The influence of poly(N-isopropylmethacrylamide) hydrogels crosslinking on their swelling capacity

AB  - Poly(N-isopropylmethacrylamide) has significant potential for designing thermosensitive new materials. It is similar with well-known poly(N-isopropylacrylamide) hydrogels. The influence of the α-methylation on the each monomer unit in the main chain exhibits weaker intermolecular interactions between the amide group. The method of thermally-initiated free radical polymerization was applied to synthesize a series of poly(N-isopropylmethacrylamide) hydrogels with different cross-linker amount. The micrographs, obtained by the scanning electron microscopy method, indicated a semi-homogeneous crosslinked network of these hydrogels which can be classified as macroporous. The swelling behavior at room temperature of these negatively thermosensitive hydrogel exhibit a little higher swelling degree in acidic media (at pH 2.0 swelling degree was about 21) in relation to the weakly basic media (at pH 7.4 swelling degree was about 18). The poly(N-isopropylmethacrylamide) hydrogel topology showed much free space within the crosslinked polymer chains in the swollen state and they could be use as absorbers of water, diluents, dissolvent, solutions, or as carriers for active substances.

AB  - Poly(N-isopropylmethacrylamide) (p(NiPMAm)) is one of the lesser known homopolymers that has significant potential for designing new “intelligent” materials. The aims of this work were the synthesis a series of cross-linked p(NiPMAm) hydrogels by the free radical polymerization method and the application of gamma-ray radiation for additional cross-linking. The synthesized p(NiPMAm) hydrogels were structurally characterized by Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). The amount of unreacted monomers was analyzed using high pressure liquid chromatography (HPLC) to evaluate conversion of monomers into polymers. The swelling behavior was monitored in dependence of pH and temperature changes. The previous aim of gamma-ray radiation was the further the cross-linkage of the obtained hydrogel sample in the gelatinous, paste-like state, but the gamma-ray radiation caused decomposition. After absorbing irradiation doses, they transformed into the liquid phase. The results obtained by the gel permeation chromatography (GPC) method indicated that only oligomers and monomers were present in the irradiated liquid material, without molecules with a higher average molar mass, i.e., that the decomposition of the hydrogels occurred. Additionally, the irradiated liquid material was analyzed using the static headspace gas chromatography mass spectrometry (HSS-GC/MS) and gas chromatography/flame ionization detection (HSS-GC/FID) methods. The presence of unchanged initiator molecule and a dominant amount of four new molecules that were different from homopolymers and the reactant (monomer and cross-linker) were determined.

Scanning electron microscopy analysis of changes of hydroxiapatite/poly-l-lactide with different molecular weight of PLLA after intraperitoneal implantation

AB  - Implantation of a biomaterial is one of the important trends in solving the problem of bone tissue loss. Calcium hydroxiapatite (HAp), as the most representative bone component is a serious candidate for such implantations. The synthetic polymer poly-L-lactide (PLLA) in HAp/PLLA is often used as a polymeric material, with a role in the substitution of bone tissue collagen fibers. Fibers of PLLA may strengthen HAp and its good bioresorption provides space for tissue remodeling. Differences in porosity, microstructure, compressive consistency as well as bioresorbility of HAp/PLLA may be achieved by using PLLA with different molecular weights. In this study HAp/PLLA composites with PLLA of different molecular weights (50,000; 160,000 and 430,000) were implanted in mouse peritoneum in order to examine the influence of the molecular weight of PLLA on morphology changes. Microstructural changes of biomaterial (HAp/PLLA) surface were analyzed one week, three weeks and four months after their implantation using Scanning Electron Microscopy. The results showed a significant difference in tissue reactions on the applied biocomposites, depending on their molecular weight. The most intense proliferation of cells was induced by HAp/PLLA 50,000 compared to HAp/PLLA 430,000 and HAp/PLLA 160,000. In the vicinity of HAp/PLLA 430,000 abundant erythrocytes were observed. The differences in biological reactions on the examined biocomposites are significant for their practical applications. HAp/PLLA composite biomaterials of different types and resorption rates require specific designing and programming to become suitable for particular purposes in an organism.

