Downregulation of fibrosis related hsa-miR-29c-3p in human CAKUT

AB  - Congenital anomalies of the kidney and urinary tract (CAKUT) represent structural and functional urinary system malformations and take place as one of the most common congenital malformations with an incidence of 1:500. Ureteral obstruction-induced hydronephrosis is associated with renal fibrosis and chronic kidney diseases in the pediatric CAKUT. We aimed to construct interaction network of previously bioinformatically associated miRNAs with CAKUT differentially expressed genes in order to prioritize those associated with fibrotic process and to experimentally validate the expression of selected miRNAs in CAKUT patients compared to control group. We constructed interaction network of hsa-miR-101-3p, hsa-miR-101-5p and hsa-miR-29c-3p that showed significant association with fibrosis. The top enriched molecular pathway was extracellular matrix-receptor interaction (adjusted p = .0000263). We experimentally confirmed expression of three miRNAs (hsa-miR-29c-3p, hsa-miR-101-3p and hsa-miR-101-5p) in obstructed ureters (ureteropelvic junction obstruction and primary obstructive megaureter) and vesicoureteral reflux. The hsa-miR-29c-3p was shown to have lower expression in both patient groups compared to controls. Relative levels of hsa-miR-101-5p and hsa-miR-101-3p showed significant positive correlations in both groups of patients. Statistically significant correlation was observed between hsa-miR-101 (-3p and -5p) and hsa-miR-29c-3p only in the obstructed group. The significant downregulation of anti-fibrotic hsa-miR-29c-3p in obstructive CAKUT could explain activation of genes involved in fibrotic processes. As miRNAs are promising candidates in therapeutic approaches our results need further measurement of fibrotic markers or assessment of extent of fibrosis and functional evaluation of hsa-miR-29c.

AB  - Schiff bases, or azomethine compounds, are commonly employed in the fields of organicsynthesis, metal complexes, materials, and engineering. Especially, they have gainedimportance in medicinal researches, considering their antimicrobial, anticancer, antiinflammatory, and antioxidant properties. On the other side, azo dyes are the most significantgroup of synthetic dyes, utilized in textile fiber dyeing. Conjugation of Schiff bases with azocompounds leads to the class of azo-azomethine dyes, which have numerous applicationsrelated to their coloration and biological properties. Viscose is a textile material widely used inthe medicine. Moreover, viscose fiber can be engineered in many ways which are significant inthe development of medical materials. The antioxidant effect is an important feature of medicaltextiles, such as wound dressings. In this work, the microwave-assisted synthesis andcharacterization of novel azo-azomethine dye are reported. The azo-azomethine dye is obtainedby the condensation between arylazo pyridone dye and 4-aminophenol. The structure ofsynthesized dye was determined by ATR-FTIR, NMR, and UV-Vis spectroscopy. Azoazomethine dye was used for dyeing viscose, and the washing fastness of dyed material wasevaluated according to the standard method. The viscose fabrics, before and after washing, wereanalyzed in terms of their color coordinates in the CIELab color space. The antioxidantproperties of azo-azomethine dye and dyed viscose fabrics were examined by the ABTSmethod.

AB  - Aims: (1) to identify the most dysregulated genes in ureter tissue affected by congenital anomalies of the kidney and urinary tract (CAKUT) and to extract the biological meaning of these markers; (2) to describe the key molecular networks in CAKUT and to provide expression validation of the genes selected from these networks. Main methods: Transcriptome data was obtained from ureter samples of CAKUT patients and controls by Illumina iScan microarray. Identification of differentially expressed genes was coupled with subsequent bioinformatics analyses. Expression of candidate genes was validated by qRT-PCR. Key findings: Analysis of the transcriptome led to the identification of 78 commonly dysregulated genes in CAKUT tissue compared to controls. Integrative bioinformatic analyses of differentially expressed genes identified 7 major networks. The targets for qRT-PCR validation were selected as members of the major molecular networks in CAKUT, which had both, the significant high fold change and biological relevance for CAKUT. By qRT-PCR the substantial increase of LCN2, PROM1, SOSTDC1, and decrease of INA, RASD1 and TAC3 mRNA levels was confirmed. Significance: Since CAKUT is a leading cause of end-stage renal disease in children, the search for molecular targets for postnatal therapy is of particular interest. Data described in this study represents the gene expression profile and significant molecular networks specific to human ureter affected by CAKUT. The discovery of impaired molecular factors and processes is the step towards the uncovering of the key mechanisms that reflect CAKUT postnatally and could lead to the affected tissue deterioration and end organ damage. © 2018 Elsevier Inc.

