In vitro radiosensitivity of Fanconi anemia fibroblasts measured by gamma-H2AX foci

Fanconi anemia (FA) is a genetically heterogeneous disease character-ized by bone marrow failure, congenital abnormalities and a significantpredisposition toward malignancies. Fanconi cells are highly sensitiveto DNA crosslinking agents but their response to ionizing radiation isstill unclear. This aimed us to investigate in vitro cellular radiosensitiv-ity of FA cell lines (eight patients) and corresponing controls (six sam-ples) employing enumeration ofc-H2AX foci. Cells were irradiated inexponential growth phase using60Co gamma-rays, radiation dose of 2Gy. Quantification of gamma-H2AX was assessed at various period oftime after irradiation (30 minutes, 2 hours, 5 hours and 24 hours afterirradiation). The mean value of DSBs/cell in unirradiated control fibro-blasts was 1.17 ± 0.43, wheras in unirradiated FA cells DSBs/cell was3.0 ± 2.13. The maximum of induced DSBs/cell was found 30 minutesafter irradiation in both study groups (20.38 ± 3.76 in control,21.99 ± 4.12 in FA cell...s, respectively). The level of radiation-inducedgamma-H2AX foci in FA cells, 2 and 5 hours after irradiation signifi-cantly decrease when compared to controls, whereas after 24 hours FAcells have significantly higher level of DSBs/cell In FA cells. Numerousnecrotic nuclei were found at recovery time of 2 and 5 hours, whereasat longer recovery period of time (24 hours) gamma-H2AX positivemicronuclei were found. This study have shown that in FA cells repairmechanisms operate very inefficiently, leading to elevated apoptosisand micronuclei in growing cell cultures.c-H2AX foci represents verysensitive assay for estimation of intrinsic genome radiosenitivity, anddemonstrate clear difference in behavior of FA and normal cells inresponse to ionizing radiation
