AB  - Indocyanine green (ICG) is the FDA-approved fluorescent dye used for in vivo medical imaging, diagnostics, and photothermal therapy. However, this dye is easily degradable in the human vascular system, and therefore its stabilization is preferable. In this work, ICG molecules were stabilized by their adsorption on the surface of the L-methionine-capped Ag and Au nanoparticles (Ag and Au @LM NPs) in aqueous colloidal dispersions. The result is the formation of hybrid metal core/ICG shell NPs in colloidal dispersions. Additionally, colloidal dispersions were stabilized, indicating a double effect of ICG adsorption. The obtained hybrid NPs were studied experimentally (UV–Vis spectrophotometry, HRTEM, DLS, FTIR) and theoretically (DFT calculations). HRTEM revealed that the interplanar spacing between adjacent planes of NPs decreases after the dye adsorption. The results obtained from the DFT study confirmed the formation of a covalent bond between the oxygen from ICG dye SO3− group and metal NPs. Considering the characteristics of both components of the NPs/ICG hybrid system, the authors assume that this hybrid system can exhibit the synergistic effect that could lead to more successful theranostic treatment of cancer in nanomedicine.

TEM measurements revealed the formation of a distinctive ~ 3 to 4 nm thick halo around the particles, while HRTEM measurements show microstructural changes in NPs. The DFT calculations were used to investigate the energetics of interaction between ICG molecule and Ag-surface. The obtained data indicate a strong interaction between Ag-atom from NPs surface and SO3− group of ICG molecule.

AB  - Treatment of various diseases, especially cancer treatment, includes the potentialuse of different types of nanoparticles and nanostructures as drug carriers. However,searching for less toxic and more efficient therapy requires further progress, wherein recentdevelopments in medicine increasingly include the use of various advanced nanostructures.Their more successful application might be achieved by leveling imbalances betweenthe potentiality of different nanostructures and the demands required for their safeuse. Biocompatibility, biodegradability, prolonged circulation time and enhanced accumulationand uptake by cells are some of the key preconditions for their usage in efficientdrug delivery. Thanks to their greatly tunable functions, they are major buildingblocks for manufacturing novel materials. Nevertheless, given that their toxicity is questionable,their practical application is challenging. Hereof, before entering the sphere ofhuman consumption, it is of critical importance to perform more studies regarding theirtoxicity and drug distribution. This review emphasizes recent advances in nanomedicine,employing different kinds of conventionally used nanoparticles as well as novel nanoparticlesand nanostructures. Special emphasis is placed on micro/nanomotors (MNMs), discussingtheir opportunities, limitations, challenges and possible applications in drug deliveryand outlining some perspectives in the nanomedicine area.

Improvement of sorption properties of natural clay pyrophyllite by ultrasonic treatment

AB  - Pyrophyllite, a naturally abundant clay material, exhibits remarkable physicochemical characteristics. Its minimal electrical and thermal conductivity, low expansion rate, strong mechanical properties, and outstanding heat resistance make it a valuable resource across diverse industries. Pyrophyllite finds applications in sectors such as paper and plastic manufacturing, brick production, ceramics, cosmetics, rubber processing, and wastewater treatment. Furthermore, its versatility extends to the production of ceramic membranes for efficient water filtration. This paper presents the characterization of natural pyrophyllite ore subjected to ultrasonic treatment at varying time intervals. The ultrasonic treatment aims to eliminate hard phases such as quartz and calcite within the ore, thereby improving its sorption capabilities. The treated samples were subjected to analysis using SEM and XRD techniques. Morphological and structural analysis revealed that as the duration of ultrasonic treatment increased, the proportion of hard phases in the sample decreased. Additionally, this study evaluated the sorption properties of pyrophyllite. A comparative analysis was conducted between a raw clay sample containing various admixtures and a sample that underwent a 30-minute ultrasonic treatment. The sorption of a methylene blue solution after 24 hours in water was assessed, with UV-Vis analysis revealing that the efficiency of the sonically treated pyrophyllite exceeded 97%, whereas the raw ore exhibited approximately 89% efficiency over the same duration. These findings suggest that the removal of hard phases from pyrophyllite ore enhances its sorption properties