AB  - In situ synthesis of Cu/Cu2O nanoparticles on the cotton fabric discussed in this study relies on adsorption of Cu2+-ions by carboxylate groups generated through the TEMPO-mediated oxidation of cellulose and their subsequent reduction by sodium borohydride. In order to establish the influence of aldehyde and carboxylate groups on the nanoparticles formation, the duration of TEMPO-mediated oxidation was varied. Chemical changes induced by TEMPO-mediated oxidation were evaluated by titrimetric determination of the amounts of aldehyde and carboxylic groups in cotton and FTIR spectroscopy. The presence of Cu/Cu2O nanoparticles on the cotton fabric was confirmed by FE-SEM, AAS and XRD analyses. Antimicrobial activity of synthesized nanoparticles was tested against Gram-negative bacteria E. coli, Gram-positive bacteria S. aureus and fungi C. albicans. The extension of TEMPO oxidation time led to an increase of carboxylate group content and consequently, formation of larger amounts of Cu/Cu2O nanoparticles. All fabricated textile nanocomposites provided excellent antibacterial and acceptable antifungal activity. They also ensured a controlled release of Cu2+-ions in physiological solution which is an imperative for infection prevention.

AB  - Background: The interindividual variability of cyclosporin A (CsA) pharmacokinetics might be explained by heterogeneity in the cytochrome P450 3A (CYP3A) subfamily. Altered CYP3A enzyme activity was associated with variant allele of P450 oxidoreductase gene (POR*28). The aim of this study was to assess the impact of age, CYP3A5*3, CYP3A4*22, and POR*28 alleles on CsA pharmacokinetics in pediatric renal transplant recipients. Methods: Renal transplant patients receiving CsA (n = 47) were genotyped for CYP3A5*3, CYP3A4*22, and POR*28. Results: CYP3A5 nonexpressers had higher overall dose-adjusted predose concentration (C0/dose; ng/mL per mg/kg) compared with expressers (31.48 +/- 12.75 versus 22.44 +/- 7.12, P = 0.01). CY-P3A5 nonexpressers carrying POR*28 allele had a lower overall dose-adjusted concentration (C2/dose) than those with POR*1/*1 genotype (165.54 +/- 70.40 versus 210.55 +/- 79.98, P = 0.02), with age as covariate. Children aged 6 years and younger had a lower overall C0/dose (18.82 +/- 4.72 versus 34.19 +/- 11.89, P = 0.001) and C2/dose (106.75 +/- 26.99 versus 209.20 +/- 71.57, P LT 0.001) compared with older children. Carriers of CYP3A5*3 allele aged LT = 6 years required higher dose of CsA and achieved lower C0/dose and C2/dose, at most time points, than older carriers of this allele. Carriers of POR*28 allele aged # 6 years required higher doses of CsA, whereas they achieved lower C0/dose and C2/dose, at most time points, in comparison to older carriers of this allele. AB  - Waste hemp (Cannabis sativa) fibers were used as sustainable and renewable raw materials for production of low-cost biocarbon sorbent for heavy metals removal. Carbon precursors of different chemical composition were obtained by oxidative and alkaline treatments of hemp fibers. Influence of lignocellulosic precursor chemical composition on hemp fibers-based biocarbon (HFB) characteristics was examined by BET surface area measurement, scanning electron microscopy and mass titration. It was found that lignin content and polymorphic transformation of cellulose increase the SBET of microporous HFBs, while hemicelluloses induce more homogeneous distribution of adsorption active sites. Heavy metal ions adsorption onto HFBs is primarily influenced by the amount of surface oxygen groups, while specific surface area plays a secondary role. Equilibrium data obtained for lead ions adsorption were analyzed by different nonlinear adsorption isotherms, and the best fitting model was chosen using standard deviation and Akaike information criterion (AIC(C)). The maximum adsorption capacities of HFBs ranged from 103.1 to 116.3 mg Pb/g. Thermodynamic parameters showed that Pb2+ adsorption onto HFBs is a spontaneous and complex endothermic process, suggesting the coexistence of physisorption and chemisorption mechanisms.

