METHYLIODIDA), was synthesized. Acid-base equilibria of METHYLIODIDA were studied potentiometrically, because these data are important for determination of complex formation conditions. It was established that METHYLIODIDA undergoes a complex acid-base equilibrium due to its zwitterionic nature and four proton-binding sites. The stoichiometric ionization constants were determined at 25 degreesC and constant ionic strength 0.1 M (NaClO4): pK(1) = 1.7 +/- 0.1; pK(2) = 2.44 +/- 0.07; pK(3) = 6.29 +/- 0.02 and pK(4) = 10.91 +/- 0.06, respectively. Electromagnetic localized structures in a relativistic laser-plasma interaction

AB  - Localized electromagnetic structures with trapped laser light inside are often found in simulations of relativistic laser-plasmas. First, we examine existence and stability of one-dimensional electromagnetic solitons formed in a relativistic laser interaction with an underdense cold plasma. In a weakly relativistic model, the original equation of the nonlinear Schrodinger type, with local and non-local cubic nonlinearities is derived. Standing electromagnetic solitons are analytically shown to be stable in agreement with the model simulation. Next, we discuss a novel example of a stimulated backscattering of a laser light from the slow trapped electron-acoustic wave, involving a standing (localized) Stokes light sideband. Conditions are examined when above underdense instability dominates over standard stimulated Raman backscattering.

Relativistic solitons in laser-plasma interaction

AB  - We discuss existence and stability of electromagnetic solitons in a relativistic interaction of a linearly polarized laser light with uniform underdense cold plasma. In a weakly relativistic NLS model with local and non-local cubic nonlinearities, an one-dimensional standing electromagnetic soliton trapped in a localized density well, is analytically shown to be stable. By fully relativistic fluid-Maxwell simulations existence of a family of large amplitude relativistic solitons is revealed. Simple analytical estimates give the relation between the maximum amplitude of the soliton vector potential and its eigen-frequency, in agreement with simulation data.

Chemical and biological properties of verapamil labeled with technetium-99m: A potential myocardial imaging agents

AB  - On the base of property to enter into myocardial cells as a calcium channel blocker, verapamil was labeled with technetium-99m in order to investigate the possibility to obtain new radiopharmaceutical for myocardial imaging. The conditions of labeling verapamil with technetium-99m for different ammounts of stannous(II) ion, mannitol, cystein and pH range 2.5-3.5 were examined. Investigation of radiochemical purity ( GT 95%) and biodistribution of (9)9mTc-verapamil in rats showed that it was stable during 2 hours after labeling. Accumulation of Tc-99m-verapamil in heart was 1.2% and in liver 9.4%, 5 minutes after injection. Biodistribution of Tc-99m-verapamil in rats in conditions of stress, pharmacologically caused by dipiridamol, showed that the elimination of Tc-99m-verapamil from the heart was slower related to the control group. In the group of rats previously treated with isoproterenol uptake of Tc-99m-verapamil in heart was lower related to the control group (0.7% versus 1.0%) 5 minutes after injection. Lipophilicity of (9)9mTc-verapamil was examined by determination of partition coefficient (log P = 0.62) and protein binding (79%). Imaging studies on dogs provided relatively good myocardial images with partially overlap of activity in the lung and liver.

AB  - Pharmaceutical interaction of the most frequently used substances with fats: lanolin, white soft paraffin and vitamin A palmitate was studied. The mixtures were examined by TLC with different ratios of lanolin and vitamin A palmitate, and white soft paraffin and vitamin A palmitate. Vitamin A in the mixtures was determined by the UV spectrophotometric method according to BP 93. It was established that the rise of lanolin concentration resulted in an increased apparent content of Vitamin A in fats as a consequence of lanolin absorption at the wave length of maximum absorption of vitamin A. By an increase of white soft paraffin concentration, an apparent decreased in the content of vitamin A in fats was obtained. Therefore, the suggested UV spectrophotometric method for vitamin A determination has a limited application on fats with high content of lanolin and white soft paraffin.