AB  - Nanocomposites synthesized by incorporating nanostructured materials within the interlayer spaces of clay minerals have gained tremendous interest lately, owing to their exceptional physicochemical properties and vast potential in various fields. This study focuses on the mechanochemical activation of pyrophyllite, a commonly used natural clay material, using AgNO3 at different concentrations (2, 5, and 10 wt%). The activation process involved grinding the materials for varying durations ranging from 20 to 320 minutes. The resulting samples were analyzed using FTIR, TGA, and DTA thermal analysis techniques, XRD, SEM with EDX as well as PSD method for particle size distribution analysis. The results have shown a homogeneous distribution of silver along the analyzed surface. The duration of grinding and the amount of added silver greatly influence the composite physicochemical properties.

This study focuses on the mechanochemical activation of pyrophyllite, a commonly used natural clay material, using AgNO3 at different concentrations (2, 5, and 10 wt%). The activation process involved grinding the materials for varying durations ranging from 20 to 320 minutes. The resulting samples were analyzed using FTIR, TGA, and DTA thermal analysis techniques, XRD, SEM with EDX as well as PSD method for particle size distribution analysis. The results have shown a homogeneous distribution of silver along the analyzed surface. The duration of grinding and the amount of added silver greatly influence the composite physicochemical properties.",

AB  - The work aims to design a pyrophyllite-modified carbon paste electrode for pesticide detection in aqueous solutions. The structural and morphological characterization of natural pyrophyllite clay for Parsovići mine, Bosnia and Herzegovina, and mechanically modified pyrophyllite was performed using X-ray diffraction analysis (XRD), scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR) and particle size analysis (PSD). The electrochemical characteristics of the constructed electrode were investigated using cyclic voltammetry in 1 mM K4Fe(CN)6 in 0.1 M KCl and 0.5 M H2SO4 and differential pulse stripping voltammetry in Britton- Robinson buffer at pH 2-8. It was shown that the maximum at + 0.96 V versus Ag/AgCl originates from oxidation by carbendazim at pH 4 in the Britton-Robinson buffer. The electrode designed in this way showed numerous advantages such as good stability and sensitivity. The developed analytical method is linear over the range of 1 ppm to 10 ppm with r=0.999 and a detection limit of 0.3 ppm.

AB  - Ovaj rad ima za cilj da dizajnira elektrodu od ugljenične paste modifikovanu pirofilitom za

karakterizacija prirodne gline pirofilit i mehanički modifikovanog pirofilita urađena je
rendgenostrukturnom analizom i skenirajućom elektronskom mikroskopijom. Elektrohemijske
karakteristike ove elektrode ispitivane su cikličnom voltametrijom u 1 mM K4Fe(CN)6 u 0,1 M
KCl i 0,5 M H2SO4 i diferencijalnom pulsnom „striping“ voltametrijom u Briton-Robinsonovom
puferu na pH 4. Pokazano je da maksimum na + 0,96 V u odnosu na Ag/AgCl elektrodu potiče
od oksidacije karbendazima na pH 4 u Briton-Robinsonovom puferu. Dobru stabilnost i
osetljivost pokazala je elektroda koja je sadržala 50% ugljenične paste i 50% pirofilita mehanički
modifikovanog 15 minuta u mlinu sa kuglama. Razvijena metoda je linearna u opsegu od 1 ppm
do 10 ppm sa r= 0,999 i granicom detekcije od 0,3 ppm.

AB  - A new approach to drug design based on nanoparticles and related nanostructures for effective drug delivery, is of great importance in future medical treatment, especially for cancer therapy. Nanomaterials hold tremendous potential for increasing the efficiency of drug delivery, with a high degree of biocompatibility. Additionally, for biomedical applications, they must be biodegradable, have prolonged circulation half-life, not tend to aggregate or cause an inflammatory response in the body and to be cost-effective. The efficacy of such structures is highly dependent on their chemical properties as well as on shape, charge, size, surface modifications and loading method. Here we focused on the potential of using different kinds of nanoparticles and similar nanostructures loaded with various drugs in order to achieve specific targeting and controlled drug release. Thereby, computational modeling on NPs-based drug delivery could help in providing a better understanding of all parts of the delivery system. This review emphasizes recent advances in the usage of various types of nanoparticles and similar nanostructures for drug delivery, aiming to provide a critical review of less toxic and more effective treatment.