AB  - Recent studies have shown that the behavior of nanoparticles in in vivo conditions is not solely dependant on their physical properties, such are size and shape. The surface of nanoparticles in biological fluids interacts with biomolecules such are proteins, adsorbs these molecules, and leads to formation of nanoparticle-biomoelcular complex known as “protein corona”. This protein corona changes the properties of nanoparticles and their behavior in vivo. In this work three types of nanomaterial based on Hap and polymers were incubated in rats plasma, and the identification and quantification of proteins in protein corona that had formed around these nanoparticles, was done by means of electrophoresis and mass spectrophotometery.

AB  - The influence of glucocorticoids on the composition and mineral/organic content of the mandible in tested animals after recovery and healing phase was investigated in this work. The results of FTIR analysis demonstrated that bone tissue composition was changed after glucocorticoid treatment. The increase of calcium, magnesium, phosphorus content and mineral part of bones was statistically significant in recovery phase and in treatment phase that included calcitonin and thymus extract. Some changes also happened in the organic part of the matrix, as indicated by intensity changes for already present IR bands and the appearance of new IR bands in the region 3500–1300 cm−1.

AB  - Indications exist that paramagnetic calcium phosphates may be able to promote regeneration of bone faster than their regular, diamagnetic counterparts. In this study, analyzed was the influence of paramagnetic cobalt-substituted hydroxyapatite nanoparticles on osteoporotic alveolar bone regeneration in rats. Simultaneously, biocompatibility of the material was tested in vitro, on osteoblastic MC3T3-E1 and epithelial Caco-2 cells in culture. The material was shown to be biocompatible and nontoxic when added to epithelial monolayers in vitro, while it caused a substantial decrease in the cell viability as well as deformation of the cytoskeleton and cell morphology when incubated with the osteoblastic cells. In the course of 6 months after the implantation of the material containing different amounts of cobalt, ranging from 5 to 12 wt%, in the osteoporotic alveolar bone of the lower jaw, the following parameters were investigated: histopathological parameters, alkaline phosphatase and alveolar bone density. The best result in terms of osteoporotic bone tissue regeneration was observed for hydroxyapatite nanoparticles with the largest content of cobalt ions. The histological analysis showed a high level of reparatory ability of the nanoparticulate material implanted in the bone defect, paralleled by a corresponding increase in the alveolar bone density. The combined effect of growth factors from autologous plasma admixed to cobalt-substituted hydroxyapatite was furthermore shown to have a crucial effect on the augmented osteoporotic bone regeneration upon the implantation of the biomaterial investigated in this study.

AB  - In this study we observed the changes in surface of nanoparticles of two different nanomaterials, cobalt-substituted hydroxyapatite (CoHAp) and calcium phosphate/poly-(DLlactide-co-glycolide) (CP/PLGA) that occurred under simulated physiological conditions in vitro. Both nanomaterials were incubated in an appropriate volume of Dulbecco's Modified Eagle Medium (DMEM) for 3 days at 37 0C and then the medium was removed and materials were dried. Surface changes of nanoparticles were studied by scanning electron microscopy (SEM). Results of SEM analysis show that changes in the size and shape of the nanoparticles took place. These changes can be explained by interactions of nanomaterials with DMEM. In the case of CP/PLGA it was probably the result of polymer degradation and changes in CoHAp nanoparticles were probably the result of ion exchange.

AB  - The aim of this study was to investigate the osteogenic potential of freshly isolated adipose tissue-derived mesenchymal stromal/stem cells (ASCs) in bone regeneration of rabbit calvaria defects. ASCs were applied together with calcium phosphate-poly-DL-lactide-co-glycolide (CPPLGA) nanoparticles that are mixed in an appropriately prepared blood clot (BC). Parallelly were analysed defects filled with fragmented adipose tissue (FAT) instead of ASCs, with CP-PLGA in BC, and only with nanoparticles. Bone density in defects was measured after I, III and VI weeks, and histological examination was done after IV and VIII weeks after filling defects. Approach to assisted osteoregeneration which is based on the use of fresh non-induced ASCs has proven to be promising due to the favorable effect on bone regeneration and simplicity of their application.

