EO-based mouthwashes: Is there something that should be known?

AB  - The possibility to utilize the antibacterial and antibiofilm potential of essential oils (EOs) in dentistry is recognized and best seen in the design of EO-based mouthwashes. The up-to-date results, obtained with the most commonly used formulation being consisted of thymol, eucalyptol, menthol, and methyl salicylate (Listerine), proved its efficacy against plaque and gingival inflammation. However, novel alcohol-free products remain to be further investigated. Commercially available mouthwash containing curcumin, clove oil, mentha oil, eucalyptol, thymol and tea tree oil, and the mouthwash based on ginger EO (a-zingiberene, b-bisabolene, b-sesquiphellandrene, curcumene), reduced plaque index and improved periodontal status of the patients. The plaque index was also reduced and overall healing was improved after the surgical removal of the third molar by mouthwash containing John wort oil. In addition, Cinnamomum zeylanicum EO-based mouthwash and mouthwash containing peppermint oil were efficient against stomatitis and xerostomia, respectively. In addition to beneficial effects achieved in the mouth, i.e. at the appropriate site of application, novel investigations pointed-out that EO-based mouthwashes could be recognized as efficient in the reduction of bacterial and viral aerosols. This has been shown for Listerine, and for several mouthwashes containing following active substances of plants' EO origin: a-farnesene, b-farnesene, farnesol, nerolidol, a-bulnesene, eremanthin, b-sesquiphellandrene, spiroether, cinnamylacetate, cinnamaldehyde, eugenol, menthol, and carvacrol. This finding could be of special interest, in order to be included in further clinical studies related to COVID -19 pandemic.

Application of LIBS technique for dental analysis

AB  - This paper evaluates the possibilities of the original LIBS (Laser Induced Breakdown Spectroscopes) laboratory setup for the elementary analysis of human tooth tissues and tooth restorations. For that reason, samples of the teeth restored with amalgam and composite fillings were analyzed. In addition, the possibility of heavy metal diffusion from restoring materials into healthy tooth tissues was also examined. Plasma was induced by the TEA CO2 laser in the air under atmospheric pressure. Optical emission spectra with sharp and well-decomposed spectral lines and a negligibly low background were obtained. The results of the elemental analysis showed the presence of various trace elements in the tooth tissues (P, Ca, Mg) as well as within restorations such as Zn, Cu, Ag, Hg. The analysis also showed that cement successfully blocked diffusion from restoring materials (amalgam and composite) into the dentine below. Obtained preliminary results led to the conclusion that this alternative and cost-effective LIBS system can find application as a useful tool for further research on laser-induced plasma in dentistry

AB  - The aim of the study was to investigate the potency of Cinnamomum camphora cineoliferum and Melaleuca ericifolia essential oils (EOs) against facultative and obligate anaerobes isolated from teeth’ infected root canals. Biocompatibility of the EOs was also in the study focus. Chemical composition of the EOs was determined by GC/MS analysis. Their antibacterial potential and cytotoxicity were evaluated by microdilution and MTT assays, respectively. Statistical analysis was performed by Student’s t-test and one way ANOVA. Two the most abundant constituents of C. camphora cineoliferum EO were 1,8-cineole and sabinene, while in M. ericifolia EO the dominant were linalool and 1,8-cineole. MICs achieved against all tested strains were in the ranges 0.95±0.50 - 22.50±0.00 mg/mL and 0.65±0.00 - 23.35±7.80 mg/mL for the C. camphora cioneoliferum and M. ericifolia EOs, respectively. Statistical analysis showed that response to both oils has not differed between facultative and obligate anaerobes. Sensitivity pattern of the tested strains was different for the EOs and the positive control triple antibiotic paste (TAP): while E. faecalis and certain strains of S. salivarius were among the most sensitive to the oils, they were the least susceptible to the conventional antimicrobial TAP. Although the observed cytotoxicity of the EOs after 30 min of cell exposition was high (50.78-66.35% and up to 70% for C. camphora cioneoliferum and M. ericifolia EOs, respectively) it was lower than that of positive control (1.5% NaOCl, p<0.05). To conclude, both EOs showed promising antibacterial potential and achieved at least the same or even better biocompatibility than the conventional intracanal antimicrobials. Obtained results strongly encourage future research with an aim to formulate C. camphora cioneoliferum and M. ericifolia EO- based irrigants and medicaments, that could be used in endodontic practice.

