AB  - This paper explains the basis for the excitation energy-independent fluorescence emission of biomass-derived carbon dots (CDs) and shows that these CDs have excellent anti-melanoma and anti-metastatic potential. Additionally, we demonstrate that the black carrots´-derived CDs can be exploited as cell cycle-sensing agents, because of the interaction with chromatin material. Besides their optical properties, fluorescent CDs have gained increased attention for image-guided cancer treatment due to their water solubility, environmental friendliness, affordability, ease of synthesis, and primary biocompatibility. CDs have excellent photostability, determined by their precursors and synthesis pathways. In this study, CDs with chemically homogenous surface functional groups were made using a hydrothermal technique from black carrot extract, an anthocyanin-rich substance derived from biomass. The anti-cancer and anti-metastatic properties of black carrot-derived CDs can be attributed to flavylium cations on the surface, spherical forms, and high water dispersibility. Most importantly, these CDs demonstrate a stable emission at a single wavelength, 612 nm, independent of the excitation energy, which we have explained theoretically for the first time.

AB  - In this work, we have applied the Informational Spectrum Method (ISM) to discover a potential protein target in the HeLa cervical cancer cell line for [Pd(dach)Cl2] complex. Since Pd complexes are considered an alternative to traditionally used Pt complexes in anti-cancer therapy, it is essential to elucidate the mechanism of their action. A detailed analysis that also involves screening the known protein databases revealed the proteins of the SOSS complex as the most probable [Pd(dach)Cl2] targets. Since this protein maintains genomic stability, this result shows the potential of the Pd(II) complex as an anti-cancer drug.

AB  - Nanocomposite system formulated from surface-modified S-doped carbon dot (S-CD) nanoparticle with a potential metallodrug, palladium(II) complex, dichloro(1,2-diaminocyclohexane)palladium(II), [Pd(dach)Cl2] (Pd@S-CD), was investigated as a model system for the treatment of cervical carcinoma (HeLa) cells. To examine the intracellular biochemical effects induced by the Pd@S-CD, we used Synchrotron Radiation-based Fourier-transform infrared spectroscopy (SR FTIR). SR FTIR spectroscopy was employed to investigate the alterations in cellular components’ biochemical composition and secondary structure upon exposure to Pd@S-CD. Spectral analysis, complemented by statistical techniques, revealed changes in biomolecules, lipids, proteins, nucleic acids, and carbohydrates caused by the treatment with Pd@CDs. These results and the increased cytotoxicity of the system demonstrate its high anti-cervical cancer therapeutic potential.

Physico-chemical and mechanical properties of geopolymer/zircon composites

AB  - The effect of zircon (ZrSiO4) on the physico-chemical and mechanical properties of geopolymer/zircon composites was examined in this study. Four geopolymer/zircon composites containing 10, 20, 30 and 40 wt.% zircon were prepared from metakaolin with alkali activators. Characterization of the obtained geopolymers was performed by X-ray diffraction (XRD), Scanning electron microscope (SEM-EDS), Fourier transform infrared spectroscopy (FTIR) and Matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF). XRD results did not confirmed the formation of interconnected phases between added zircon, starting aluminum silicates and alkali activators. Compressive strength of prepared geopolymer was examined. The maximum obtained compressive strength of 70.15 MPa was measured in sample containing the smallest fraction of zircon, i.e., 10 wt.%. Addition of larger amount of zircon (20 wt.%) hinders the progress of geopolymerization reaction and consequently decreases compressive strength.

Potential of MALDI TOF mass spectrometry for detection and quantification of corticosterone in the blood of loggerhead sea turtles

AB  - The endangered loggerhead sea turtles (Caretta caretta) are a relatively abundant sea turtle species in Madeiran waters, where they encounter various environmental stressors: from natural to anthropogenic. The physiological stress response is increased corticosterone (CS) level in the blood, achieving 2 ng/mL. Although there are various analytical methods for determining the CS concentration in turtle blood, most of them require tedious procedures for the preparation. In this work, we have tested the possibility of using matrix-assisted laser desorption and ionization time-of-flight mass spectrometry (MALDI TOF MS) for fast screening and detection of CS in blood from a turtle captured at the coast of Madeira Island. MALDI does not require sample derivatization and is suitable for low sample volume. The linearity, precision, and limit of detection and quantification with three organic MALDI matrices were investigated in this study. The signal-to-noise ratio of the CS-derived signal was used as a parameter for quantification. Isotopically labelled corticosterone was added as a correction factor at fixed concentration to achieve better linearity and precision. Our results demonstrate that this method has the potential for quantification of CS in turtle blood. However, the lowest concentration of CS extracted from plasma that was still detectable by MALDI TOF MS was about 0.04 mg/mL, which is about ten times higher than the expected CS concentration in blood.

