AB  - Two new Schiff base cobalt(III) ([Co(LH)Cl2], 1) and nickel(II) ([Ni(LH)ClO4], 2) complexes with a diimine-dioxime ligand, (4,9-diaza-3,10-diethyl-3,9-dodecadiene-2,11-dione bis oxime (LH2)), were synthesized and characterized. The compounds were obtained in MeOH from corresponding metal salts and LH2 in molar ratio 1:1 and further characterized by mass spectrometry, IR spectroscopy, electrochemistry, and elemental analysis. Previously reported copper(II) analog ([Cu2(LH)2]·(ClO4)2, 3) was joined to 1 and 2, and the three metal analogs, 1–3, were further investigated in terms of their electrochemical behavior. Binding studies of the complexes with deoxyribonucleic acid (DNA) and human serum albumin (HSA) were carried out using both spectrophotometric and electrochemical methods. All three complexes exhibit binding affinity towards the DNA chain through intercalative interactions. The binding reaction with HSA showed for 1 and 3 decrease in the peak current obtained in the case of complexes before the addition of HSA, while the Ni complex–HSA possesses the same electroactivity as starting complex. The cytotoxicity of LH2 as well as its metal complexes, and cisplatin were evaluated on CT-26 mouse colon carcinoma and human LS174T cancer cell lines employing MTT assay. The copper(II) complex exhibited very promising anticancer activity compared with cisplatin.

90Y-CA/SPIONs for dual magnetic hyperthermia-radionuclide nanobrachytherapy of solid tumours

AB  - Radiolabelled superparamagnetic iron oxide nanoparticles (SPIONs) are a promising nanomaterial for the development of dual radiation/hyperthermia cancer therapy. To that purpose, flower-shaped SPIONs with an exceptional heating capability were synthesised and coated with citrate, dextran or (3-aminopropyl)triethoxysilane. Both non-coated and coated SPIONs were nontoxic to CT-26 mouse colon cancer cells up to 1.0 mg ml−1 in vitro. In an oscillating magnetic field, citrate-coated SPIONs (CA/SPIONs) displayed the highest heating rate (SAR ∼ 253 W g−1) and the strongest hyperthermia effects against CT-26 cells. Labelling of the CA/SPIONs by the 90Y radionuclide, emitting β− radiation with an average/maximum energy of 0.94/2.23 MeV, and deep tissue penetration generated 90Y-CA/SPIONs intended for the therapy of solid tumours. However, intravenous injection of 90Y-CA/SPIONs in CT-26 xenograft-bearing mice resulted in low tumour accumulation. On the contrary, intratumoural injection resulted in long-term retention at the injection site. A single intratumoural injection of 0.25 mg CA/SPIONs followed by 30-min courses of magnetic hyperthermia for four consecutive days caused a moderate antitumour effect against CT-26 and 4T1 mouse tumour xenografts. Intratumoural application of 1.85 MBq/0.25 mg 90Y-CA/SPIONs, alone or combined with hyperthermia, caused a significant (P ≤ 0.01) antitumour effect without signs of systemic toxicity. The results confirm the suitability of 90Y-CA/SPIONs for monotherapy or dual magnetic hyperthermia-radionuclide nanobrachytherapy (NBT) of solid tumours.

AB  - Surface modification of magnetic nanoparticles with poly-L-lysine, proline, and tryptophan was used to design potential theranostic agents for the application in cancer diagnosis and radionuclide-hyperthermia therapy. Characterization of bare and functionalized magnetic nanoparticles was performed in detail. The transparency of the examined magnetic nanoparticles was measured in the non-alternating magnetic field for a complete and better understanding of hyperthermia. For the first time amino acid-functionalized magnetic nanoparticles were labeled with theranostic radionuclides 131I and 177Lu. The specific absorption rate (SAR) procured for poly-L-lysine functionalized magnetic nanoparticles (SAR values of 99.7 W/g at H0 = 15.9 kA/m and resonant frequency of 252 kHz) demonstrated their possible application in magnetic hyperthermia. Poly-L-lysine functionalized magnetic nanoparticles labeled with 177Lu showed the highest radiochemical purity (>99.00 %) and in vitro stability in saline and serum (>98.00 % up to 96 h). The in vivo analysis performed after their intravenous administration in healthy Wistar rats presented good in vivo stability for several days. Encouraging results as well as magnetic and radiochemical properties of 177Lu–PLL-MNPs (80 °C) justify their further testing toward the potential use as theranostic agents for diagnostic and combined radionuclide-hyperthermia therapeutic applications.

