AB  - In this work, thermal diffusivity of crystalline high-density polyethylene samples of various thickness, and prepared using different procedures, was evaluated by transmission gas-microphone frequency photoacoustics. The samples composition analysis and their degree of crystallinity were determined from the wide-angle X-ray diffraction, which confirmed that high-density polyethylene samples, obtained by slow and fast cooling, were equivalent in composition but with different degrees of crystallinity. Structural analysis, performed by differential scanning calorimetry, demonstrated that all of the used samples had different levels of crystallinity, depending not only on the preparing procedure, but also on sample thickness. Therefore, in order to evaluate the samples thermal diffusivity, it was necessary to modify standard photoacoustic fitting procedures (based on the normalization of photoacoustic amplitude and phase characteristics on two thickness levels) for the interpretation of photoacoustic measurements. The calculated values of thermal diffusivity were in the range of the expected literature values. Besides that, the obtained results indicate the unexpected correlation between the values of thermal diffusivity and thermal conductivity with the degree of crystallinity of the investigated geometrically thin samples. The results indicate the necessity of additional investigation of energy transport in macromolecular systems, as well as the possible employment of the photoacoustic techniques in order to clarify its mechanism.

AB  - Aim The anastomosis technique used following right-sided colonic resection is widely variable and may affect patient outcome. This study aimed to assess the association between leak and anastomosis technique (stapled vs handsewn). Method This was a prospective, multicentre, international audit including patients undergoing elective or emergency right hemicolectomy or ileo-caecal resection operations over a 2-month period in early 2015. The primary outcome measure was the presence of anastomotic leak within 30 days of surgery, determined using a prespecified definition. Mixed effects logistic regression models were used to assess the association between leak and anastomosis method, adjusting for patient, disease and operative cofactors, with centre included as a random-effect variable. Results This study included 3208 patients, of whom 78.4% (n = 2515) underwent surgery for malignancy and 11.7% (n = 375) underwent surgery for Crohns disease. An anastomosis was performed in 94.8% (n = 3041) of patients, which was handsewn in 38.9% (n = 1183) and stapled in 61.1% (n = 1858). Patients undergoing hand-sewn anastomosis were more likely to be emergency admissions (20.5% handsewn vs 12.9% stapled) and to undergo open surgery (54.7% handsewn vs 36.6% stapled). The overall anastomotic leak rate was 8.1% (245/3041), which was similar following handsewn (7.4%) and stapled (8.5%) techniques (P = 0.3). After adjustment for cofactors, the odds of a leak were higher for stapled anastomosis (adjusted OR = 1.43; 95% CI: 1.04-1.95; P = 0.03). Conclusion Despite being used in lower-risk patients, stapled anastomosis was associated with an increased anastomotic leak rate in this observational study. Further research is needed to define patient groups in whom a stapled anastomosis is safe.

CXCL16 in Vascular Pathology Research: from Macro Effects to microRNAs

AB  - Chemokines and their receptors have become significant factors in atherosclerosis research. CXCL16 is a multifunctional agent located on a separate locus to all other known chemokines and binds only to its unique receptor named CXCR6. As a scavenger receptor, adhesion molecule, and chemokine, it quickly became an interesting target in atherosclerosis research as all its functions have a role in vascular pathology. The investigation of the role of CXCL16 in atherosclerosis, although shown in in vitro studies, animal knockout models, and CXCL16 gene polymorphisms, haplotypes, and circulating levels, still shows puzzling results. Genetic and epigenetic studies have just scratched the surface of research necessary for a better assessment of the significance and perspective of this marker in plaque development and progression. In this review, we will summarize current knowledge about CXCL16 in atherosclerosis. Additionally, we will point out the importance of bioinformatics tools for the detection of potentially new CXCL16 regulatory networks through microRNA activity. This review aims to provide a better understanding of the underlying mechanisms, define more specific biomarkers, and discover new therapeutic targets.