AB  - In the last decade, targeting membrane lipids in cancer cells has been a promising approach that deserves attention in the field of anticancer drug development. To get a comprehensive understanding of the effect of the drug [Ru(η5-Cp)(PPh3)2CN] (RuCN) on cell lipidic components, we combine complementary analytical approaches, matrix-assisted laser desorption and ionization time-of-flight mass spectrometry (MALDI TOF MS) and synchrotron radiation-based Fourier transform infrared (SR-FTIR) spectroscopy. Techniques are used for screening the effect of potential metallodrug, RuCN, without and with drug carriers (carbon dots (CDs) and nitrogen-doped carbon dots (N-CDs)) on the lipids of the human ovarian cancer cell line A2780. MALDI TOF MS results revealed that the lysis of ovarian cancer membrane lipids is promoted by RuCN and not by drug carriers (CDs and N-CDs). Furthermore, SR-FTIR results strongly suggested that the phospholipids of cancer cells undergo oxidative stress after the treatment with RuCN that was accompanied by the disordering of the fatty acid chains. On the other hand, using (N-)CDs as RuCN nanocarriers prevented the oxidative stress caused by RuCN but did not prevent the disordering of the fatty acid chain packing. Finally, we demonstrated that RuCN and RuCN/(N-)CDs alter the hydration of the membrane surface in the membrane–water interface region.

AB  - Carbon dots (CDs) and N-carbon dots (N-CDs) loaded with Ru-complex (CDs@RuCN, N-CDs@RuCN, respectively) were investigated as media imposing biochemical changes induced by UV illumination of ovarian cancer, A2780, and osteosarcoma, CAL72, cells. Synchrotron radiation-based Fourier Transform Infrared Spectroscopy was performed, and the spectra were subjected to a Principal Component Analysis. The CDs@RuCN and N-CDs@RuCN effects on cancer cells were analyzed by the theoretical modelling of the stability of the composite systems and a protein database search. Moreover, a detailed evaluation of surface and optical properties of CDs@RuCN and N-CDs@RuCN was carried out. Results demonstrated selective action of the CDs@RuCN and N-CDs@RuCN-based photodynamic therapy, with N-CDs@RuCN being the most active in inducing changes in A2780 and CDs@RuCN in CAL72 cells. We assume that different surface charges of nanoparticles led to direct interactions of N-CDs@RuCN with a Wnt signalling pathway in A2780 and those of CDs@RuCN with PI3–K/Akt in CAL72 cells and that further biochemical changes occurred upon light illumination.

ROS-mediated proapoptotic antitumor effects of Ru(II) complex on pancreatic cancer cells

AB  - Existing therapies for the treatment of pancreatic cancer are insufficiently effective and accompanied by a large number of side effects. Ruthenium complexes have shown promising antitumor properties in the previous studies 1,2 . Thus, in this investigation, anticancer effects of cis-dichlorobis (2,2'-bipyridyl-4,4'dicarboxylic acid)ruthenium(II) (Ru(II) complex) were evaluated using human pancreatic carcinoma cell lines MIA PaCa-2 and PANC-1 in vitro. Cell viability estimated with SRB assay showed significant antitumor activity of Ru(II) complex on MIA PaCa-2 (~55% of control) 48 and 72 h after treatment. On the other hand, PANC-1 cell viability was decreased only 72 h after treatment with the highest concentration of Ru(II) complex (~70% of control). Seven days after the treatment, analysis of cell survival using clonogenic assay showed a significant decrease in cell growth in both cell lines. Ru(II) complex also caused G 1 cell cycle arrest of ~13% in both cell lines. The highest percentage of apoptotic MIA PaCa-2 cells was obtained 48 h after treatment. In addition, the intracellular level of reactive oxygen species (ROS) was significantly increased, whereas cell migration was reduced in both cell lines. Summarized, Ru(II)complex demonstrates antitumor properties mediated by increased oxidative stress and also implies the antimetastatic potential, which deserves further study.