AB  - The combination of two imaging modalities in a single agent has received increasing attention during the last few years, since its synergistic action guarantees both accurate and timely diagnosis. For this reason, dual-modality contrast agents (DMCAs), such as radiolabeled iron oxide (namely Fe3O4) nanoparticles, constitute a powerful tool in diagnostic applications. In this respect, here we focus on the synthesis of a potential single photon emission computed tomography/magnetic resonance imaging (SPECT/MRI) DMCA, which consists of Fe3O4 nanoparticles, surface functionalized with 2,3-dicarboxypropane-1,1-diphosphonic acid (DPD) and radiolabeled with 99mTc, [99mTc]Tc-DPD-Fe3O4. The in vitro stability results showed that this DMCA is highly stable after 24 h of incubation in phosphate buffer saline (~92.3% intact), while it is adequately stable after 24 h of incubation with human serum (~67.3% intact). Subsequently, [99mTc]Tc-DPD-Fe3O4 DMCA was evaluated in vivo in mice models through standard biodistribution studies, MR imaging and gamma-camera imaging. abstract = "The combination of two imaging modalities in a single agent has received increasing attention during the last few years, since its synergistic action guarantees both accurate and timely diagnosis. For this reason, dual-modality contrast agents (DMCAs), such as radiolabeled iron oxide (namely Fe3O4) nanoparticles, constitute a powerful tool in diagnostic applications. In this respect, here we focus on the synthesis of a potential single photon emission computed tomography/magnetic resonance imaging (SPECT/MRI) DMCA, which consists of Fe3O4 nanoparticles, surface functionalized with 2,3-dicarboxypropane-1,1-diphosphonic acid (DPD) and radiolabeled with 99mTc, [99mTc]Tc-DPD-Fe3O4. The in vitro stability results showed that this DMCA is highly stable after 24 h of incubation in phosphate buffer saline (~92.3% intact), while it is adequately stable after 24 h of incubation with human serum (~67.3% intact). Subsequently, [99mTc]Tc-DPD-Fe3O4 DMCA was evaluated in vivo in mice models through standard biodistribution studies, MR imaging and gamma-camera imaging. AB  - Type of the wood used for the aging highly influences the quality of alcoholic beverages. In this research we explored the potential of cyclic voltammetry (CV) and differential pulse voltammetry (DPV) to establish fingerprints characteristic for each wood and to enable determining the type of the wood used in the aging process. Eleven different wood samples were used to prepare three different types of spirits during 15 months. The highest extraction rate was obtained during the first month, while further aging was followed with almost constant amount of extracted polyphenols. Black locust, myrobalan plum, and mulberry extracts were discriminated from the spirits aged in oak and wild cherry wood when statistical analysis was applied. Although clear classification of all samples was not achieved, this long term study demonstrated a potential of both CV and DPV for differentiating wood species used in the aging, hence in the quality control of spirits. © 2020 Elsevier Ltd

AB  - This work presents advanced electrochemical platform based on screen printed diamond electrode (SPDE) system for the single drop “point-of-care” testing. Proposed approach was applied for the quantification of doxorubicin hydrochloride (DOX) in biological fluids and pharmaceutical product. Using a single drop (~30 μL) of the tested solution at the electrode surface, DOX showed high electroactivity over a wide range of pHs. In these conditions, single oval shaped, well-defined and pH dependent oxidation peak was observed in the potential range from 0.5 V to 1.3 V. In the reverse scan, two cathodic peaks, were noted – around 0.3 V and – 0.5 V. Similarly, first reduction peak was pH dependent, while second one was independent in the studied range. Experimental conditions for DOX quantification were optimized and natures of the electrode reactions were investigated. Working linear range obtained for DOX detection was from 0.1 to 2.5 μM. Diffusion controlled electrode reaction reveal long life time of the proposed electrode as well high potential for practical application. Developed procedure was successfully applied for the DOX analysis in biological fluids – urine and pharmaceutical formulation. Obtained results clearly indicated that given procedure can be easily implemented for pharmaceutical control and medical analysis, in both, laboratory and field conditions. © 2021 Elsevier B.V.

