Optical evidence of magnetic field-induced ferrofluid aggregation: Comparison of cobalt ferrite, magnetite, and magnesium ferrite

AB  - Light-transmitting measurements of white light and a consequentially chosen laser beam of 655 nm propagating through citrate- and oleate-coated CoFe 2 O 4 , FeFe 2 O 4 and MgFe 2 O 4 nanoparticles under the influence of an external magnetic field were presented. New experimental settings were developed for the optical study of ferrites’ behavior in ferrofluid with the applied magnetic field strength within the 30–400 mT range. A magnetic field-induced change of light transmittance occurred and a precipitation of all studied samples was obtained. Optical investigations of ferrofluid suspensions revealed that, contrary to the linear aggregates of colloidal CoFe 2 O 4 and FeFe 2 O 4 , spheroidal aggregates were formed in the case of MgFe 2 O 4 . In all three cases, the surface modification resulted in decreased dipole–dipole interactions between ferrite cores, and thus, less precipitates were noticed. All oleate-coated nanoparticles have demonstrated weaker magnetic responses compared to the citrate-encapsulated samples. The aggregation of nanoparticles potentially increases cytotoxicity. Regarding non-linear clustering of MgFe 2 O 4 suspensions, it can be concluded that its excretion from the organism may likely be easier and faster when used in diagnosis and/or therapy. Therefore, more attention should be paid to the lowly toxic MgFe 2 O 4 regarding its medical application. © 2019

AB  - The extensive development of radiopharmaceuticals towards early tumour detection and treatment has increased the demand for new ligands with higher tumour selectivity. Research has been done on the potential of the novel O,O-diethylethylenediamine-N,N-di-3-propanoate (L) ligand as a radionuclide vehicle for tumour targeting. Under alkaline conditions, L hydrolyses and produces half ester ligand (L) and diacid ligand (L), with characteristic donor atom array N, N, O. Ligand L was successfully labelled with Tc-99m at pH=9 by coordination with the octahedral fac-[Tc-99m(CO)(3)(H2O)(3)](+) intermediate, forming the main radioproduct fac-[(TcL)-Tc-99m(CO)(3)] (Tc1). The Tc-99m complex showed a low lipophilic character (log P = 0.48) and low binding affinity to human serum albumin (2.51 +/- 0.48%). In vitro stability studies in saline and human plasma, as well as challenge studies with cysteine and histidine, revealed high stability of the complex during 24 h. Biodistribution studies of Tc1 in female C57BL/6 mice bearing B16/F1 melanoma metastases showed significant tumour uptake: 9.81 +/- 1.19% ID g(-1) in the liver, 5.87 +/- 0.54% ID g(-1) in the lungs and 3.17 +/- 0.33% ID g(-1) in the ovary at 30 min post-injection. Favourable physicochemical properties, satisfactory in vitro/in vivo stability and biodistribution profile in the experimental metastatic melanoma model indicate the possible application of the radiolabelled ligand in tumour diagnosis. AB  - This work presents the simultaneous determination of catechol (CC) and hydroquinone (HQ), employing a modified carbon paste electrode (CPE) with ferrite nanomaterial. Ferrite nanomaterial was doped with different amount of cobalt and this was investigated toward simultaneous oxidation of CC and HQ. It was shown that this modification strongly increases electrochemical characteristics of the CPE. Also, electrocatalytic activity of such materials strongly depends on the level of substituted Co in the ferrite nanoparticles. The modified electrodes, labeled as CoFerrite/CPE, showed two pairs of well-defined redox peaks for the electrochemical processes of catechol and hydroquinone. Involving of ferrite material in the structure of CPE, cause increase in the potentials differences between redox couples of the investigated compounds, accompanied with increases in peaks currents. Several important parameters were optimized and calibration curves, with limits of detection (LOD) of 0.15 and 0.3 mu M for catechol and hydroquinone, respectively, were constructed by employing amperometric detection. Effect of possible interfering compounds was also studied, and proposed method was successfully applied for CC and HQ quantification in real samples.

AB  - In this study, tin fluoride colloid (SnF-c) was prepared, labeled with 90Y and characterized with respect to its physicochemical properties. Particle size of SnF-c, at constant concentration of SnF2, was dependent on pH, concentration of NaF, temperature and time. The particle size of SnF-c decreased with an increase in NaF concentration and a decrease in reaction mixture pH. Radiolabeling yield of 90Y-SnF-c at higher temperature increased and it was greater than 98% for the preparation at 95 oC. Due to high labeling yield and stability, 90Y-SnF-c might be a promising agent for radiosynovectomy.

AB  - There is an increasing interest for the 99mTc labeling of biomolecules by using
bifunctional chelating agents like ethylenediamine-N,N´-di-3-propanoate diethyl
ester (deeddp). To find new ligand, which can be linked to the small biomolecules
and coordinated with [99mTc(CO)3(H2O)3]
+
 precursor, is a challenging task.
Radiolabeling of deeddp with [99mTc(CO)3(H2O)3]
+
 precursor, stability studies and
biodistribution of formed complexes were carried out, including challenge with
histidine. Radiochemical yield of 99mTc(I)-tricarbonyl-deeddp complexes was
higher than 95%. These complexes were stable in vitro and showed a very good
biological behavior. The radiochemical and biological features of the novel
99mTc(I)-complexes, as well as, the nature of the ligands, make them very
promising candidates for labeling of tumor specific biomolecules.