Transcriptome-wide based identification of miRs in congenital anomalies of the kidney and urinary tract (CAKUT) in children: the significant upregulation of tissue miR-144 expression

AB  - Background: The genetic cause of most congenital anomalies of the kidney and urinary tract (CAKUT) cases remains unknown, therefore the novel approaches in searching for the common disease denominators are required. miRs regulate gene expression in humans and therefore have potentially therapeutic and biomarker properties. No studies thus far have attempted to explore the miRs in human CAKUT. We applied a new strategy to identify most specific miRs associated with CAKUT, in pediatric patients. Methods: Data from the whole genome expression, gathered from ureter tissue samples of 19 patients and 7 controls, were used for the bioinformatic prediction of miRs activity in CAKUT. We integrated microarray gene expression data and miR target predictions from multiple prediction algorithms using Co-inertia analysis (CIA) in conjunction with correspondence analysis and between group analysis, to produce a ranked list of miRs associated with CAKUT. The CIA included five different sequence based miR target prediction algorithms and the Co-expression Meta-analysis of miR Targets. For the experimental validation of expression of miRs identified by the CIA we used tissue from 36 CAKUT patients and 9 controls. The results of gene ontology (GO) analysis on co-expressed targets of miRs associated with CAKUT were used for the selection of putative biological processes relevant to CAKUT. Results: We identified 7 miRs with a potential role in CAKUT. The top ranked miRs from miRCos communities 4, 1 and 7 were chosen for experimental validation of expression in CAKUT tissue. The 5.7 fold increase of hsa-miR-144 expression in human tissue from CAKUT patients compared to controls (p = 0.005) was observed. From the GO we selected 7 biological processes that could contribute to CAKUT, which genes are potentially influenced by hsa-miR-144. The hsa-miR-200a, hsa-miR-183 and hsa-miR-375 werent differentially expressed in CAKUT. Conclusions: This study shows that integrative approach applied here was useful in identification of the miRs associated with CAKUT. The hsa-miR-144, first time identified in CAKUT, could be connected with biological processes crucial for normal development of kidney and urinary tract. Further functional analysis must follow to reveal the impact of hsa-miR-144 on CAKUT occurrence.

AB  - Carbonized hemp fibers obtained by carbonization of origin and chemically modified waste hemp fibers was used as a low-cost sorbent for heavy metals removal. The efficiency of carbonized hemp fiber samples as a sorbent was tested through the adsorption of lead ions as a heavy metal model ion. Although changes in carbon precursor structure, induced by chemical modification, affect the sorption process and sorption capacity of carbonized hemp fibers, all carbonized hemp fibers samples show good adsorption properties toward lead ions. Obtained experimental data was used for development of the mathematical model that describes both the phenomenon of metal ions transport through the porous sorbent matrices, and the structure of carbonized hemp fibers. Effective diffusion coefficient, damping coefficient and the lead ion concentration profile within the carbonized hemp fibers, obtained as results of proposed mathematical model, give the insight in the mechanism and the rate of adsorption process, while average tortuosity connected the sorbent structure and ions transport through the sorbent. A good agreement between model prediction for both structural and ion transport model parameters and the experimental data, indicates that the proposed mathematical model can be successfully used for optimization of heavy metal ions adsorption process by correlating the model parameters to the carbonized hemp fibers performances. AB  - Congenital anomalies of the kidney and urinary tract (CAKUT) are a common cause of progressive chronic kidney disease that may lead to end-stage renal disease and renal replacement therapy in childhood. Altered expression or activity of matrix metalloproteinases (MMPs) have been found in CAKUT. The MMP-1, -3, and -8 polymorphisms studied here are located in the gene promoters and alter expression. Our aim was to investigate associations of MMP polymorphisms, solely and in haplotypes, with CAKUT in children. A case-control study with 101 pediatric patients and 281 controls was performed. The MMP-1 (-1607 1G/2G), -3 (5A/6A), and -8 (-799 C/T) genotypes were determined by PCR-restriction fragment length polymorphism. We found statistically significant associations of MMP-3 5A/6A polymorphism (p LT 0.0001) and 1G(-1607)-6A haplotype, with no preferences for MMP-8 -799C or T alleles, with CAKUT (OR = 2.93, 95 % CI 1.43-5.98, adjusted for gender, p = 0.003) and with obstructive uropathies in a subgroup of patients (OR = 4.57, 95 % CI 2.74-7.61, adjusted for gender, p LT 0.0001). MMP-3 genotypes and MMP-3 and -1 haplotypes encompassing either MMP-8 -799C or T alleles were associated with CAKUT and obstructive uropathies in pediatric patients. Still, functional and association studies are needed to elucidate evident roles of MMPs in CAKUT.