Cymbopogon martinii essential oil nanoemulsions: Physico-chemical characterization, antibacterial and antibiofilm potential against Enterococcus faecalis

AB  - Cymbopogon martinii is an aromatic crop cultivated for the production of its essential oil (EO) which has different applications. Bearing in mind that C. martinii EO (CMEO) has already been proved for high antibacterial potential the aim of this study was to formulate stable CMEO nanoemulsions, being capable to combat Enterococcus faecalis biofilm, especially within infected tooth’s root canal. Ten emulsions with variable CMEO content (2.5–10 % v/v) and Tween 80 (T80, 2.5–15 % v/v) were formulated (designated as ECMEO:T80) and screened for volume-weighted mean diameter (D4,3) and stability, using dynamic light scattering measurements. Chemical composition of stable nanoemulsions was monitored by UV–VIS and FTIR, while their antibacterial activity was estimated in microdilution assay. Antibiofilm properties of selected nanoemulsions were tested in vitro (cristal violet assay) and ex vivo (within root canals of the extracted teeth). Results showed that formulations containing 10 % of T80 and CMEO volume ≤ 6 %, as well as those having constant 2.5 % of CMEO and variable T80 volume (≤ 10%), namely E2.5:2.5, E2.5:5, E2.5:7.5, E2.5:10, E4:10, and E6:10, were stable and nanosized (D4,3 <100 nm). FTIR and UV–VIS analyses confirmed successful encapsulation of the CMEO by T80 and showed that nanoemulsification did not affect CMEO composition. Evaluation of antibacterial potential showed that the most efficient were E6:10, E2.5:2.5 and E4:10 (MIC values 0.37–1.97 mg mL−1). Analysis of dependency of determined MICs on mean diameter and CMEO volume showed that antibacterial potential increased with both parameters. E6:10, E2.5:2.5 and E4:10 also reduced in vitro biofilm (inhibitory range 17.6–58.4 %), as well as intracanal biofilm (reduction ~2Log CFU). Presented results suggest E6:10, E4:10 and E2.5:2.5 as the good candidates for further research.

Essential oils as adjuvants in endodontic therapy: myth or reality?

AB  - To enhance endodontics, the search for new antibacterials that can improve infected tooth root canal treatment is ongoing. As potent antibacterial and antibiofilm agents, essential oils (EOs) have been suggested as novel endodontic materials. Several studies indicate that EO-based irrigants and medicaments show promising reductive potential against the most important intracanal pathogen, Enterococcus faecalis, and notably contribute to intracanal biofilm eradication. In terms of additional benefits that EO-based endodontic materials can provide, their antioxidant and anti-inflammatory potential are also important, but they have only scarcely been explored in research. Investigations into the benefits of EO-based endodontic materials together with their biocompatibility are needed. The results presented in this review strongly encourage further research on this topic.

AB  - Aim: The objective was to formulate and characterize the nanoemulsion based on Cymbopogon citratus oil, intended for use in infected teeth root canal therapy. The investigation of the antioxidant and antibiofilm potential toward Enterococcus faecalis was aimed as well. Materials & methods: Characterization of oil (by GC/MS analysis) and nanoemulsion (by dynamic light scattering instrument), and determination of antibacterial (by microdilution assay), antibiofilm (by crystal violet assay) and antioxidant properties (by 2,2-diphenyl-1-picryl-hydrazyl-hydrate and thiobarbituric acid assay methods) were provided. Antibiofilm efficacy of irrigation procedure including nanoemulsion was screened on extracted teeth (by CFU-counting assay). Results: Notable antibacterial and antibiofilm activity, both against forming and preformed biofilms of oil, was observed. Irrigation involved nanoemulsion showed remarkable antibiofilm potential. Both substances induced some antioxidant activity. Conclusion: Results encourage further research with the aim of application of the nanoemulsion in dental practice.