AB  - The lack of bone in the jaw tegmenta inflicts major problem and leads to an inability to adequately treat stomatoprosthetic patients. If the bone tissue damage is minor, the balanced activities of osteoblasts and bone osteoclasts can repair it independently. If the damage is bigger it is necessary to support the biological potential to repair the bone, and for that reason nanoparticlebiomaterial Ca / Co-HAp was used in this study. The research was done on rats with uniform anatomical and physiological characteristics. Assessment of repair and consolidation of the jaw bone tegmenta was performed by istomorphometric and SEM analysis. The best results were obtained in the experimental group of animals where the Ca / Co-Hap was mixed with autologousplasma. Following the implementation of the above mentioned nanocomposites, a significant formation of new bone was evident on the SEM analysis, as well as the rising of histomorphometric parameters of bone formation, which indicates that the Ca / Co-HAp nanocomposite is the material of choice for the rapid regeneration and repair of bone jaw tegmenta.

AB  - We have examined the effect of extracts of the calcium phosphate/poly(DL-lactide-co-glycolide) (CP/PLGA) and cobalt-exchanged hydroxyapatite (CoHAp), on the viability of Saos2 osteoblastlike cells. Extracts were prepared by incubation for 3 days at 37°C in the cultivation medium. Conductivity and pH value of extracts were measured before viability assay. Cell viability was estimated by MTT test performed after 24 h incubation of cells with various concentrations of extracts. Extract of CP/PLGA acted more cytotoxic on osteoblasts than the extract of CoHAp. This difference in the effect of examined extracts can be explained by their different pH value and conductivity.

AB  - Huge range of tested biomaterials in recent decades has emerged as an ideal scaffold for cell growth, but few have demonstrated clinical efficacy. Among them, synthetic hydroxyapatite (HAp, Ca10(PO4)6(OH)2) is the most promising because of its biocompatibility, bioactivity, and osteoconductivity. Biocompatibility represents the primary concern for any material to be used as a substitute for natural tissue. Hydroxyapatite particles interact with numerous cellular systems in vivo, and some of these interactions may lead to cell damage and to stimulate platelet activation, coagulation and thrombus formation. The aim of this work was to examine the hemocompatibility of nanocalcium hydroxyapatite substituted with 5% and 12% cobalt (Ca /CoHAp) and hydroxyapatite/polylactidcoglicolid (HAp / PLGA) in relation to pure HAp by testing their hemolytic activities. The results show the discrepancy in hemolytic activity of implanted matherials. The degree of crystallinity of samples had a more dominant influence on hemolysis than the percentage of substituted cobalt. Hemolysis ratios of the nanocalcium hydroxyapatite substituted with cobalt samples were below 3%, indicating good blood compatibility and that they are promising for medical application.

AB  - Bone loss during the systemic osteoporosis has an important role in dentistry and medicine. The aim of the study was the application of bioactive micro and nanocomposite materials, alone, and in combination with autologous plasma in osteoporotic jaw bones of rats with artificially induced osteoporosis. The effect of these composites was measured by histomorphometric and atomic absorption spectrophotometric analysis. According to the best obtained results in regeneration and recovery of the resorbed alveolar bone, it can be concluded that nanocomposite combined with autologous plasma may be the material of choice to replace the osteoporotic damaged jaw bone.

AB  - The aim of this study was to compare the effects of two types of nanomaterials, cobalt-exchanged hydroxyapatite (CoHAp) and calcium phosphate/poly-(DL-lactide-co-glycolide) (CP/PLGA), on the viability of Saos-2 osteoblast-like cells, using MTT test. We have examined the suspension of this materials in the following concentrations: 1.6, 8, 40, 200, 1000 and 2500 μg/ml, as well as extracts in concentration range from 2,5% to 100%. Both materials showed cytotoxic effect at higher concentrations of suspension and extract, respectively, but they were not cytotoxic at lower concentrations. CP/PLGA acted stronger cytotoxic compared to CoHAp, regardless of whether it is examined suspension or extract. CoHAp in small concentrations of suspension and extract acted slightly stimulatory on cells. This suggests that CoHAp may have advantage for use in the in vivo systems.