All the investigated compounds showed a strong inhibitory potency toward both enzymes with IC50 values in micromolar range of concentration; compound 1 displayed the most potent inhibitory behaviour toward both enzymes. The mechanism of the AChE and BuChE inhibition was examined by enzyme kinetic measurements. The obtained kinetic parameters, Vmax and Km indicated an uncompetitive type of inhibition of both enzymes by compound 1. For the other two compounds a non-competitive inhibition mode was observed. To get further insight into the mechanism of action and to elucidate binding modes in details we examined the interactions of 1–3 with acetylcholinesterase, using molecular docking approach. Grid based docking studies indicated that these compounds can bind to peripheral anionic site (PAS) of the AChE with Ki values in micromolar range. Moreover, blind docking revealed the capability of investigated compounds to bind to new allosteric site (i.e. binding site II) distinct from PAS. Showing that these Cu-based compounds can act as new allosteric inhibitors of AChE and identifying novel allosteric binding site on AChE represents a significant contribution toward the design of novel and more effective inhibitors of AChE. © 2020 Elsevier Inc.

AB  - Abstract: This paper presents the study of the interaction between gold nanoparticles (AuNPs) and antitumor gold complexes, [Au(OH)2(bipy)][PF6], [Au(CH3COO)2(pydmb-H)], and [Au(bipydmb-H)(OH)][PF6], in order to estimate the possibility for metal complex tracking in cells using nanospectroscopy approach. Decrease of intensity of the surface plasmon absorption band at 524 nm and the appearance of a new broad band at ∼640 nm, followed by their red shift were observed in the presence of the complexes. TEM images showed that the average size and shape of AuNPs did not change after the addition of gold complexes. DLS and zeta potential measurements pointed out to the metal complex adsorption on the surface of AuNPs followed by flocculation process. In addition, kinetic studies indicated strong bond formation between these complexes and AuNPs. Accordingly, AuNPs/complex conjugates have the potential to be applied for the tracking of these metal complexes in the cells using nanospectroscopy approach. © 2019, Pleiades Publishing, Ltd.

AB  - The present paper deals with investigation of the interaction between selected simple structure Au(iii) ([AuCl4]-, [AuCl2(dmso)2]+, [AuCl2(bipy)]+) and Pt(ii) ([PtCl2(dmso)2]) complexes with Na/K-ATPase as the target enzyme, using an experimental and theoretical approach. Reaction stoichiometries and binding constants for these enzyme/complex systems were determined, while kinetic measurements were used in order to reveal the type of inhibition. Based on the results obtained by quantum mechanical calculations (electrostatic surface potential (ESP), volume and surface of the complexes) the nature of the investigated complexes was characterized. By using the solvent accessible surface area (SASA) applied on specific inhibitory sites (ion channel and intracellular domains) the nature of these sites was described. Docking studies were used to determine the theoretical probability of the non-covalent metal binding site positions. Inhibition studies implied that all the investigated complexes decreased the activity of the enzyme while the kinetic analysis indicated an uncompetitive mode of inhibition for the selected complexes. Docking results suggested that the main inhibitory site of all these complexes is located in the ion translocation pathway on the extracellular side in the E2P enzyme conformation, similar to the case of cardiac glycosides, specific Na/K-ATPase inhibitors. Also, based on our knowledge, the hydrolyzed forms of [AuCl4]- and [PtCl2(dmso)2] complexes were investigated for the first time by theoretical calculations in this paper. Thereby, a new inhibitory site situated between the M2 and M4 helices was revealed. Binding in this site induces conformational changes in the enzyme domains and perturbs the E1-E2P conformational equilibrium, causing enzyme inhibition.

AB  - In vitro oxidative stress responses of two Keggin-type polyoxotungstates, 12- tungstosilicic (WSiA) and 12-tungstophosphoric acid (WPA), were investigated using rat synaptosomes as a model system. WSiA induced concentration-dependent increase in catalase activity, up to about 6 times compared to the control activity in untreated synaptosomes, and glutathione peroxidase was not significantly affected after WSiA treatment. On the contrary, WPA treatment resulted in the increase of glutathione peroxidase activity, while synaptosomal catalase was even reduced related to the control, at all investigated WPA concentrations. Both investigated polyoxotungstates did not significantly change malondialdehyde content in synaptosomal preparations. It could accordingly be concluded that WSiA and WPA probably induce reactive oxygen species generation, resulting in the activation of the antioxidant defense enzymes. However, these polyoxotungstates are not strong prooxidants being able to cause oxidative stress, and consequently synaptosomal membrane lipid damage.