AB  - S and N-doped carbon dots (S-CDs and N-CDs) and their cisplatin (cis-Pt) derivatives. (S-CDs@cis-Pt and N-CDs@cis-Pt) were tested on two ovarian cancer cell lines: A2780 and A2780 cells resistant to cis-Pt (A2780R). Several spectroscopic techniques were employed to check S-CDs@cis-Pt and N-CDs@cis-Pt: solid- and solution-state nuclear magnetic resonance, matrix-assisted laser desorption, ionization time-of-flight mass spectrometry, and X-ray photoelectron spectroscopy. In addition, synchrotron-based Fourier Transformed Infrared spectro-microscopy was used to evaluate the biochemical changes in cells after treatment with cis-Pt, S-CDs, N-CDs, or S-CDs@cis-Pt and N-CDs@cis-Pt, respectively. Computational chemistry was applied to establish the model for the most stable bond between S-CDs and N-CDs and cis-Pt. The results revealed the successful modification of S-CDs and N-CDs with cis-Pt and the formation of a stable composite system that can be used for drug delivery to cancer cells and likewise to overcome acquired cis-Pt resistance. Nanoparticle treatment of A2780 and A2780R cells led to the changes in their structure of lipids, proteins, and nucleic acids depending on the treatment. The results showed the S-CDs@cis-Pt and N-CDs@cis-Pt might be used in the combination with cis-Pt to treat the adenocarcinoma, thus having a potential to be further developed as drug delivery systems.

AB  - Metal acetate complexes are highly interesting in designing and synthesizing metal–organic frameworks, which have potential applications in gas storage, molecular sieves, ion exchange, and catalysis. Cadmium is well recognized as an industrial pollutant and a food contaminant that severely affects human health. For these reasons, the detection and analysis of Cd are of great interest. A major challenge in studying cadmium compounds' structure is that Cd2+ can have a wide range of coordination numbers and diverse geometries. We applied LDI to detect and characterize the Cd-related clusters and MALDI approach to investigate the Cd(OAc)2x2H2O reactivity and detectability with the five different organic matrices. The results revealed that in the solid-state Cd(OAc)2x2H2O tend to form coordination polymers, and in the presence of coordinating ligands (matrix molecules) form smaller complexes. In addition, the correlation between Cd-related LOD and matrix features was observed and discussed, indicating that the LODs of this analyte strongly depend on the optical absorption of the matrix-analyte adduct.

Blue light-enhanced cytotoxic activity of nitrogen-doped TiO2 nanoparticles in human cervical cancer cells

AB  - We have studied the cytotoxicity and phototoxicity of nitrogen-doped TiO2 (N-TiO2) nanoparticles (NPs) of the anatase crystal form for a possible human cervical (HeLa) cancer therapy. TiO2 is biocompatible with living structures (a) and its electrons in the valence band can be excited under ultraviolet (UV) light irradiation to produce reactive oxygen species and kill cancer cells. On the other hand, UV light is harmful and its penetration depth is short, which hinders its applications as a photosensitizer. Thus, we have chosen doping of TiO2 with nitrogen, since it was already shown that N-doping shifts the absorbance towards the visible region and enables photoactivation of the TiO2 with visible light, whereas the biocompatibility is preserved. (b) To assess the biocompatibility of N-TiO2 with the cancer cells, we have treated the HeLa cell line with three different concentrations of N-TiO2 NPs suspensions. After 24h of incubation in the dark, we have observed significant cytotoxicity in samples treated with concentration of 100 μg mL-1 of N-TiO2 NPs, whereas no significant cytotoxicity of the concentrations ≤50 μg mL-1 was detected. Hence, we have selected the treatment of 50 μg mL-1 of the N-TiO2 NPs for phototoxicity and cell imaging. Given that the N-TiO2 absorbs in the region of 400-450 nm, we have examined the viability of the HeLa cancer cell line upon the N-TiO2 treatment followed by continuous blue light irradiation (420 nm, 10 μW) for 15 minutes. The survival fraction of the cells treated with N-TiO2 NPs decreased significantly upon blue light irradiation. The intracellular distribution of N-TiO2 NPs was examined by laser scanning confocal microscopy. Most of the cells survived the treatment after 24h, and the agglomerates of N-TiO2 NPs were distributed mainly inside the cancer cells. The results of viability in the dark show that N-TiO2 NPs are not toxic for HeLa cells in the lower concentrations, and the evidence of cellular uptake points towards their biocompatibility. Furthermore, we have shown that it is feasible to induce cells’ phototoxicity upon treatment with N-TiO2 NPs and blue light irradiation.