AB  - There is a growing interest in utilization of abundantly available materials, bio-mass or industrial byproducts, as precursors for the preparation of carbon materials. Short hemp fibers, acquired as waste from textile production, were used as low-cost precursor for production of carbon materials as a sorbent in the solid-phase extraction, for pesticide analysis in water samples. Different carbon materials were prepared by carbonization of unmodified and chemically modified hemp fibers. Activation of carbonized materials with potassium hydroxide improves sorption properties of carbonized hemp fibers by increasing the specific surface area (up to 2192 m(2)/g) and the amount of surface oxygen groups. The following parameters that may affect the solid-phase extraction procedure efficiency were optimized: different elution solvents and the pH value of pesticide solution. Extracts were analyzed by liquid chromatography-tandem mass spectrometry technique. For this study pesticides belonging to the different chemical classes were chosen. Obtained results indicate that carbonized and activated hemp fibers could be successfully applied as a solid-phase sorbent for the pesticide analysis in water samples. (C) 2012 Elsevier B.V. The IL-6-174G/C polymorphism and renal scarring in children with first acute pyelonephritis

AB  - Urinary tract infections (UTI) are common in infants and children and may result in serious complications, such as renal scarring, hypertension, and renal failure. Identification of the new markers in relation to acute pyelonephritis (APN) and its treatment is essential for designing interventions that would minimize tissue damage. This prospective study investigated the first UTI infection in 71 children (age range: 1-24 months) in respect to interleukin-6 (IL-6) -174G/C polymorphism and renal scarring. The patients were divided into an APN group and a lower UTI group according to dimercaptosuccinic acid (DMSA). The IL-6 -174G/C genotypes were determined by tetra-primer ARMSPCR. Serum IL-6 was significantly higher in the APN group than in the group with lower UTI (p LT 0.05). In both groups, the -174G/C genotype and allele frequencies did not differ significantly from the control group. The highest white blood cell (WBC) count was observed in the CC genotype (p LT 0.05). A non-significant trend toward higher serum IL-6 was observed in children with CC genotype. On follow-up DMSA imaging performed 6 months later, renal scarring was detected in 36.9% of APN children. We did not find the significant association of IL-6 -174G/C polymorphism with APN and/or postinfectious renal scarring. These results indicate that serum IL-6 concentrations were significantly higher in children with APN than in patients with lower UTI.

The angiotensin II receptor type 2 (AT2R) is one of proposed candidate genes for CAKUT, but the expression was never explored in humans. The aim was to establish the AM gene expression in human CAKUT concerning -1332A/G polymorphism, which might affect alternative splicing. Design and methods: Forty-eight patients with CAKUT constitute the basis of this study. Genotyping for -1332A/G, RT-PCR for AT2R gene expression and confirmation sequencing were performed. Results: The expression of Ex 1/2/3 and Ex 1/3 transcript splice variants of the AT2R mRNA were detected in human CAKUT tissue. The pattern was observed independently of A to G transition. Conclusions: The expression of AT2R mRNA in human CAKUT was established for the first time and was not affected by -1332A/G polymorphism in children with CAKUT.