AB  - The study aimed to investigate the potency of Leptospermum petersonii, Eucalyptus citriodora, Pelargonium graveolens, and Pelargonium roseum essential oils (EOs) against Streptococcus sanguinis and Streptococcus salivarius dental isolates. To examine the variation in sensitivity among strains of different origin, isolates from symptomatic and asymptomatic root canal infections, as well as the reference, strains S. sanguinis ATCC 10556 and S. salivarius ATCC 9222 were used in the study. EOs were chemically characterised (GC/MS). Their antibacterial and antibiofilm properties were detected by microdilution and CV assay respectively. Statistical analysis was performed by One way ANOVA. Examined bacterial strains, regardless clinical or ATCC, exhibited the highest sensitivity to L. petersonii and E. citriodora EOs (MIC ranged 0.53±0.00 - 5.88±1.27 mg/ml and 0.08±0.03 - 9.37±1.25 mg/ml respectively), while two other EOs were less efficient. Comparison of MIC values showed that sensitivity of the isolates recovered from symptomatic infections was the least, while the susceptibility of asymptomatic isolates and especially of reference strains were higher. EO inhibition of biofilm formation differed between clinical isolates of S. salivarius and their reference strain. Inhibition was higher in the case of S. salivarius ATCC 9222 (84 %-89 %) in comparison to that of clinical isolates (24 %-75 %). Regardless of the strains tested, E. citriodora exhibited the highest antibiofilm potential. To conclude: L. petersonii induced notable antibacterial potential, while E. citriodora EO proved promising antibacterial and antibiofilm activity. Due to the variable sensitivity of the isolates to EOs, it is suggested to screen activity on a large number of strains, especially from symptomatic infections.

AB  - OBJECTIVE The objective was to determine the antibiofilm efficacy of Cymbopogon martinii and Thymus zygis essential oils and to estimate the disruption potential of oil-based endodontic irrigants on the multispecies biofilm formed in the root canals of extracted teeth. DESIGN The essential oils were characterized (GC-MS), while their antibacterial and cytotoxic properties were detected by microdilution and MTT assays. Particles sizes and polydispersity indices were determined for the irrigants. The isolates from root canals of pediatric patients were identified (MALDI-TOF). The multispecies biofilms were formed from Streptococcus mitis, Streptococcus sanguinis and Enterococcus faecalis. Antibiofilm properties of the essential oils and irrigants were determined by crystal violet and plate counting assay. Statistical analysis was performed by one-way ANOVA. RESULTS The screening of biofilm biomass revealed a disruption potential of C. martinii essential oil. The plate counting assay showed the efficacy of both oils in diminishing cell viability: high in biofilms (reduction of log10CFU was 2.75-2.87) and moderate in the planktons formed above. The essential oil-based irrigants showed the same antibiofilm activity as in the control (1.5 % sodium hypochlorite) for C. martini and almost 2-fold higher for T. zygis. Successive irrigations with 1.5 % sodium hypochlorite, saline and an oil-based irrigant was more efficient for C. martini than for the control (reductions of log10CFU was 1.69 and 1.14, respectively). CONCLUSIONS Notable activities achieved by essential oils and oil-based irrigants, particularly regarding C. martini, against the biofilm consisting of S. mitis, S. sanguinis, and E. faecalis, are expected to encourage further research in endodontics.

Microbiological analysis of primary infected root canals with symptomatic and asymptomatic apical periodontitis of young permanent teeth

AB  - Background/Aim: Understanding the composition of bacteria in infected root canals is important for ameliorating the treatment strategies that lead to the elimination of pathogens and infection control, but also prevent reinfection. Aim of this study was to investigate microbial composition of primary infected root canals with apical periodontitis of young permanent teeth, originating form school children in Serbia, and its association with clinical symptoms. Material and Methods: To determine the bacterial composition of infected root canals in children, 35 endodontic samples were obtained. The identification of cultured bacteria was performed by MALDI-TOF MS analysis. The presence or absence of clinical symptoms were recorded. Results: Facultative anaerobes were 2,2 times more frequent than obligate anaerobes. The most common facultative anaerobes belonged to following genera, Streptococcus (58 isolates), Actinomyces (10) and Enterococcus (8), while predominant obligate anaerobes, belonged to genera Veillonella (15), Prevotella (9) and Fusobacterium (8). The most common clinical isolates recovered from infected root canals with symptomatic apical periodontitis were Veillonella parvula (10) and Fusobacterium nucleatum (7), while from the asymptomatic ones, they were Streptococcus mitis/Streptococcus oralis (5). Prevalence of Parvimonas micra, Prevotella buccae and Streptococcus constellatus within the root canals might be associated to clinical symptoms. Conclusions: Species of genera Streptococcus and Veillonella were the most common isolates from primary infected root canals with apical periodontitis in Serbian school children. Facultative anaerobes were predominant over obligate anaerobes. The prevalence of obligate anaerobes was much higher in symptomatic compared to asymptomatic root canal infections. No specific bacterial strain might be associated to a single examined clinical symptom (pain, tenderness to percussion or swelling), but majority of the strains are associated to all of the examined three symptoms.