AB  - In this study, C-doped TiO2 nanoparticles (C-TiO2) were prepared and tested as a photosensitizer for visible-light-driven photodynamic therapy against cervical cancer cells (HeLa). X-ray diffraction and Transmission Electron Microscopy confirmed the anatase form of nanoparticles, spherical shape, and size distribution from 5 to 15 nm. Ultraviolet–visible light spectroscopy showed that C doping of TiO2 enhances the optical absorption in the visible light range caused by a bandgap narrowing. The photo-cytotoxic activity of C-TiO2 was investigated in vitro against HeLa cells. The lack of dark cytotoxicity indicates good biocompatibility of C-TiO2. In contrast, a combination with blue light significantly reduced the survival of HeLa cells: illumination only decreased cell viability by 30% (15 min of illumination, 120 µW power), and 60% when HeLa cells were preincubated with C-TiO2. We have also confirmed blue light-induced C-TiO2-catalyzed generation of reactive oxygen species in vitro and intracellularly. Oxidative stress triggered by C-TiO2/blue light was the leading cause of HeLa cell death. Fluorescent labeling of treated HeLa cells showed distinct morphological changes after the C-TiO2/blue light treatment. Unlike blue light illumination, which caused the appearance of large necrotic cells with deformed nuclei, cytoplasm swelling, and membrane blebbing, a combination of C-TiO2/blue light leads to controlled cell death, thus providing a better outcome of local anticancer therapy.

Antitumor effect of Ru(II) complex on A375 and HeLa cell growth, migration and adhesion ability

AB  - Platinum-based complexes represent the mainstay of treatment for various cancer types. However, their usage is often restricted by numerous side effects or intrinsic and acquired resistance. Therefore, significant research efforts have focused on developing therapeutics based on other transition metals, such as ruthenium 1,2. In this study, effects of transition metal complex, cis-dichlorobis (2,2ʹ-bipyridyl-4,4ʹ-dicarboxylic acid)ruthenium(II) (Ru(II) complex) were analyzed on A375 human melanoma and HeLa cell growth, adhesion ability andmigration. Cell viability assay indicated significant antitumor activity of Ru(II) complex on A375 (~60% of control) up to 72 h after treatment, but not on HeLa cells. However, analysis by clonogenic assay showed that growth of both cell lines was decreased 7 days after treatment. Growth inhibition was followed by G1 phase cell cycle arrest (5–10% G1 increase for A375 and 5–8% for HeLa cells compared to control). Moreover, Ru(II) complex increased adhesivity of A375 and HeLa cells by 11 and 16 % respectively and decreased cell migration, as shown by scratch assay. The obtained results indicate that the analyzed Ru(II) complex is a promising metallodrug, as itinduced growth inhibition of A375 and HeLa cells through induction of G1 arrest and decreased metastatic potential of these cells through the increase ofadhesivity and decrease of cell migration.