AB  - The homemade, porous carbon material, thermolysis prepared from Novolac phenol-formaldehyde resin, in situ modified with Co3O4 nanoparticles and mixed with single-wall carbon nanotubes, was used in selective sensing of prominent antioxidant α-lipoic acid (LA). XRD, SEM and EIS measurements were used for characterization of material composition, structure, morphology and improved conductivity. The quantification of LA at TPCo3O4&SWCNTCPE was done by a square-wave voltammetric technique in BR buffer solution at pH 6. The linear working range was recorded from 2 to 100 μM of LA and the proposed electrode material was successfully applied in the determination of LA in dietary supplements.

AB  - The goal of this work was to develop green electrode material that unites all advantages of domestic made, synthesized porous carbon powder and ceria dioxide nanoparticles known due to exceptional catalytic properties. Thermal decomposition of Novolac phenol-formaldehyde resin and cerium sulfate resulting in producing a high-performance CeO2 porous carbon material highly sensitive to dopamine (DA) electrooxidation. Morphological and structural characteristics of the material were determined by SEM and XRD measurements, while electrochemical characterization was performed by EIS and CV. The sensitivity of DA determination on the proposed CeO2-doped carbon material was enhanced by adding multi-wall carbon nanotubes to finally prepare a mixture for a specific carbon paste electrode (TPCeO2&MWCNT@CPE). SWV technique was used for quantification of dopamine in Britton-Robinson buffer pH 6 in the concentration range of 0.5-100 μM of DA, with the detection limit of 0.14 μM and quantification limit of 0.44 μM. Good selectivity overstudied bioactive compounds enables the successful and efficient application of the proposed electrode and developed an analytical procedure for the determination of dopamine in spiked urine samples.

AB  - Purpose Recent studies with doxycycline as adjuvant therapy to conventional chemotherapy have shown promising results in cancer therapy. The current study aimed to examine the capability of 177Lu-labeled tetracycline ligand, doxycycline hyclate, to use as an anticancer agent.Materials and methods Doxycycline was radiolabeled with beta-emitting radioisotope 177Lu. Complex formation and its interaction with DNA were investigated electrochemically. Binding of 177Lu-doxycycline to CT 26 cell line was done. Biodistribution of 177Lu-doxycycline was examined in healthy Wistar rats and CT26 colon carcinoma tumor-bearing mice by i.v. and i.p. administration, respectively.Results Doxycycline hyclate was successfully radiolabeled with 177Lu in high radiolabeling yield (>99%). The radiolabeled complex was stable in vitro in saline and human serum over 72 h. Non-radioactive Lu-doxycycline complex formation was demonstrated electrochemically as well. Intercalative interactions of the doxycycline and Lu-doxycycline with DNA were proved using simultaneously spectrophotometric and electrochemical methods. The binding of the radiolabeled complex with plasma proteins was 4.0 ± 0.4%. The partition coefficient showed the lipophilic nature of the complex similar to the free ligand. The binding curve demonstrates binding from 0.1 nM concentrations of 177Lu-doxycycline, with half-binding estimated ∼100 nM. Biodistribution studies of 177Lu-doxycycline in CT26 colon tumor-bearing mice showed a satisfactory accumulation rate in the tumor (2.88 ± 0.85% ID/g) 3 h after intraperitoneal injection. abstract = "Purpose Recent studies with doxycycline as adjuvant therapy to conventional chemotherapy have shown promising results in cancer therapy. The current study aimed to examine the capability of 177Lu-labeled tetracycline ligand, doxycycline hyclate, to use as an anticancer agent.Materials and methods Doxycycline was radiolabeled with beta-emitting radioisotope 177Lu. Complex formation and its interaction with DNA were investigated electrochemically. Binding of 177Lu-doxycycline to CT 26 cell line was done. Biodistribution of 177Lu-doxycycline was examined in healthy Wistar rats and CT26 colon carcinoma tumor-bearing mice by i.v. and i.p. administration, respectively.Results Doxycycline hyclate was successfully radiolabeled with 177Lu in high radiolabeling yield (>99%). The radiolabeled complex was stable in vitro in saline and human serum over 72 h. Non-radioactive Lu-doxycycline complex formation was demonstrated electrochemically as well. Intercalative interactions of the doxycycline and Lu-doxycycline with DNA were proved using simultaneously spectrophotometric and electrochemical methods. The binding of the radiolabeled complex with plasma proteins was 4.0 ± 0.4%. The partition coefficient showed the lipophilic nature of the complex similar to the free ligand. The binding curve demonstrates binding from 0.1 nM concentrations of 177Lu-doxycycline, with half-binding estimated ∼100 nM. Biodistribution studies of 177Lu-doxycycline in CT26 colon tumor-bearing mice showed a satisfactory accumulation rate in the tumor (2.88 ± 0.85% ID/g) 3 h after intraperitoneal injection. Magnetic nano- and micro-particles based on Gd-substituted magnetite with improved colloidal stability