The most common facultative anaerobes belonged to following genera, Streptococcus (58 isolates), Actinomyces (10) and Enterococcus (8), while predominant obligate anaerobes, belonged to genera Veillonella (15), Prevotella (9) and Fusobacterium (8). The most common clinical isolates recovered from infected root canals with symptomatic apical periodontitis were Veillonella parvula (10) and Fusobacterium nucleatum (7), while from the asymptomatic ones, they were Streptococcus mitis/Streptococcus oralis (5). Prevalence of Parvimonas micra, Prevotella buccae and Streptococcus constellatus within the root canals might be associated to clinical symptoms. Conclusions: Species of genera Streptococcus and Veillonella were the most common isolates from primary infected root canals with apical periodontitis in Serbian school children. Facultative anaerobes were predominant over obligate anaerobes. The prevalence of obligate anaerobes was much higher in symptomatic compared to asymptomatic root canal infections. No specific bacterial strain might be associated to a single examined clinical symptom (pain, tenderness to percussion or swelling), but majority of the strains are associated to all of the examined three symptoms.",

AB  - Background/Aim. The main task of endodontic treatment is to eliminate pathologically altered tissue, to disinfect root canal space and to obtain its three-dimensional hermetic obturation. The main purpose of this study was to evaluate antimicrobial activity of new endodontic nano-structured highly active calcium silicates based materials albo-mineral plyoxide carbonate aggregate (ALBO-MPCA) and calcium silicates (CS) in comparison to mineral trioxide aggregate (MTA+) and UltraCal XS (CH). Methods. The antimicrobial activity of materials was tested against Staphylococcus aureus (ATCC 25923) and Enterococcus faecalis (ATCC 14506) strains, and following clinical isolates: Rothia dentocariosa, Enterococcus faecalis, Staphylococcus aureus, Streptococcus anginosus and Streptococcus vestibularis using a double layer agar diffusion test. The pH measurements were performed using the pH meter. Total amount of released ions was determined by inductively coupled plasma optical emission spectrometry (ICP-OES). Results. All tested materials showed the best antibacterial potential after 1 h of incubation. After 3h and 24h of the incubation period, the antibacterial potential of all tested materials were similar. The Agar diffusion test showed that ALBO-MPCA, CS and MTA+ had similar inhibition zones (p > 0.05), except in the activity against Staphylococcus aureus where ALBO-MPCA showed better antimicrobial properties than MTA+ in 3h and 24h of the incubation period (p < 0.05). Following 24h of the incubation, the inhibition zones were the strongest with CH against Staphylococcus aureus (16.67 ± 2.34 mm) followed by ALBO-MPCA (14.67 ± 1.21 mm) and the weakest with CS against Enterococcus faecalis (6.50 ± 1.76 mm). CH showed the highest pH, followed by ALBO-MPCA, CS and MTA+. Conclusion. The expressed antibacterial effects indicate that materials based on nano-structured highly active calcium silicates represent effective therapeutic agents for root canal obturation in one-visit apexification treatment, therefore they are recommend for further examination and clinical trials as they are proposed for MTA substitution. © 2019, Inst. Endodontic treatment of traumatized teeth with chronic periapical lesions using antibiotic paste and mineral trioxide aggregate obturation - a preliminary study

AB  - Introduction/Objective The purpose of this study was to assess effectiveness of endodontic root canal procedures in traumatized permanent teeth with necrotic pulps and chronic periapical lesions after definitive obturation with mineral trioxide aggregate (MTA) products. Adobe Photoshop CS (San Jose, CA, USA) image-analysis software was used for healing assessment. Methods Twenty-seven traumatized single-rooted permanent teeth diagnosed with necrotic pulps and chronic periapical lesions were treated with non-surgical procedure using a tri-antibiotic paste and calcium hydroxide as intra-canal medication. Definitive obturation was performed with ProRoot MTA (Dentsply Tulsa Dental Specialties, Tulsa, OK, USA) or MTA+ Cerkamed (Cerkamed, Stalowa Wola, Poland). Control follow-ups were done three, six, 12, and 24 months following the completion of treatment. Results The positive clinical outcome was recorded in 24 (88.9%) cases, while radiographic success was present in 26 (96.3%) cases. A statistically significant decrease in the sizes of periapical lesions was consistently observed at follow-up periods (p < 0.001). There was no statistically significant difference between the two tested MTA materials (p > 0.05). Conclusion The MTA products were effective for the root canal obturation and appeared to contribute to the significant reduction or complete regression of periapical lesions in teeth treated. The presented procedure may be proposed for everyday clinical practice. © 2019, Serbia Medical Society. All rights reserved.