The results showed that both CDs are sensitive to the cholesterol and can be used either alone or in a binary system with 2,5-dihydroxybenzoic acid (DHB) to enhance the detection process. It was found that both COOH and NH2 groups on CDs surface contributed to the enhancement in the cholesterol detection by MALDI mass spectrometry in the presence of inorganic cations. Nevertheless, in the presence of NaCl, N-CDs led to a better reproducibility of results. It was due to the coexistence of positive and negative charge on N-CDs surface that led to a homogeneous analyte/substrate distribution, which is an important detection parameter. The enhancing effect of carbon dots was linked to a negative Gibbs energy of the complex formation between CDs, Na+, cholesterol and DHB, and it was supported by theoretical calculations. Moreover, upon the addition of CDs/N-CDs, such features as a low ionization potential, vertical excitation, dipole moment and oscillator strength positively affected the cholesterol detection by MALDI in the presence of Na+. © 2021 Elsevier Inc.

AB  - This paper outlines the production of an inorganic polymer/geopolymer using a metakaolin by an environmental friendly, energy saving, clean technology to conserve natural environment and resources. The influence of alkali activation, i.e. different concentration of NaOH as a component of alkali activator mixture on the process of geopolymerization of metakaolin is investigated. Also, process of aging time of geopolymer is followed by several analytical methods. The structure of metakaolin and metakaolin based geopolymers and their physicochemical properties were studied using X-ray diffraction (XRD), Fourier transformation infrared spectroscopy (FTIR) and after 28 days scanning electron microscopy (SEM) with energy dispersive spectroscopy (EDS) was applied for the surface characterization of the samples. A shift of the Si-O or Si-O-X (X=Al, Si, Na...) bands as the molarity of activator increasing during geopolymerization process was observed by FTIR. Mass spectra of geopolymers were characterized by MALDI TOF mass spectrometer. Structural reorganization of geopolymer samples occurs during the curing/aging in accordance with a geopolymerization mechanism.

AB  - In this work, the performances of two ionic liquid matrices (ILMs) with the same ammonium counterpart for mass spectrometric analysis of the insoluble and soluble sucralfate were compared. Matrix-assisted laser desorption/ionization mass spectrometry (MALDI MS) was performed assisted by the butylammonium salts of α-cyano-hydroxycinnamic acid (CHCAB) and 2,5-dihydroxybenzoic acid (DHBB). CHCAB has a higher IE than DHBB, but better optical properties. CHCAB is more suitable for the analysis of sucralfate, although molecular ions of both compounds were detectable only with low intensities. Thus, optical properties of ILMs are crucial to enhance the sensitivity of MALDI MS detection of polysulfated oligosaccharides. © 2019, © 2019 Taylor & Francis Group, LLC.

On the other hand, there is great concern about toxicity in the dark of TiO2 nanoparticles in general. In this work, we have investigated the biocompatibility of TiO2 PNSs of the anatase crystal form (length between 100 and 300 nm and width 50 nm) in the dark with immune cells and light-induced cytotoxicity on several cancer cell lines. The effects of the treatment of different cell lines with several concentrations of TiO2 PNSs suspensions showed the specifics of cells’ viability and the intracellular localization. The results of in vitro studies obtained by cytotoxicity assays adjusted to individual cell lines’ metabolism point towards the biocompatibility of TiO2 PNSs at low and moderate concentrations in the dark, which neither kill the cells, nor induce activation of the immune system cells. Laser scanning confocal microscopy revealed that PNSs are taken up by cells, and insight into the intracellular distribution was obtained in this study.

AB  - Objects of the present study are improved fullerene C60 drug carrier properties trough encapsulation by microbial polysaccharides, levan (LEV), pullulan (PUL), and their hydrophobized cholesterol-derivatives (CHL and CHP), that show better interaction with cancer cells. The zeta potential, polydispersity index, and the diameter of particles were determined, and their cytotoxicity against three cancer cell lines were tested. Biochemical changes in HeLa cells are analyzed by synchrotron radiation (SR) FTIR spectro-microscopy combined with the principal component analysis (PCA). The most significant changes occur in HeLa cells treated with LEV-C60 and correspond to the changes in the protein region, i.e. Amide I band, and the changes in the structure of lipid bodies and membrane fluidity are evident. The highest cytotoxicity was also induced by LEV-C60. In HeLa cells, cytotoxicity could not be strictly associated with biochemical changes in lipids, proteins and nucleic acids, but these findings are significant contribution to the study of the mechanism of interaction of C60-based nanoparticles with cellular biomolecules. In conclusion, LEV, PUL, CHL, and CHP enhanced fullerene C60 potential to be used as target drug delivery system with the ability to induce specific intracellular changes in HeLa cancer cells.