AB  - A series of Fe3-xGdxO4 (x = 0, 0.1, 0.2) nanoparticles with an average diameter of around 8 nm were prepared by the coprecipitation method and coated by citric acid (CA). The nanoparticles show superparamagnetic behavior at room temperature and transition to a blocked state, at a temperature from ~ 89 K to ~ 213 K, depending on Gd concentration. The saturation magnetization of Fe3-xGdxO4 tended to drop for samples with a higher content of Gd. High colloidal stability is mandatory in medical applications of magnetic nanoparticles, and here we demonstrate a new procedure for its improvement. A colloidal sample of Fe3O4@CA was mechanically milled, after which dynamic light scattering and zeta potential measurements were used to monitor the hydrodynamic size and colloidal stability of the acquired suspensions. After 90 min of milling, the average hydrodynamic diameter decreased by 40%, and size distribution changed from polymodal to monomodal, while the negative zeta potential increased from − 30.5 mV to − 52.8 mV. Additionally, Fe2.80Gd0.20O4@CA nanoparticles were embedded in human serum albumin to produce magnetic microspheres (MMS), which could be used as a drug delivery platform. Both measurements are in good agreement while TEM micrographs additionally indicate to a large multivesicular inner structure of prepared liposomes. A simple and straightforward procedure was used for liposome radiolabeling with 177Lu, a well–known and commonly used radionuclide in radiotherapy with favorable properties, that can be exploited in RSO therapy. Radiolabeled 177Lu–liposomes were tested in vitro for stability and then injected into the knee joints of Wistar rats where liposome in vivo behavior was followed up to 30 days post injection. Results from both ex vivo biodistribution and in vivo imaging studies presented a high stability and retention (>94 %ID) of 177Lu–micro liposomes in the synovial liquid for the entire observation period. Leakage of free 177Lu or 177Lu–liposomes from the synovial fluid has not been detected, indicating to a possible application of 177Lu–liposomes in radiosynoviorthesis (RSO) therapy.

The liposomes were characterized with TEM and DLS measurements, and MNPs encapsulation rate was determined using ICP-OES. Prepared liposomes were stored at 5 °C for 30 days and subsequently exposed to an external magnetic field (20 mT) with varying exposure times (2‒20 min), at room temperature. The release of fluorescein from GMLs induced by the magnetic field exposures was quantified, showing a high release rate (25‒85%) depending on the concentration of MNPs in GMLs. EPR measurements were conducted during the liposomes storage period in order to provide semi-quantitative information of possible MNPs oxidation from Fe3O4 to Fe2O3 inside the liposomes, impacting MNPs magnetic properties. In contrast to the MNPs water dispersion, no significant change in the EPR signal of MNPs encapsulated inside GMLs was detected over the course of 30 days. The data presented in this study indicate that GMLs loaded with MNPs maintain a high stability for prolonged periods of time and that this delivery system may be used for magnetically assisted controlled drug release.