Nanostructured endodontic materials mixed with different radiocontrast agents — biocompatibility study

AB  - The aim of this study was to investigate the biocompatibility of nanostructured materials based on highly active calcium silicates mixed with different radiocontrast agents in comparison to MTA+ using in vitro and in vivo model. Morphology of materials’ samples was analyzed using SEM while the phase compositions were identified by XRD. pH values of materials’ suspensions were conducted by pH-meter. The cytotoxicity of materials’ solutions was tested by MTT test (100, 50, 25 and 12.5 mg/ml). LDH and 3H-thymidine assay were utilized for biocompatibility investigations of materials’ eluates (24 h, 7 day and 21 day). Eighteen Guinea pigs were used for intramuscular implantation, as teflon tubes with freshly prepared materials were placed into intramuscular pockets. All samples were composed of round and needle-like particles equally distributed with Ca/Si ratio ~2.7 at%, with the presence of hydrated calcium silicate phases. The pH values of ALBO-MPCA1 and ALBO-MPCA2 were high alkaline, while in case of MTA+ they were lower and continuously declined (p < 0.05). Investigated materials didn’t exhibit dose-dependent effect on metabolic activity of L929 cells (p > 0.05). Significant differences in the percentage of cytotoxicity between diluted and undiluted extracts between all tested materials after 24 h and 7 day were noticed (p < 0.05). Increase in L929 cells proliferation was noticed in case of undiluted eluates of ALBO-MPCA1 and ALBO-MPCA2 after 7 day (p < 0.05). There were no statistically significant differences in the intensity of inflammatory response between investigated materials and control group after 60 day (p > 0.05). Evaluation of biocompatibility of both ALBO-MPCA1 and ALBO-MPCA2 indicate their potential clinical use. AB  - This study examined the potential of a new porous calcium hydroxyapatite scaffold covered with poly (lactide-co-glycolide) (PLGA) as a bone substitute, identifying its advantages over Geistlich Bio-Oss (R), considered the gold standard, in in vivo biofunctionality investigations. Structural and morphological properties of the new scaffold were analyzed by scanning electron and atomic force microscopy. The biofunctionality assays were performed on New Zealand white rabbits using new scaffold for filling full-thickness defects of critical size. The evaluated parameters were: the presence of macrophages, giant cells, monoocytes, plasma cells, granulocytes, neo-angiogenesis, fibroplasia, and the percentage of mineralization. Parallel biofunctionality assays were performed using Geistlich Bio-Oss (R). The appearance of bone defects 12 weeks after the new scaffold implantation showed the presence of a small number of typical immune response cells. Furthermore, significantly reduced number of capillary buds, low intensity of fibroplasia and high degree of mineralization in a lamellar pattern indicated that the inflammation process has been almost completely overcome and that the new bone formed was in the final phase of remodeling. All biofunctionality assays proved the new scaffolds suitability as a bone substitute for applications in maxillofacial surgery. It showed numerous biological advantages over Geistlich Bio-Oss (R) which was reflected mainly as a lower number of giant cells surrounding implanted material and higher degree of mineralization in new formed bone.

Biocompatibility Investigation of New Endodontic Materials Based on Nanosynthesized Calcium Silicates Combined with Different Radiopacifiers