AB  - Rationale: Changes in lipid composition might be associated with the onset and progression of various neurodegenerative diseases. Herein, we investigated the changes in the plasma phosphatidylcholine (PC)/lysophosphatidylcholine (LPC) ratios in patients with Parkinson's disease (PD) in comparison with healthy subjects and their correlation with clinico-pathological features. Methods: The study included 10 controls and 25 patients with PD. All patients were assigned to groups based on clinico-pathological characteristics (gender, age at examination, duration of disease and Hoehn and Yahr (H&Y) stage). The analysis of the PC/LPC intensity ratios in plasma lipid extracts was performed using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. Results: PD patients exhibited an increased PC/LPC intensity ratio in comparison with the control group of healthy subjects. Furthermore, the investigated ratio was shown to be correlated with clinico-pathological parameters, in particular with H&Y stage and disease duration. The PC/LPC intensity ratio in plasma samples of PD patients was found to be elevated in all examined H&Y stages and throughout the disease duration. Conclusions: To our knowledge, this is the first study examining the PC/LPC ratios in plasma of patients with PD and illustrating their correlation with clinico-pathological features. Although the presented results may be considered as preliminary due to the limited number of participants, the observed alterations of PC/LPC ratios in plasma might be a first step in the characterization of plasma lipid changes in PD patients and an indicator of lipid reconfiguration. © 2019 John Wiley & Sons, Ltd.

AB  - There has been a growing demand for development and improvement of cancer treatment in order to overcome disadvantages and limitations of traditional chemotherapy such as resistance, toxicity, and activity against a small number of tumor types. Photodynamic therapy is a rapidly developing cancer treatment that utilizes the combination of photoactive drug and/or carrier and light as an external stimulus to destroy tumors, achieve a maximum concentration of the drug on tumor site, reduce side effects of drug onto the healthy tissue, enable dosage control, and more effective treatment which could improve outcome of therapy [1]. We investigated possibility to use ТiО2 nanoparticles as a carrier for potential antitumor drug [Ru(II)(dcbpy)2Cl2], cis-dichlorobis (2, 2'-bipyridyl-4, 4'-dicarboxylicacid) ruthenium(II). Nanocomposite system has been formed by binding of ruthenium complex to the ТiО2 nanoparticles. Components of the system are selected according to their preferences, such as biocompatibility, photo activity, and easy surface modification for ТiО2, and less toxicity, significant activity against cancer metastases, and activity on tumors that were resistant to a variety of standard chemotherapeutic agents for Ru-complexes. Both components of the system are also photoactive, therefore potential for system manipulation and use in photodinamic therapy has been investigated by irradiation with UV and visible light. Additionally, the feasibility of using this system as a light-triggered drug delivery system was shown on amelanotic melanoma cancer line. The experimental results revealed a potential of nanocomposite system for long-term constant release of complex which is suitable for clinical practice. The further investigation of nanocomposite system indicated that it exhibited UV and red light susceptible drug release behavior [2]. More precisely, the system demonstrated slower complex release upon visible and increased release rate upon UV light illumination which was also in good correlation with the light–dependent cytotoxicity of the system demonstrated on amelanotic melanoma cancer line. The melanoma cancer cell death is enhanced by UV and reduced by red light in the presence of investigated nanocomposite system [3]. All obtained results suggested that nanocomposite system may have a potential use as a light sensitive drug delivery system in photodynamic therapy which could significantly contribute to development of more efficient and less invasive therapy of melanoma which is highly aggressive and deadliest form of skin cancer.