AB  - Liposomes are promising drug's delivery systems due to decreased toxicity of the liposome-encapsulated drug, but wider clinical application requires their more efficient tumor targeting with uptake, controlled drug release and higher shelf life. The unique metabolic characteristics of cancer cells based on higher demand for energy and therefore increased glucose utilization were exploited in the design of glucose modified liposomes (GML) with the aim to provide increased tumor targeting via glucose transporters and increased ability of drug delivery into tumor cells. Tumor accumulating potential of GML and non-glucose liposomes (NGL) were investigated on CT26 and LS174T tumor-bearing mice by simple and reliable radiotracer method using 177Lu as radioactive marker. Both liposomes, GML and NGL were radiolabeled in high radiolabeling yield, showing high in vitro stability in biological media, as the main prerequisite for the biodistribution studies. Tumors displayed significantly better accumulation of 177Lu-GML with the maximum uptake 6 h post-injection (5.8 ± 0.2%/g in LS174T tumor and 5.1 ± 0.5%/g in CT26 tumor), compared to negligible uptake of 177Lu-NGL (0.6 ± 0.1%/g in LS174T tumor and 0.9 ± 0.2%/g in CT26 tumor). Results of comparative biodistribution studies of 177Lu-NGL and 177Lu-GML indicate that increased accumulation of GML is enabled by glucose transporters and subsequent endocytosis, resulting in their prolonged retention in tumor tissues (up to 72 h). Direct radiolabeling of liposomes with 177Lu may be used not only for biodistribution studies using radiotracking, but also for cancer treatment. © 2021 Elsevier B.V.

AB  - Different phosphates and phosphonates have shown excellent coating ability toward magnetic nanoparticles, improving their stability and biocompatibility which enables their biomedical application. The magnetic hyperthermia efficiency of phosphates (IDP and IHP) and phosphonates (MDP and HEDP) coated Fe3O4 magnetic nanoparticles (MNPs) were evaluated in an alternating magnetic field. For a deeper understanding of hyperthermia, the behavior of investigated MNPs in the non-alternating magnetic field was monitored by measuring the transparency of the sample. To investigate their theranostic potential coated Fe3O4-MNPs were radiolabeled with radionuclide 177Lu. Phosphate coated MNPs were radiolabeled in high radiolabeling yield (> 99%) while phosphonate coated MNPs reached maximum radiolabeling yield of 78%. Regardless lower radiolabeling yield both radiolabeled phosphonate MNPs may be further purified reaching radiochemical purity of more than 95%. In vitro stabile radiolabeled nanoparticles in saline and HSA were obtained. The high heating ability of phosphates and phosphonates coated MNPs as sine qua non for efficient in vivo hyperthermia treatment and satisfactory radiolabeling yield justifies their further research in order to develop new theranostic agents. © 2021, The Author(s), under exclusive licence to Springer Science+Business Media, LLC, part of Springer Nature.

Investigation of 177Lu-labeled HEDP, DPD, and IDP as potential bone pain palliation agents

AB  - Application of bone-seeking radiopharmaceuticals is one of the modalities in the management of metastatic bone pain. The present study aimed to investigate the potential of Lu-177-labeled phosphate/phosphonate ligands: 1-hydroxyethane 1,1-diphosphonic acid (HEDP), 2,3-dicarboxypropane-1,1-diphosphonic acid (DPD), and imidodiphosphate tetrasodium salt (IDP), as bone pain palliation agents. HEDP, DPD, and IDP were radiolabeled with Lu-177 in high radiolabeling yield (98.49%, 93.31%, and 90.69%, respectively), forming in vitro stable radiolabeled complexes in saline and human serum after 96 h. Biodistribution was followed by imaging studies and ex vivo measurement of radioactivity in organs in healthy Wistar rats. Significant bone accumulation and long retention even after 96 h (3.85 +/- 0.91%ID/g), as well as relatively low uptake in soft tissue such as liver and spleen (<1%ID/g), were observed for Lu-177-HEDP. Two other radiolabeled ligands showed lower accumulations in bone (<1% ID/g) and higher accumulations in liver and spleen at examined time points (>1.5% ID/g). Obtained results suggest that difference in the chemical structure of phosphonates/phosphates influences the rate of bone incorporation of Lu-177-radiolabeled complexes. Desirable biodistribution pattern of Lu-177-HEDP makes it suitable for its further preclinical investigations as a potential bone pain palliation agent.