AB  - Introduction: The aim of this article was to analyze biocompatibility and bioactivity of new endodontic materials on the basis of nanosynthesized calcium silicates (ALBO-MPCA(1) and ALBO-MPCA(2)) combined with different radiopacifiers in comparison with MTA(+). Methods: Morphology of the samples was studied by scanning electron microscopy, and the pH and ion release analysis were also assessed. Biocompatibility of materials eluates (24-hour, 7-day, and 21-day) was conducted by using MIT test. Twelve New Zealand white rabbits were used for intraosseous implantation. Four calvarial defects per animal were created and filled with freshly prepared investigated materials. Results: Samples mostly consisted of agglomerates built up from nanoparticles, preferably spherical and rod-like. There was no significant difference among pH values of materials eluates after 24 hours (P GT .05). The amount of calcium and aluminum ion release decreased, whereas the amount of magnesium and bismuth (ALBO-MPCAl, MTA(+)) and barium (ALBO-MPCA(2)) increased during 21-day period. The metabolic activity of cells increased after the extraction time, except in case of undiluted elutes of ALBO-MPCA(2) and ALBO-MPCAI (21-day). Histologic analysis of the samples revealed newly formed bone tissue with moderate inflammation for all investigated materials, which subsided during 90-day period to mild. Both MIA(+) and ALBO-MPCAI were in direct contact with the newly formed bone tissue. After 90 days, statistically significant difference in hard tissue formation was observed in comparison of MIA(+) and ALBO-MPCki with control group (P LT .05). Conclusions: Experimental materials ALBOMPCA, and ALBO-MPCA(2) possess both biocompatibility and bioactivity. Because ALBO-MPCAA provokes favorable biological response, it is especially good candidate for further clinical investigations.

AB  - A synthetic bone substitute based on calcium hydroxyapatite (CHA) and poly(lactic-co-glycolic) acid (PLGA), described in this paper, was synthesized to fulfill specific requirements like biodegradability, satisfying mechanical properties, optimal porosity and nanotopology, osteoinductive and osteoconductive properties, and so forth. Structural and morphological properties of the new scaffold were analyzed by micro computed tomography and scanning electron microscopy, while its physicochemical properties were investigated by X-ray diffraction and infrared spectroscopy. In vivo biological investigations of the synthesized scaffold were conducted over the cutaneous irritation and biofunctionality assays on rabbits and the test of acute systemic toxicity on mice. The results showed that the scaffold is not irritant and that it does not exhibit any symptoms of acute toxicity. Biofunctionality assays which include evaluation of the presence of various cells of immune response, the presence of neoangiogenesis, percentage of mineralization of newly formed bone, and fibroplasia in the tissue indicated that the new scaffold is suitable for the application in maxillofacial and dental surgery as a bone substitute. Also, it showed significant advantages over commercial product Geistlich Bio-Oss(R) from the aspect of some parameters of immunological response.

AB  - Introduction Biodegradable porous scaffolds are found to be very promising bone substitutes, acting as a temporary physical support to guide new tissue regeneration, until the entire scaffold is totally degraded and replaced by the new tissue. Objective The aim of this study was to investigate cytotoxicity of a synthesized calcium hydroxyapatite-based scaffold, named ALBO-OS, with high porosity and optimal topology. Methods The ALBO-OS scaffold was synthesized by the method of polymer foam template. The analysis of pore geometry and scaffold wallstopography was made by scanning electron microscope (SEM). The biological investigations assumed the examinations of ALBO-OS cytotoxicity to mouse L929 fibroblasts, using 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromidefor (MU) and lactate dehydrogenase (LDH) tests and inverse phase microscopy. Results The SEM analysis showed high porosity with fair pore distribution and interesting morphology from the biological standpoint. The biological investigations showed that the material is not cytotoxic to L929 cells. Comparison of ALBO-OS with Bio-Oss, as the global gold standard as a bone substitute, showed similar results in MTT test, while LDH test showed significantly higher rate of cell multiplication with ALBO-OS. Conclusion The scaffold design from the aspect of pore size, distribution, and topology seems to be very convenient for cell adhesion and occupation, which makes it a promising material as a bone substitute. The results of biological assays proved that ALBO-OS is not cytotoxic for L929 fibroblasts. In comparison with Bio-Oss, similar or even better results were obtained.