AB  - The mechanism responsible for trafficking of monocyte-derived macrophages into kidney in the puromycin aminonucleoside model of nephrotic syndrome in rats (PAN-NS), and the significance of this infiltration, remain largely unknown. CXCL10, a chemokine secreted in many T helper type 1 (Th1) inflammatory diseases, exhibits important roles in trafficking of monocytes and activated T cells. We hypothesized that induction of circulating interferon (IFN)- and glomerular tumour necrosis factor (TNF)- during PAN-NS would stimulate the release of CXCL10 by podocytes, leading to infiltration of activated immune cells and greater glomerular injury. We found that serum IFN-, glomerular Cxcl10mRNA and intra- and peri-glomerular macrophage infiltration were induced strongly during the late acute phase of PAN-NS in Wistar rats, but not in nude (Foxn1(rnu/rnu)) rats lacking functional effector T lymphocytes. Wistar rats also developed significantly greater proteinuria than nude rats, which could be abolished by macrophage depletion. Stimulation of cultured podocytes with both IFN- and TNF- markedly induced the expression of Cxcl10mRNA and CXCL10 secretion. Together, these data support our hypothesis that increased circulating IFN- and glomerular TNF- induce synergistically the production and secretion of CXCL10 by podocytes, attracting activated macrophages into kidney tissue. The study also suggests that IFN-, secreted from Th1 lymphocytes, may prime proinflammatory macrophages that consequently aggravate renal injury.

AB  - This paper presents a model for indirect photoacoustic response that includes thermal memory effects. At low frequencies, the model reduces to the well-known thermal piston model of photoacoustic response given by Rosencweig and Gersho. AB  - Fructose-rich diets (FRD) cause cardiac insulin resistance manifested by impairment of Akt/endothelial NO synthase (eNOS) signalling. In contrast, oestradiol (E2) activates this signalling pathway in the heart. To study the ability of E2 to revert the detrimental effect of fructose on cardiac Akt/eNOS, female rats were subjected to a FRD and ovariectomy followed with or without E2 replacement. We also analysed the effects of the FRD and E2 on cardiac extracellular signal-regulated kinase (Erk 1/2) signalling related to their role in cardiac hypertrophy development. Expression of Akt, eNOS and Erk 1/2, as well as regulatory phosphorylations of these molecules were determined. The protein expression of cardiac Akt and eNOS was not affected by the diet or E2 treatment. However, the FRD was accompanied by a decrease in Akt phosphorylation at Ser(473) and Thr(308), and eNOS at Ser(1177), while the phosphorylation of eNOS at Thr(495) was increased. E2 replacement in ovariectomised fructose-fed rats caused a reversion of the diet effect on Akt and eNOS serine phosphorylation, but mostly had no effect on threonine phosphorylation of the molecules. The FRD and E2 treatment did not influence Erk 1/2 expression and phosphorylation and heart mass as well. The data show that E2 selectively suppress the negative effects of a FRD on Akt/eNOS signalling and probably point to the different effects of E2 on kinase/phosphatase pathways responsible for phosphorylation/dephosphorylation of Akt and eNOS. Furthermore, the results suggest that the heart of females in the reproductive period is partially protected against the damaging effects of increased fructose intake.

A comparison is made between the derived model and the classic one, which neglects the influence of thermal memory. It has been shown that, at modulation frequencies lower than a certain boundary frequency of the light source, these models tend to overlap, while at higher frequencies, noticeable differences occur. The boundary frequency depends on heat propagation velocity through the sample and its thickness. This observation limits the validity domain of previous models to a range lower than the boundary frequency, offering, at the same time, the possibility of obtaining thermal memory properties using PA effects at frequencies above it.

A generalized description of the distribution is proposed, including the effects of a finite heat propagation velocity and a finite, non-zero time of thermal relaxation (known as thermal memory effects in generalized heat conduction theory), and finally the generated thermoelastic bending is calculated by using both a thin solid-plate approximation and a decoupling system of thermoelastic equations. The comparison between this model and the classical one, which does not account for thermal memory influence, has been made. If the sample is thicker than the value of its minimal thermal diffusion length, the difference between the two models becomes insignificant. Otherwise, it has been shown that the two models tend to overlap at low and high modulation frequencies of the excitation light, while in the mid-frequency range, some deviations become more apparent and thermal memory properties of the sample must be taken into account. The suggested model enables evaluation of thermal memory properties for such a solid.

AB  - In this study, a model of thermoconducting components of the transmission photoacoustic response is analyzed, which includes thermal memory properties of both the examined solid sample and its fluid environment. It is shown that, at low modulation frequencies, lower than minimum among the inverse thermal relaxation time of air, the inverse thermal relaxation time of the sample, and the certain frequency at which the sample is thinner than one-quarter of the wavelength of time-varying temperature perturbation, thermal memory does not affect the photoacoustic response, while, at high frequencies, noticeable effects are observed. These results indicate the possibility of evaluating thermal memory properties of solid samples using classical, transmission, high frequency photoacoustic measurements.