AB  - Simulated body fluid (SBF) is an artificial fluid which has ionic composition and ionic concentration similar to human blood plasma. Purpose: This paper compares the interaction between the nanomaterial containing calcium phosphate/poly-dl-lactide-co-glycolide (N-CP/PLGA) and SBF, in order to investigate whether and to what extent inorganic ionic composition of human blood plasma leads to the aforementioned changes in the material. Methods: N-CP/PLGA was incubated for 1, 2, 3, and 5 weeks in SBF. The surface of the material was analyzed on SEM-EDS and FTIR spectrometer, while SBF was subjected to pH and electrical conductivity measurement. Results: Our results indicate that dissolution of the polymer component of the material N-CP/PLGA and precipitation of the material similar to hydroxyapatite on its surface are based on the morphologic changes seen in this material. Conclusions: The mechanism of the apatite formation on the bioceramic surface was intensively studied and was considered crucial in designing the new biomaterials. The results obtained in this work indicate that N-CP/PLGA may be a good candidate for application to bone regeneration.

Results: Our results indicate that dissolution of the polymer component of the material N-CP/PLGA and precipitation of the material similar to hydroxyapatite on its surface are based on the morphologic changes seen in this material. Conclusions: The mechanism of the apatite formation on the bioceramic surface was intensively studied and was considered crucial in designing the new biomaterials. The results obtained in this work indicate that N-CP/PLGA may be a good candidate for application to bone regeneration.",

AB  - In recent years, calcium hydroxyapatite (HAp) and its substituents are increasingly used in dentistry and medicine. The influence of nanoparticles Ca / Co-HAp in strengthening weak osteoporotic bone jaw tegmenta was tested in an experimental model. The study was conducted on Wistar soy rats, aged 6-8 weeks. The biomaterial was implanted in the osteoporoticly weakened mandible of these animals. The best results in the strengthening of the lower jaw bone tegmenta were achieved 24 weeks after implantation of hydroxyapatite nanoparticles in which the calcium ion was substituted with 12% of cobalt ions. Histochemical parameters of bone synthesis were in a statistically significant increase. SEM analysis showed a high degree of osteogenetic ability of nano-particulate material implanted in the bone defect.

AB  - The aim of this study was to examine the effects of nanoparticles of calcium phosphate/poly-(DL-lactide-co-glycolide) (NPs-CP/PLGA) on viability and growth of different cell lines in vitro. HeLa and MDCK cells were incubated with different concentrations of suspension and extract of NPs-CP/PLGA. Concentrations of suspension were in the range from 1.6 μg/ml to 5000 μg/ml and extract in the range from 2.5% to 50%. After incubation, MTT test was performed. Our results indicate that examined nanomaterial shows different effects depending on type of the cells, applied concentration of nanomaterial as well as whether it is examined suspension or extract.

AB  - One of the trends in the treatment of reduced bone density is in oral implantology through the use of new biomaterials with paramagnetic properties. In this in vivo study the effect of Ca /Co-HAp nanoparticles (calcium/cobalt-hydroxiapatite) in the regeneration of the reduced alveolar bone density in experimental animals was examined by analyzing both the biochemical and histochemical markers and through analysis of alveolar bone density. The research was carried out on female Westar rats, aged 6-8 weeks, whom have been implanted nanobiomaterial in reduced alveolar bone density. Good results in regeneration of reduced bone density were achieved in six weeks after implantation of the nanoparticles with paramagnetic properties. Biochemical markers of osteogenesis showed statistically significant rise. Histological analysis revealed high level reparatory skills of the biocomposite implanted in the bone defect, while X-ray analyses evidenced rise of bone density. Implantation of Ca/Co-Hap enables rapid new formation, thus becoming the material of choice for accelerated reduced bone density regeneration.