Analysis of chemical composition of the most efficient essential oils towards Enterococcus faecalis referent strain ATCC 29212 and clinical isolates

AB  - Twenty one essential oils (EOs) documented their significant antimicrobial effect with regard to our pre-set criterion of the Minimal Inhibitory Concentration (MIC ≤ 200 μL / mL) of EOs towards Enterococcus faecalis (ATCC 29212 and or clinical isolates); the best effect MIC 0.4 μL / mL (approx. 0.26 μg / mL) achieved Satureja horvatii L. EO, while the EOs with the lowest antimicrobial efficacy were Rosmarinus officinalis L. and Achilea milefolium L., both with MIC s160.0 μg / mL. Analysis of the MIC values within the groups revealed that ATCC strain of E. faecalis is generally lower, ranging from 0.26 to 156 μg / mL, in comparison to those for clinical isolateswhich ranged from 10 to 160 μg / mL. Twelve 12 components that are common in EOs whith MIC s ≤ 200 μg / mL in testings towards both, the clinical and referent strains are given in descending order according to number of oils they are present in: trans-β-caryophyllene (13) > myrcene (8) > α-pinene (8) > linalool (7) > p-cymene (7) > borneol (7) > geraniol (6) > camphene (6) > limonene (5) > 1,8- cineol (5) > γ -terpinene (5) > α- terpinene (4). Comparison of EO constituents reviled that only, geraniol and 1,8-cineol, contributed with ≥ 10 % to more than one EO (MIC 0.3-200 μg / mL) efficient against both E. faecalis strains. Thirteen components in 11 EOs with MIC ≤ 200 μg / mL towards ATCC 29212 were representative based on their contents in EOs: eugenol 82.9 % > thymol 63.7 % > hexadecanoic acid 47.8 % > menthol 46.6 % > cis-b-ocimene 44.2 % > geranial 42.1 % > trans-β-caryophyllene 40.8 % > citronellal 36.7 % > α-pinene 31.2 % > neral 30.5 % > α-eudesmol 22.4 % > citronellol 13.1 % > menthone 11.3 %. Following seven components, representative in 10 EOs with MIC ≤ 200 μg / mL towards clinical isolates, are presented in order of their contribution to EOs: phenylethyl alcohol 57.7 % > geranial 32.9 % > neral 22.2 % > p- cymene 20 % > carvacrol 14 % > α-pinene 11.5 % > linalool 11.4 %. Out of 21 highly efficient EOs selected in this study, six EOs proved to be the most efficient (MIC ≤ 30 μg / mL ); three oils in control of E.faecalis ATCC strain (Satureja horvatii, Mentha pulegium and Rosmarinus officinalis) and other three in control of E. faecalis clinical isolates (Leptospermum petersonii, Thymus algeriensis, Thymus serpyllum). Thymol is a major component in three out of the six aforementioned most efficient EOs. The aim of our study was to investigate differences in efficacy of selected EOs that proved to possess great antimicrobial activity, towards the referent strain ATCC 29212 and clinical isolates of E. faecalis on, and to estimate which of their constituents might contribute to desired activity, as 'markers compunds'.

AB  - Millions of patients worldwide need surgery to repair or replace tissue that has been damaged through trauma or disease. To solve the problem of lost tissue, a major emphasis of tissue engineering (TE) is on tissue regeneration. Stem cells and highly porous biomaterials used as cell carriers (scaffolds) have an essential role in the production of new tissue by TE. The cellular component is important for the generation and establishment of the extracellular matrix, while a scaffold is necessary to determine the shape of the newly formed tissue and facilitate migration of cells into the desired location, as well as their growth and differentiation. This review describes the types, characteristics and classification of stem cells. Furthermore, it includes functional features of cell carriers - biocompatibility, biodegradability and mechanical properties of biomaterials used in developing state-of-the-an scaffolds for TE applications, as well as suitability for different tissues. Moreover, it explains the importance of nanotechnology and defines the challenges and the purpose of future research in this rapidly advancing field.

AB  - The aim of this study was to examine the differential capacity of isolated dental pulp stem cells (SHED) cultured onto four different scaffold materials. The differential potential of isolated SHED was examined on the following scaffolds: porous hydroxyapatite (pHAP) alone or combined with three polymers [polylactic-co-glycolic acid (PLGA), alginate, and ethylene vinylacetate / ethylene vinylversatate (EVA/EVV)]. SHED were isolated by outgrowth method and characterized by the flow cytometry. Viability of cells grown with scaffolds was assessed by MTT and LDH assays. No significant cytotoxic effect of any of the tested materials was shown. Staining with alizarin red and estimated alkaline phosphatase activity to identify differentiation, demonstrated osteoblastic phenotype of SHED and newly deposited and mineralized extra cellular matrix (ECM) in presence of all tested scaffolds. The developed ECM seen at scanning electronic micrographs additionally confirmed the osteogenic differentiation and biocompatibility between cells and materials. AB  - A synthesis of a new dental material, based on nano-structured highly active calcium silicates and calcium carbonates, is described in this paper. Phase analysis of this material, before and after hydration, was performed by X-ray diffraction and showed that active silicate phases were transformed into tobermorite phase, while carbonates remained unchanged. The mechanism of hydration was fully described. The morphology of the sample was studied by SEM, and typical appearance of the present phases was particularly discussed. The mechanical properties and setting time of this material make it very promising for potential application in dental practice.