AB  - Vascular endothelium is a key regulator of homeostasis. In physiological conditions it mediates vascular dilatation, prevents platelet adhesion, and inhibits thrombin generation. However, endothelial dysfunction caused by physical injury of the vascular wall, for example during balloon angioplasty, acute or chronic inflammation, such as in atherothrombosis, creates a proinflammatory environment which supports leukocyte transmigration toward inflammatory sites. At the same time, the dysfunction promotes thrombin generation, fibrin deposition, and coagulation. The serine protease thrombin plays a pivotal role in the coagulation cascade. However, thrombin is not only the key effector of coagulation cascade; it also plays a significant role in inflammatory diseases. It shows an array of effects on endothelial cells, vascular smooth muscle cells, monocytes, and platelets, all of which participate in the vascular pathophysiology such as atherothrombosis. Therefore, thrombin can be considered as an important modulatory molecule of vascular homeostasis. This review summarizes the existing evidence on the role of thrombin in vascular inflammation.

AB  - Vascular endothelium, as a key regulator of hemostasis, mediates vascular dilatation, prevents platelet adhesion, and inhibits thrombin generation. Endothelial dysfunction caused by acute or chronic inflammation, such as in atherosclerosis, creates a proinflammatory environment which supports leukocyte transmigration toward inflammatory sites, and at the same time promotes coagulation, thrombin generation, and fibrin deposition in an attempt to close the wound. Life-long persistent infection with human cytomegalovirus (HCMV) has been associated with atherosclerosis. In vivo studies have revealed that HCMV infection of the vessel wall affects various cells including monocytes/macrophages, smooth muscle cells (SMCs) and endothelial cells (ECs). HCMV-infected SMCs within vascular lesions display enhanced proliferation and impaired apoptosis, which contribute to intima-media thickening, plaque formation and restenosis. Monocytes play a central role in the process of viral dissemination, whereas ECs may represent a viral reservoir, maintaining persistent infection in HCMV-infected atherosclerotic patients following the primary infection. Persistent infection leads to dysfunction of ECs and activates proinflammatory signaling involving nuclear factor kappa B, specificity protein 1, and phosphatidylinositol 3-kinase, as well as expression of platelet-derived growth factor receptor. Activation of these pathways promotes enhanced proliferation and migration of monocytes and SMCs into the intima of the vascular wall as well as lipid accumulation and expansion of the atherosclerotic lesion. Moreover, HCMV infection induces enhanced expression of endothelial adhesion molecules and modifies the proteolytic balance in monocytes and macrophages. As a consequence, infected endothelium recruits naive monocytes from the blood stream, and the concomitant interaction between infected ECs and monocytes enables virus transfer to migrating monocytes. Endothelial damage promotes thrombin generation linking inflammation and coagulation. HCMV, in turn, enhances the thrombin generation. The virus carries on its surface the molecular machinery necessary to initiate thrombin generation, and in addition, may interact with the prothrombinase protein complex thereby facilitating thrombin generation. Thus, infection of endothelium may significantly increase the production of thrombin. This might not only contribute to thrombosis in patients with atherosclerosis, but might also induce thrombin-dependent proinflammatory cell activation. This review summarizes the existing evidence on the role of HCMV in vascular inflammation.

AB  - Vascular smooth muscle cells (VSMC) respond to arterial wall injury by intimal proliferation and play a key role in atherogenesis by proliferating and migrating excessively in response to repeated injury, such as hypertension and atherosclerosis. In contrast, fully differentiated, quiescent VSMC allow arterial vasodilatation and vasoconstriction. Exaggerated and uncontrolled VSMC proliferation appears therefore to be a common feature of both atherosclerosis and hypertension. Signal transduction pathways in eukaryotic cells integrate diverse extracellular signals, and regulate complex biological responses such as growth, differentiation and death. One group of proline-directed Ser/Thr protein kinases, the mitogen-activated protein kinases (MAPKs), plays a central role in these signalling pathways. Much attention has focused in recent years on subfamilies of MAPKs, the extracellular signal regulated kinases (ERKs). Here we overview the work on ERKs 1 to 2, emphasising when possible their biological activities in VSMC proliferation. It is clear from numerosus studies including our own, that ERK1/ERK2 pathway has an imoprtant role in VSMC proliferation induced by insulin (INS) and thrombin. Despite the physiological and pathophysiological importance of INS and thrombin, possible signal transduction pathways involved in INS and thrombin regulation of VSMCs proliferation remains poorly understood. Thus, this review examines recent findings in signalling mechanisms involved in INS and thrombin-triggered VSMCs proliferation with particular emphasis on ERK1/2 signalling pathways. Future investigations should now focus on the mechanisms of MAPK activation which might therefore represent a new mechanism involved in the antiproliferative effect revealed in this review.