Tailoring IONP shape and designing nanocomposite IONS@GN toward modification of SPCE to enhance electrochemical degradation of organic dye

AB  - Iron oxide nanoparticles (IONP) with different distinctive morphologies (spherical, cubic, flower-like and needles) were utilized for modification of screen-printed carbon electrodes (SPCE) to be used for synthetic organic dye degradation by an electrochemical approach. This platform was implemented for removal of the synthetic organic dye, Reactive Black 5 (RB5) in aqueous solution. Modified SPCE with spherically shaped IONP (IONS) had the highest dye removal efficiency. Thus, IONS were then used for surface decoration of the most common carbon-based materials (graphene, graphene oxide, carboxylated graphene, graphene nanoribbons, graphene nanoplatelets, single- and multi-wall carbon nanotubes), and the nanocomposites formed were deposited on the electrode surfaces. Using IONS/graphene composite (IONS@GN) for electrode modification resulted in the best effect. Removal of RB5 with this electrode was 51% better in comparison with bare SPCE, reducing the time required for complete dye degradation from 61 to 30 min Using IONS-modified SPCE, total RB5 removal occurred in 51 min, improving the performance by 16% over that of bare SPCE. The effects determined, i.e., the best IONP morphology and best type of carbon-based material for nanocomposite formation to enhance RB5 removal will provide guidelines for further modifications of SPCE with nanomaterials and nanocomposites, for application of this electrochemical approach in the degradation of organic pollutants. © 2020 The Author(s). A single drop histamine sensor based on AuNPs/MnO2 modified screen-printed electrode

AB  - In order to ensure high food quality, one of the prime importance is the detection and quantification of histamine, well known marine food poison. In this work, we constructed novel electrochemical biosensor for the detection of histamine based on gold nanoparticles decorated on manganese dioxide (Au/MnO2) and used for modification of screen-printed carbon electrode (Au/MnO2@SPCE). The constructed sensor was then used for the estimation of histamine content in a single drop. Materials used in this study were synthesized and characterized using HR-TEM, XRPD and electrochemical methods. The amperometric detection method was optimized and, under selected operating parameters (supporting electrolyte pH 6, working potential of 1 V), the proposed sensor possesses linear working range from 0.3 µM to 5.1 µM, with a detection limit of 0.08 µM. The effect of selected interferences was investigated and it was found that the developed approach offers accurate, precise, selective, fast and reproducible quantification of histamine using only one drop of the sample. In the end, this work stands as a proof-of-concept of the modified electrodes and electrochemical detection as a promising and prospective approach for the applications in real-time monitoring of the food quality.

The amperometric detection method was optimized and, under selected operating parameters (supporting electrolyte pH 6, working potential of 1 V), the proposed sensor possesses linear working range from 0.3 µM to 5.1 µM, with a detection limit of 0.08 µM. The effect of selected interferences was investigated and it was found that the developed approach offers accurate, precise, selective, fast and reproducible quantification of histamine using only one drop of the sample. In the end, this work stands as a proof-of-concept of the modified electrodes and electrochemical detection as a promising and prospective approach for the applications in real-time monitoring of the food quality.",

AB  - Herein, we proposed a novel approach and state-of-the-art technology for the improvement of the materials and enzyme immobilization at the electrode surface and construction of impedimetric glucose biosensor. We silanized titanium dioxide nanoparticles using (3-aminopropyl)triethoxysilane (APTES), for the preparation of cross-linked material nanoparticles, with carboxylic graphene. The silanization of titanium dioxide nanoparticles and an increase in electron shuttle was proven feasible when this composite was able to achieve about 30% higher current than non-silanized material. The proposed approach was used for the modification of the printed three-electrode system and the development of the impedimetric glucose biosensor. The material morphology and electrochemical characteristics were confirmed by spectroscopic and electrochemical methods. The present combination effectively modified the electrode surface and serve as a promising basis for the construction of Point-of-Care devices. Developed biosensor possesses wide operating linear range toward glucose detection from 50 µmol to 1000 µmol, with the limit of detection of 24 µmol. Finally, negligible interference effect and application in the real sample indicate that the proposed mechanism can be successfully applied to the assessment of glucose level in only one drop of real sample.