AB  - Inhibition of acetylcholinesterase (AChE) is presented as a promising strategy in the treatment of Alzheimer disease providing inspiration for new discoveries and investigations less toxic and more effective potential anti Alzheimer drugs. In this paper, it demonstrated that the activity of acetylcholinesterase can be effectively inhibited by polyoxometalates (POMs), 12-tungstosilicic acid (WSiA) and 12-tungstophosphoric acid (WPA) without significant changes on the secondary structure of this enzyme. The obtained values of partition coefficient implicated on smooth pass of these POMs trough cell membrane and satisfied necessary criteria for the drugs used in the treatment of the central nervous system disease. Based on these obtained results it is possible to conclude that POM could represent new generation of potential anti Alzheimer drugs.

AB  - The aim of this work was to investigate the formation of J-aggregates of thiacyanine dye (TC, 5,5-disulfopropyl-3,3-dichlorothiacyanine sodium salt) in the presence of 6 nm spherical silver nanoparticles (Ag NPs) using spectrophotometric and fluorescence methods. The formation of J-aggregates was concentration dependent and characterized by the appearance of the new absorption band with the maximum at 481 nm. Spectrophotometric study of J-aggregate formation and time stability suggested that they were formed on the account of monomer form of TC. Moreover, the stability of J-aggregates increased with the lowering AgNPs concentration. The measurements of fluorescence of the NPs-dye assembly clearly indicated that the fluorescence of TC was quenched by Ag NPs on the concentration dependent manner. The spectrophotometric and fluorescence properties of NPs-dye assembly were found to be quantitatively related to the surface coverage of the dye on the Ag NPs.

AB  - The in vitro influence of gold(III) complexes, H[AuCl4], [Au(DMSO)(2)Cl-2]Cl and [Au(bipy)Cl-2]Cl (bipy = 2,2-bipyridine), upon commercially available Na+ /K+ ATPase activity, purified from porcine brain cortex, was investigated. Additionally, the complexes were tested on human lymphocytes, and incidence of micronuclei and cell proliferation index was determined. Concentration-dependent inhibition of the enzyme for all three compounds was obtained, but with differing potencies. Calculated IC50 from Hill analysis were (in M): 5.75 x 10(-7), 5.50 x 10(-6) and 3.98 x 10(-5), for H[AuCl4], [Au(DMSO)(2)Cl-2]Cl and [Au(bipy)Cl-2]Cl, respectively, while Hill coefficient values, n, were above 1 in all cases. This inhibition can be prevented using -SH donating ligands such as L-Cys and glutathione, and these ligands can also cause a recovery of the enzyme activity after the induced inhibition. Kinetic analysis demonstrated that each of the studied gold(III) complexes affects Na+ /K+ ATPase reducing maximum enzymatic velocity, V-max, but not significantly changing the affinity for the substrate (K-M value), implying a noncompetitive mode of the interaction. Furthermore, among investigated gold(III) complexes, the [Au(bipy)Cl-2]Cl complex exhibits a strong cytotoxic effect on human lymphocytes, which suggests its potential for use in antitumor therapy. AB  - Nanoparticles (NPs) modified with fluorescent cyanine dyes can exhibit fluorescent quenching properties due to energy and/or electron transfer, and these systems have many applications in catalysis, drug delivery, nanoelectronics, medical diagnostics, and chemical sensing. The aim of this work was to investigate the adsorption of cyanine dye 5,5-disulfopropyl-3,3-dichlorothiacyanine (TC) on the surface of citrate capped gold nanoparticles (AuNPs) with the average core sizes of 9, 17, and 30 nm using various experimental techniques. The measurements of fluorescence corrected for inner filter effects (IFE) of the particle dye assembly clearly indicated that the fluorescence of TC was quenched by AuNPs on the concentration dependent 3 manner, and the equilibrium constants for the sorption of TC on the NP surface were calculated. Significant increase of fluorescence quenching was noticed when NP size increased, keeping the concentration of NPs of different size constant. By comparing experimental and calculated results for quenching of TC dye fluorescence, we assumed that the maximum quenching was restricted to full monolayer coverage of TC on the NP surface. The conclusion was drawn that the most probable orientation of TC dye molecules on the NP surface was slanted and that the dye approached NP surface with its positively charged thiazole ring.