The treatment considers a semiconductor elastic plate with isotropic and homogeneous plasma and elastic properties. The solution of the coupled system of plasma and elastic equations are given for a typical photothermal configuration including the carrier surface and volume recombination processes. The analysis of the plasma density and elastic fields shows that the coupling plasmaelastic effects show the attenuation and disperse phenomena.

AB  - The theoretical models of optically induced elastic bending for a semiconductor circular plate (clamped and simply supported) was derived including both plasmaelastic (PE) and thermoelastic (TE) wave influences. The PE and TE effects versus the modulation frequency of focused laser excitation were analyzed. Obtained results were compared with the optically excited elastic bending produced by homogeneous surface heat source and with experimental measurements.

Human cytomegalovirus increases HUVEC sensitivity to thrombin and modulates expression of thrombin receptors

AB  - Human cytomegalovirus (HCMV) establishes a life-long persistent infection. HCMV infection could be associated with chronic inflammatory diseases, such as cardiovascular disease and atherosclerosis. Here we observed that in HCMV (AD-169) pre-exposed human umbilical vein endothelial cells (HUVEC), thrombin-induced expression of IL-1 alpha and M-CSF is markedly enhanced compared to the un-exposed cells. Study of the expression of thrombin receptor genes in HUVEC showed that HCMV triggered a time- and concentration-dependent expression of the thrombin receptors PAR1, PAR3 and PAR4 at the mRNA level. Induction of PAR1 and PAR3 mRNA expression is due to transcriptional activation of their promoters as shown by gene reporter assay. Furthermore, the virus induced expression of PAR1 and PAR3 but not PAR4 proteins, as analyzed by Western immunoblotting. However, flow cytometric analysis revealed that only PAR3, expressed at very low level in control HUVEC, is induced at the surface during the exposure to the virus. Our data suggest that although exposure to HCMV induces a minor increase of cell-surface receptors expression, it does make endothelial cells more responsive to additional thrombin stimulation.

Here we observed that in HCMV (AD-169) pre-exposed human umbilical vein endothelial cells (HUVEC), thrombin-induced expression of IL-1 alpha and M-CSF is markedly enhanced compared to the un-exposed cells. Study of the expression of thrombin receptor genes in HUVEC showed that HCMV triggered a time- and concentration-dependent expression of the thrombin receptors PAR1, PAR3 and PAR4 at the mRNA level. Induction of PAR1 and PAR3 mRNA expression is due to transcriptional activation of their promoters as shown by gene reporter assay. Furthermore, the virus induced expression of PAR1 and PAR3 but not PAR4 proteins, as analyzed by Western immunoblotting. However, flow cytometric analysis revealed that only PAR3, expressed at very low level in control HUVEC, is induced at the surface during the exposure to the virus. Our data suggest that although exposure to HCMV induces a minor increase of cell-surface receptors expression, it does make endothelial cells more responsive to additional thrombin stimulation.",

Milling was performed in order to strengthen the copper matrix by grain size refinement and Al2O3 particles. Milling in air of prealloyed copper powder promoted formation of finely dispersed nano-sized Al2O3 particles by internal oxidation. On the other side, composite powders with commercial micro-sized Al2O3 particles were obtained by mechanical alloying. Following milling, powders were treated in hydrogen at 400 degrees C for 1h in order to eliminate copper oxides formed on their surface during milling. Hot pressing (800 degrees C for 3 h in argon at pressure of 35 MPa) was used for compaction of milled powders. Hot pressed composite compacts processed from 5 and 20 h milled powders were additionally subjected to high temperature exposure (800 degrees C for 1 and 5 h in argon) in order to examine their thermal stability. The results were discussed in terms of the effects of different size of starting powders, the grain size refinement and different size of Al2O3 particles on strengthening, thermal stability and electrical conductivity of copper-based composites.