AB  - Combined radionuclide therapy with magnetic nanoparticles-mediated hyperthermia has been under research focus as a promising tumor therapy approach. The objective of this study was to investigate the potential of 131I-radiolabeled superparamagnetic iron oxide nanoparticles (SPIONs) prepared as the ~40 nm flower-shaped structures with excellent heating efficiency (specific absorption rate at H0 = 15.9 kA∙m−1 and resonant frequency of 252 kHz was 123.1 W∙g−1) for nano-brachytherapy of tumors. 131I-radiolabeled CC49 antibody attached to SPIONs via reactive groups of 3-aminopropyltriethoxysilane (APTES) provided specificity and long-lasting localized retention after their intratumoral application into LS174T human colon adenocarcinoma xenografts in NOD-SCID mice. The results demonstrate feasibility and effectiveness of magnetic hyperthermia (HT), radionuclide therapy (RT) and their combination (HT + RT) in treating cancer in xenograft models. Combined therapy approach induced a significant (p < 0.01) tumor growth suppression in comparison to untreated groups presented by the tumor volume inhibitory rate (TVIR): 54.38%, 68.77%, 73.00% for HT, RT and HT + RT, respectively in comparison to untreated group and 48.31%, 64,62% and 69,41%, respectively, for the SPIONs-only injected group. Histopathology analysis proved the necrosis and apoptosis in treated tumors without general toxicity. Obtained data support the idea that nano-brachytherapy combined with hyperthermia is a promising approach for effective cancer treatment.

AB  - Dual-modality contrast agents (DMCA), such as radiolabeled magnetic nanoparticles, have attracted significant attention in diagnostic applications due to their potency for the timely and accurate diagnosis of diseases. The hemocompatibility of a candidate DMCA with human blood is essential for the investigation of its application in vivo. In this respect, here we focused on the evaluation of the hemocompatibility of a new DMCA, that is based on iron oxide nanoparticles (i.e. Fe3O4 magnetite), with human red blood cells (RBCs). The specific iron oxide nanoparticles are surface functionalized with 2,3-dicarboxypropane-1,1-diphosphonic acid (-DPD) and radiolabeled with gallium-68 (68Ga), resulting in 68Ga-DPD-Fe3O4. RBCs of five healthy individuals are incubated at room temperature for 120 min without and with 68Ga-DPD-Fe3O4 at concentrations 0.1 and 1.0 mg/ml. Optical microscopy (OM) and atomic force microscopy (AFM) are employed to assess detailed information on the overall morphological and geometrical characteristics of the entire cell at the microscopic (10−6 m) level and on the membrane morphology at the nanoscopic (10−9 m) level. In addition, a standard hematology analyzer (HA) is used to obtain complete blood count information. At the microscopic level, the combined OM, AFM and HA data revealed that the overall shape/size characteristics of RBCs were preserved upon incubation with 68Ga-DPD-Fe3O4. However, at the nanoscopic level, the AFM results revealed two different kinds of local deconstructions of the RBCs membrane, termed holes and ulcer-like abnormalities, that were observed in both the DMCA-free and DMCA-incubated samples. Holes did not exhibit any statistically significant difference upon incubation with the 68Ga-DPD-Fe3O4 DMCA. On the contrary, ulcer-like abnormalities exhibited two statistically significant differences upon incubation with the 68Ga-DPD-Fe3O4 DMCA. First, increased percentage of RBCs having at least one ulcer-like abnormality; in DMCA-incubated samples 78.6 ± 11.6% for CDMCA = 0.1 mg/ml and 80.4 ± 11.1% for CDMCA = 1.0 mg/ml, while in DMCA-free samples 61.2 ± 8.4% prior to and 63.6 ± 13.5% after incubation. Second, increased number of ulcer-like abnormalities per RBC; in DMCA-incubated samples 4.26 ± 0.62 for CDMCA = 0.1 mg/ml and 3.99 ± 0.97 for CDMCA = 1.0 mg/ml, while in DMCA-free samples 2.84 ± 0.54 prior to and 2.98 ± 0.50 after incubation. The combined OM, AFM and HA results prove fair hemocompatibility of the 68Ga-DPD-Fe3O4 DMCA with human RBCs, thus documenting its potential use in imaging applications.