The efficacy of hydrothermally obtained carbonated hydroxyapatite in healing alveolar bone defects in rats with or without corticosteroid treatment

AB  - Background/Aim. Autogenous bone grafting has been the gold standard in clinical cases when bone grafts are required for bone defects in dentistry. The study was undertaken to evaluate multilevel designed carbonated hydroxyapatite (CHA) obtained by hydrothermal method, as a bone substitute in healing bone defects with or without corticosteroid treatment in rats as assessed by histopathologic methods. Methods. Bone defects were created in the alveolar bone by teeth extraction in 12 rats. The animals were initially divided into two groups. The experimental group was pretreated with corticosteroids: methylprednisolone and dexamethasone, intramuscularly, while the control group was without therapy. Posterior teeth extraction had been performed after the corticosteroid therapy. The extraction defects were fulfilled with hydroxyapatite with bimodal particle sizes in the range of 50-250 mu m and the sample from postextocactional defect of the alveolar bone was analyzed pathohystologically. Results. The histopatological investigations confirmed the biologic properties of the applied material. The evident growth of new bone in the alveolar ridge was clearly noticed in both groups of rats. Carbonated HA obtained by hydrothermal method promoted bone formation in the preformed defects, confirming its efficacy for usage in bone defects. Complete resorption of the materials particles took place after 25 weeks. Conclusion. Hydroxyapatite completely meets the clinical requirements for a bone substitute material. Due to its microstructure, complete resorption took place during the observation period of the study. Corticosteroid treatment did not significantly affect new bone formation in the region of postextractional defects.

Use of mineral trioxide aggregate in the treatement of traumatized teeth in children - Two case reports

AB  - Introduction. Dental injuries in immature permanent teeth often result in endodontic complications. Apexification technique using calcium hydroxide is associated with certain flaws, such as long treatment time, the possibility of tooth fracture and incomplete calcification. The use of an apical plug employing mineral trioxide aggregate (MTA) is an alternative treatment option. Case report. We reported the successful treatment of 4 maxillary incisors (in a 7-year-old boy and a 10-year-old girl) with open apices and periapical lesions. Apical portions of the canals were filled with MTA plugs in both cases. Coronal parts of the root canals were filled with gutta-percha and sealer. Clinical findings were clear 6 months after the definite obturation with no pathological changes on the radiographs in both cases. Conclusion. The use of MTA for apical plugging appears to be a valid treatment option in traumatized immature teeth with endodontic complications.

Self assembly of biomimetic hydroxyapatite on the surface of different polymer thin films

AB  - The role of the process of designing a scaffold in bone tissue engineering is to provide optimal conditions for new bone tissue growth. The primary concern of such engineering is to create an adequate nanotopology of the scaffold inner walls, which can initiate the growth and activity of bone cells. Here, we present a completely new scaffold designing process based on a biomimetic approach in order to improve the nanostructure of pore walls of previously-made calcium hydroxyapatite (CHA) porous scaffolds. CHA porous scaffolds were covered with different polymer thin films (alginate, cellulose and PLGA) and exposed to simulated body fluid (SBF) for 42 days. SBF induced in situ formation of bone-like apatite phases on the surface of CHA/polymer composites. Fourier Transformed Infrared (FTIR) spectroscopy showed that the biomimetically-assembled phase is CHA of slightly shifted stoichiometry. X-ray diffraction confirmed that CHA is self-assembled on the surface of all investigated thin films. The calculation of crystallite sizes showed small differences in the degree of crystallinity between different samples. Scanning electron microscopy revealed a dominant blow-ball morphology of CHA particles (size 1 - 5 mu m) with nano-sized branches on their surfaces.

X-ray diffraction confirmed that CHA is self-assembled on the surface of all investigated thin films. The calculation of crystallite sizes showed small differences in the degree of crystallinity between different samples. Scanning electron microscopy revealed a dominant blow-ball morphology of CHA particles (size 1 - 5 mu m) with nano-sized branches on their surfaces.",