AB  - We studied the effect of HCMV (AD-169) on the regulation of PAR1 reporter activity in HUVEC. In HUVEC, HCMV significantly induced activation of PAR1 reporter constructs carrying AP-2-like and two complexes of AP-2/Sp1-binding elements. Shorter constructs with no AP-2-like binding elements had lower activity upon virus inoculation. Moreover, activity of the shortest construct without binding elements was not significantly increased in virus-exposed cells. Furthermore, in HCMV-exposed cells, activity of the PARI reporter was significantly augmented upon p52/RelB transfection. Taken together, the data indicate that activity of the PAR1 promoter is highly dependent on AP-2/Sp1 binding elements and mediated by p52/RelB regulatory factors.

AB  - Background: Recently a high-resolution HLA and SNP map was defined and the analysis provided informative tag SNPs that capture much of the common variation in the MHC region. This concept enables detection of smaller number of SNPs, making it surrogate markers for haplotype associated with certain disease. The SNP rs3135388 was proposed as a tagging SNP for DRB1*1501/DQB1*0602 alleles, associated with MS. The aim of the study was to investigate the HLA rs3135388 genotypes in association with MS in patients from Serbia. Methods: Two hundred sixty nine consecutive patients from Serbia with relapse-remitting and secondary progressive MS were recruited for the study. Genomic DNA was isolated from peripheral blood cells. We designed the TaqMan assay for high-throughput genotyping of HLA rs3135388 on 7500 Real-Time PCR System. Results: We found significantly higher frequency of rs3135388 A allele carriers in MS patients compared to controls (p LT 0.001, chi(2)). In our population the carriers of one A allele had adjusted OR 2.09 (95% CI 1.41-3.09, p LT 0.001) for MS susceptibility. Conclusion: We assessed significant association of rs3135388 A allele carriership with MS in patients from Serbia. This HLA-DRB1*1501 surrogate marker is useful in association studies in MS. First, a general photothermal mathematical model for thermal gradient materials is presented. Then, we discuss the effects of inhomogeneous thermal properties in photothermal amplitude and phase spectra of coatings. Finally, a method for quantitative depth profiling that makes use of prior knowledge about the type of profile existing in a sample to reduce the instabilities associated with the mathematically ill conditioned task is demonstrated.

AB  - A study of ion beam modification of structural and electrical properties of TiN thin films is presented. The layers, were deposited by reactive ion sputtering on (100) Si substrates to a thickness of similar to 240 nm. After deposition the structures were implanted with argon ions at 120 keV, to the fluences from 1 x 10(15) - 1 x 10(16) ions/cm(2). Structural analysis of the samples was performed by cross-sectional transmission electron microscopy, x-ray diffraction and Rutherford backscattering spectrometry. It was found that the as-deposited layers have a columnar structure, individual columns stretching from the substrate to the surface and being a few tells of nanometers wide. Ion irradiation rearranges their crystalline structure, which remains polycrystalline, but the columns are broken, and nanocrystals of the same phase are formed.

AB  - Polycrystalline cobalt layers, deposited on Si(110) wafers via electron beam evaporation to a thickness of 55 nm, were irradiated at room temperature with 200 keV Xe ions to fluences of tip to 15 x 10(15) ions/cm(2). The atomic and magnetic force microscopy (AFM and MFM) were used to investigate the changes in the roughness and magnetic properties of the Co/Si bilayers. Ion beam induced structural changes were correlated with magnetic properties.

The influence of orientation on the radiation-induced crosslinking/oxidative behavior of different PEs

AB  - This paper studies the influence of orientation and structural peculiarities on the radiation-induced oxidation/crosslinking changes in different polyethylenes (PEs). For this reason, low density polyethylene (LDPE), linear low density polyethylene (LLDPE) and high density polyethylene (HDPE) were oriented via solid-state stretching at an elevated temperature. Later on, oriented samples were gamma-irradiated in air to various absorbed doses (up to 700 kGy). In order to investigate orientation- and radiation-induced changes in morphology, surface microstructures were analyzed by optical (OM) and scanning electron microscopy (SEM). Differential scanning calorimetry (DSC), IR spectroscopy and gel measurements were used to determine changes in crystallinity, oxidative degradation and degree of network formation, respectively. Conclusions derived according to these methods were compared. The differences in morphology, crystallinity, oxidation and gel behavior are caused primarily by the structural peculiarities of different types of PEs, the heterogeneous nature of radiation effects in semicrystalline polymers and, in the case of oriented samples, by the molecular orientation as well. The presented results reveal complicate crosslinking behavior with orientation while increase/decrease in crystallinity/oxidation with draw ratio was clearly evident. (C) 2007 Elsevier B.V.