AB  - The partition coefficient (log P) for n-octanol/water system was calculated applying PACO program for various theoretically possible mono and dihalogenated IDA derivatives. Some of the synthesized ligands (SOLCOIODIDA, IODIDA and DIIODIDA) were labeled with the technetium-99m. The biodistribution and influence of bilirubin on their biokinetics were investigated in rats. The correlation between partition coefficients of ligands increase (log P) and better hepatobiliary properties of Tc-99m-IDA derivatives was determined. The values of log P increase from 1.16 for SOLCOIODIDA, 3.11 for IODIDA to 3.47 for DIIODIDA. In correlation with these results, biliary excretion decreased for 59% for Tc-99m-SOLCOIODIDA and 11% for Tc-99m-IODIDA and Tc-99m-DIIODIDA under hyperbilirubinemia (3.5 min after injection) and 45%, 11% and 0.38% respectively (15 min after injection). The highest biliary excretion had Tc-99m-DIIODIDA (55.4% for 3.5 min). Considering the correlation between hepatobiliary properties and log P, the evaluation of biological properties for various trifluoromethyl mono and dihalogenated IDA derivatives was performed on the basis of the calculated log P in order to synthetize a new radiopharmaceutical for hepatobiliary scintigraphy.

AB  - A nonlinear three-wave interaction in an open dissipative plasma model of a stimulated Raman backscattering is studied. An anomalous kinetic dissipation due to electron trapping and plasma wave breaking is accounted for in a hybrid kinetic-fluid scheme. We simulate a finite plasma with open boundaries and vary a transport parameter to examine a route to spatio-temporal complexity. An interplay between self-organization at micro (kinetic) and macro (fluid) scales is found through quasi-periodic and intermittent evolution of dynamical variables, dissipative structures and related entropy rates. AB  - A new diiodine substituted LDA derivative, 2,4-diiodine-6-methyl IDA (DIIODIDA) was synthesized and labeled with Tc-99m. It was established that Tc-99m-DIIODIDA had high radiochemical purity. Biodistribution and influence of bilirubin on Tc-99m-DIIODIDA biokinetics has been studied in rats and compared to the corresponding results for Tc-99m-SOLCOIODIDA. Related to Tc-99m-SOLCOIODIDA, Tc-99m-DIIODIDA has much better biliary exretion (55.18 versus 43.63%). No change of Tc-99m-DIIODIDA biokinetics, under influence of bilirubin was noticed. Biliary excretion of Tc-99m-SOLCOIODIDA has been reduced for about 60%. The protein binding of Tc-99m-DIIODIDA and Tc-99m-SOLCOIODIDA were also determined, using in vitro method of precipitation. These results showed that Tc-99m-DIIODIDA hepatobiliary imaging agent is superior to the presently used Tc-99m-monoiodine IDA derivatives.

Effect of wave damping on the nonlinear saturation of stimulated Raman scattering in a finite homogeneous plasma

AB  - A question of stationarity of a nonlinear saturated regime of stimulated Raman backscattering in a finite homogeneous plasma is considered. Slowly varying envelope equations with the damping and the nonlinear frequency shift terms included are treated numerically. It is demonstrated by a space-time simulation of these equations that the variation in the damping rate drives the system through various dynamical regimes (from steady-state to chaotic). A simplified, space-only approach, based on the use of a numerical shooting method was applied to calculate the critical damping values corresponding to transitions between stationary and nonstationary saturated regimes. The corresponding bifurcation diagrams that enable prediction of the type of asymptotic behaviour of Raman instability are obtained and discussed.

Spatiotemporal intermittency and chaos in stimulated Raman backscattering

AB  - Spatiotemporal dynamics of stimulated Raman backscattering in a finite, weakly dissipative plasma, with non-linear phase detuning taken into account, is examined numerically. The non-linear model of a three-wave interaction, involving quadratic coupling of slowly varying complex amplitudes of the laser pump, the backscattering and the electron plasma waves, exhibits spatiotemporal intermittency and chaos following a quasi-periodic scenario. Qualitative analysis of spatiotemporal patterns reveals increasing complexity both for the backscattered and the electron plasma wave as the relative pump strength increases. The transition from spatiotemporal intermittency to chaos is identified using methods from the theory of critical phenomena.

AB  - The spatiotemporal evolution of stimulated Raman backscattering in a bounded, uniform, weakly dissipative plasma is studied. The nonlinear model of a three-wave interaction involves a quadratic coupling of slowly varying complex amplitudes of the laser pump, the backscattered and the electron plasma wave. The corresponding set of coupled partial differential equations with nonlinear phase detuning that is taken into account is solved numerically in space time with fixed nonzero source boundary conditions. The study of the above open, convective, weakly confined system reveals a quasiperiodic transition to spatiotemporal chaos via spatiotemporal intermittency. In the analysis of transitions a dual scheme borrowed from fields of nonlinear dynamics and statistical physics is applied. An introduction of a nonlinear three-wave interaction to a growing family of paradigmatic equations which exhibit a route to turbulence via spatiotemporal intermittency is outlined in this work.