AB  - Novel theranostic nanoplatform is expected to integrate imaging for guiding and monitoring of the tumor therapy with great therapeutic efficacy and fewer side effects. Here we describe the preparation of a multifunctional 99mTc–bisphosphonate–coated magnetic nanoparticles (MNPs) based on Fe3O4 and coated with two hydrophilic bisphosphonate ligands, i.e., methylene diphosphonate (MDP) and 1–hydroxyethane-1,1- diphosphonate (HEDP). The presence of the bisphosphonates on the MNPs surface, enabled their biocompatibility, colloidal stability and successful binding of the radionuclide. The morphology, size, structure, surface charge and magnetic properties of obtained bisphosphonate–coated Fe3O4 MNPs were characterized by transmission electron microscopy, X–ray powder diffraction, dynamic light scattering, laser Doppler electrophoresis, Fourier transform infrared spectroscopy and vibrating sample magnetometer. The specific power absorption values for Fe3O4–MDP and Fe3O4–HEDP were 113 W/g and 141 W/g, respectively, indicated their heating ability under applied magnetic field. Coated MNPs were radiolabeled with 99mTc using stannous chloride as the reducing agent in a reproducible high yield (95% for Fe3O4–MDP and 97% for Fe3O4–HEDP MNPs) and were remained stable in saline and human serum for 24 h. Ex vivo biodistribution studies presented significant liver and spleen uptake in healthy Wistar rats after intravenous administration at all examined time points due to the colloidal nature of both 99mTc–MNPs. Results of scintigraphy studies are in accordance with ex vivo biodistribution studies, demonstrating high in vivo stability of radiolabeled MNPs and therefore results of both methods were proved as accurate information on the biodistribution profile of investigated MNPs. Overall, in vitro and in vivo stability as well as heating ability, indicate that biocompatible radiolabeled bisphosphonate magnetic nanoparticles exhibit promising potential as a theranostic nanoagent. © 2019 Elsevier B.V.

AB  - Development of a complex based on iron oxide nanoparticles (IONPs) for diagnosis and dual magnetic hyperthermia/radionuclide cancer therapy accomplishing high yields of radiolabeling and great magnetic heat induction is still a challenge. We report here the synthesis of citric acid, poly(acrylic acid) (PAA) and poly(ethylene glycol) coated IONPs and their labeling with three radionuclides, namely, technetium (99mTc), yttrium (90Y), and lutetium (177Lu), aiming at potential use in cancer diagnosis and therapy. Polyol-synthesized IONPs are a flowerlike structure with 13.5 nm spherically shaped cores and 24.8 nm diameter. PAA-coated nanoparticles (PAA@IONP) showed the best characteristics such as easy radiolabeling with very high yields (&gt;97.5%) with all three radionuclides, and excellent in vitro stabilities with less than 10% of radionuclides detaching after 24 h. Heating ability of PAA@IONP in an alternating external magnetic field showed intrinsic loss power value of 7.3 nH m2/kg, which is one of higher reported values. Additionally, PAA@IONP itself presented no significant cytotoxicity to the CT-26 cancer cells, reaching IC50 at 60 μg/mL. However, under the external magnetic field, they show hyperthermia-mediated cells killing, which correlated with the magnetic field strength and time of exposure. Since PAA@IONP are easy to prepare, biocompatible, and with excellent magnetic heat induction, these nanoparticles radiolabeled with high-energy beta emitters 90Y and 177Lu have valuable potential as agent for dual magnetic hyperthermia/radionuclide therapy, while radiolabeled with 99mTc could be used in diagnostic imaging. AB  - It was reported that novel O,O′-diethyl-(S, S)-ethylenediamine-N,N′-di-2-(3-cyclohexyl) propanoate dihydrochloride (DE-EDCP) displayed in vitro antiproliferative activity on several human and mouse cancer cell lines, which was comparable to that of the prototypical anticancer drug cisplatin. In order to reveal its toxicity profile, acute and repeated-dose toxicity studies were performed in Naval Medical Research Institute (NMRI) Han mice. The intravenous LD50 values of DE-EDCP were found to be 95.3 and 101.3 mg/kg body weight in female and male mice, respectively. In the subacute toxicity study, DE-EDCP was administered intravenously at the doses of 15, 25, and 40 mg/kg/day for a period of 28 days. There were no adverse effects on general condition, growth, feed and water consumption, and hematological parameters. There was a significant increase in urea and alanine aminotransferase in female mice and aspartate aminotransferase and alkaline phosphatase in both genders in 40 mg/kg/day dose-treated group. The histopathological changes confined to the liver and kidney, but in other organs were not found. Satellite group revealed that changes in the kidney and liver were less pronounced, suggesting their reversibility. Interactions with DNA could also be of importance for understanding DE-EDCP toxic side effects. Hyperchromic effect obtained with ultraviolet–visible, suggested electrostatic interactions between DE-EDCP and calf thymus DNA. The toxicity testing of DE-EDCP was conducted to predict human outcomes. © The Author(s) 2018.