The characterization of AuNPs in the presence and absence of TC was performed by measurements of the absorption, fluorescence and Raman spectra, TEM, DLS and zeta potential. The measurements of fluorescence of the NPs-TC assembly in the presence of low TC concentration clearly indicated that the fluorescence of TC was quenched by AuNPs in the concentration dependent manner. This process was found to be quantitatively related to the surface coverage of AuNPs by TC molecules. NPs surface coverage by TC was obtained from the dependence of TC fluorescence on NPs or TC concentration, keeping the concentration of one of the components of the NPs-TC assembly constant. The calculated values were obtained under the assumption that the monolayer of TC molecules, which were in direct contact with the NPs surface, was adsorbed. The experimentally obtained values were in accordance with the calculated ones for TC orientation along the vertical short side. The formation of TC J-aggregates was evident only in the presence of 6 nm NPs, when TC concentration was high. In this case the experimentally obtained value for C6 concentration needed to completely quench TC dyes fluorescence was much lower than the calculated, indicating that the dye was accommodated on the surface of these NPs in more than one layer, and most probably in the slanted configuration.

AB  - The aim of this study was to investigate the inhibitory activity of Gentiana lutea extracts on the enzyme myeloperoxidase (MPO), as well as the antioxidant activity of these extracts and their correlation with the total polyphenol content. Extracts were prepared using methanol (100%), water and ethanol aqueous solutions (96, 75, 50 and 25% v/v) as solvents for extraction. Also, isovitexin, amarogentin and gentiopicroside, pharmacologically active constituents of G. lutea were tested as potential inhibitors of MPO. Antioxidant activity of extracts was determined using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging test and also using cyclic voltammetry (CV). Among all extracts, the antioxidant capacity of 50% ethanol aqueous extract was the highest, both when measured using the DPPH test, with IC50 = 20.6 mu g/ml, and when using CV. Also, 50% ethanol extract, showed the best inhibition of MPO activity in comparison with other extracts. In the group of the selected G. lutea constituents, gentiopicroside has proved to be the strongest inhibitor of MPO, with IC50 = 0.8 mu g/ml. AB  - The kinetics of J-aggregate assembly on the surface of noble metal nanoparticle colloids are described, providing new mechanistic insight into the interaction of the dye molecules with the surface of metal nanoparticles. We specifically studied the J-aggregation of a thiacyanine dye (TC, 3,3-disulfopropyl-5,5-dichlorothiacyanine sodium salt) on the surface of gold nanopartide colloids. The hybrid J-aggregate Au colloidal dispersions were characterized by UV vis spectrophotometry, fluorescence measurements, zeta potential measurements, and TEM analysis. Kinetic measurements were carried out using a stopped-flow method, indicating that the J-aggregate formation on the surface occurs via a two-step process. The first step includes adsorption of the initial dye layer, followed by an order of magnitude slower growth of consecutive layers. Activation parameters determined from the fluorescence measurements yielded further details about the nature of the interaction.

AB  - The spherical gold nanoparticles (NPs) of different average size (15 nm and 47 nm) prepared by citrate reduction were used to examine their cytotoxic effects. UV-vis spectrophotometry, transmission electron microscopy (TEM), dynamic light scattering (DLS) and zeta potential measurements were used to characterize the gold NPs. We have investigated the concentration and size dependent cytotoxic effects of gold NPs, using two in vitro human cells model systems: proliferating lymphocytes and connective tissue fibroblasts. Treatment of lymphocyte cultures with Au NPs caused cytotoxic effects as revealed by significant enhancement of incidence of micronuclei but non significant increase of cell proliferation potential when compared to the control. In a fibroblast cell line, the same doses of gold NPs induced the slightly higher level of gamma-H2AX foci than in a control. The presented results clearly indicate that the enhancement of micronuclei incidence and proliferation index depend not only on concentration but also the size of the nanoparticles.