AB  - Abstract: In this study, we have synthesized a series of dinuclear and trinuclear gold(III) complexes of the general formula [Au2(N–N)Cl6] (1–3) for dinuclear and [Au3(N–N)2Cl8]+ (4–6) for trinuclear compounds, respectively, in which N–N is a bidentate ligand (1,4-diaminobutane; 1,6-diaminohexane or 1,8-diaminooctane). These complexes were characterized by elemental analysis, molar conductivity, and spectroscopic techniques (IR, UV–Vis, 1H NMR, ESI–MS). We performed DFT calculations to get insight into the geometry of the studies complexes. DNA-binding studies were performed by UV–Vis spectrophotometry and fluorescence spectroscopy. The results of competitive reactions between gold(III) complexes and ethidium bromide (EB) towards DNA have shown that selected complexes can displace EB from DNA-EB adduct. In addition, these experiments confirm that polynuclear gold(III) complexes interact with DNA covalently or via intercalation. Furthermore, high values of binding constants of gold(III) complexes towards bovine serum albumin (BSA) protein indicate good binding affinity. In addition, redox stability of complexes in the presence of DNA/BSA was confirmed by cyclic voltammetry. Results of the interactions between gold(III) complexes with DNA/BSA were discussed in reference to molecular docking data obtain by Molegro virtual docker. The cytotoxic activity of synthesized gold(III) complexes was evaluated on human breast cancer cell line (MDA-MB-231), human colorectal cancer cell line (HCT-116), and normal human lung fibroblast cell line (MRC-5). All complexes dose-dependently reduced cancer and normal cells viabilities, with significant cytotoxic effects (IC50 < 25 μM) for trinuclear gold(III) complexes (4, 5) on HCT-116 cells. In addition, redox stability of complexes in the presence of DNA/BSA was confirmed by cyclic voltammetry. Results of the interactions between gold(III) complexes with DNA/BSA were discussed in reference to molecular docking data obtain by Molegro virtual docker. The cytotoxic activity of synthesized gold(III) complexes was evaluated on human breast cancer cell line (MDA-MB-231), human colorectal cancer cell line (HCT-116), and normal human lung fibroblast cell line (MRC-5). All complexes dose-dependently reduced cancer and normal cells viabilities, with significant cytotoxic effects (IC50 < 25 μM) for trinuclear gold(III) complexes (4, 5) on HCT-116 cells. Positive and negative nano-electrospray mass spectrometry of ruthenated serum albumin supported by docking studies: an integrated approach towards defining metallodrug binding sites on proteins

AB  - Binding of three ruthenium(ii) compounds of general formula mer-[Ru(L3)(N-N)X][Y] (where L3 = 4-chloro-2,2:6,2-terpyridine (Cl-tpy); N-N = 1,2-diaminoethane (en), 1,2-diaminocyclohexane (dach) or 2,2-bipyridine (bipy); X = Cl; Y = Cl) to human serum albumin (HSA) has been investigated by nano-LC/nano-ESI MS and docking studies. A bottom-up proteomics approach has been applied for the structural characterization of metallated proteins and the data were analyzed in both the positive and negative ion mode. The negative ion mode was achieved after the post-column addition of an isopropanol solution of formaldehyde that enabled sample ionization at micro-flow rates. The negative ion mode MS has been proved to be beneficial for the analysis of binding sites on ruthenated protein in terms of ion charge reduction and consequent simplification of target sequence identification based on isotopic differences between ruthenated and non-ruthenated peptides. Moreover, the negative ion mode ESI MS shows the advantage of singly charged ion formation and, unlike MALDI MS, it does not cause complete ligand fragmentation, merging the benefits of each method into a single experiment. Six target sequences were identified for the binding of en and dach compounds, and four sequences for the binding of bipy. All compounds have been found to bind histidine and one aspartate residue. Docking studies showed that the identified sequences are the constituents of five distinct binding sites for en and dach, or two sites for the bipy complex. The selection of binding sites seems to be dependent on the chelate ligand and the form of the complex prior or after hydrolysis of the leaving chloride ligand.

AB  - Photodynamic therapy implies a combined use of a photosensitizing medicament and low-intensity light to cause selective damage to the target tissue—tumor. As potential medicament, we use Ru(II)(dcbpy)2Cl2complex, and in order to achieve better photosensitization properties, the Ru complex was attached to the nano carrier—TiO2nanoparticles. Additionally, this nanocomposite system was encapsulated in the phospholipid vesicles, which could be classified as small unilamellar vesicles, based on the technique of production. The complex-release tests were performed under light illumination, at pH 5, characteristic for tumor cells` interior and compared with the release pattern at pH 7, characteristic for the serum, i.e. physiological solution.
