Cardiac Nitric Oxide Synthases and Na+/K+-ATPase in the Rat Model of Polycystic Ovary Syndrome Induced by Dihydrotestosterone

AB  - Nitric oxide synthases (NOSs) and Na+/K+-ATPase are enzymes essential for regular functioning of the heart. Since both enzymes are under insulin and androgen regulation and since insulin action and androgen level were disturbed in polycystic ovary syndrome (PCOS), we hypothesized that cardiac nitric oxide (NO) production and sodium/potassium transport would be deteriorated in PCOS. To test our hypothesis we introduced animal model of PCOS based on dihydrotestosterone (DHT) treatment of female Wistar rats and analyzed protein expression, phosphorylation or subcellular localization of endothelial NOS (eNOS), inducible NOS (iNOS) and alpha subunits of Na+/K+-ATPase in the heart. Obtained results indicate that DHT treatment significantly decreased cardiac eNOS protein level and activating phosphorylation at serine 1177, while inhibitory phosphorylation at threonine 495 was increased. In contrast to expression of eNOS, iNOS protein level in the heart of DHT-treated rats was significantly elevated. Furthermore, cardiac protein level of alpha 1 subunit of the ATPase, as well as its plasma membrane content, were decreased in rats with PCOS. In line with this, alpha 2 subunit protein level in fraction of plasma membranes was also significantly below control level. In conclusion, DHT treatment impaired effectiveness of NOSs and Na+/K+-ATPase in the female rat heart. Regarding the importance of NO production and sodium/potassium transport in the cardiac contraction and blood flow regulation, it implicates strong consequences of PCOS for heart functioning.

AB  - Phosphor materials are of special interest due to their important applications ranging from solid state lightning and displays to biomedicine. Understanding the spectroscopic properties of these materials is important for optimizing their emission for technological applications. Zinc silicate (Zn2SiO4) has been identified as a very suitable host matrix for many rare-earth and transition metal dopant ions with excellent luminescent properties in the blue, green and red spectral zones. Here, we present high resolution measurements of low-temperature photoluminescence of trivalent europium doped zinc silicate particles prepared by combination of so-gel and combustion synthesis route. We observed and analyzed 5D(0) - GT F-7(J) spin forbidden f-f electronic transitions at low temperatures, as well as emission kinetics from D-5(0) level and lifetime-temperature dependence.

AB  - Recently, trivalent rare earth doped materials have received significant attention due to the strong temperature dependence of the fluorescence emission of these materials, which can be useful in temperature sensing. Here, we investigated Y2O3 ceramic powders doped with Yb3+ and co-doped with either Tm3+ or Ho3+. The powders were obtained via spray pyrolysis at 900 degrees C and additionally thermally treated at 1100 degrees C for 24 h. Structural characterization using X-ray powder diffraction confirmed the cubic bixbyte structure. Scanning electron microscopy (SEM) revealed that the particles exhibit a uniform spherical morphology. The up-conversion emissions were measured using laser excitation at 978 nm, resulting in the following transitions: blue emission in the range of 450-500 nm, weak red emission in the range of 650-680 nm and near infrared emission in the range of 765-840 nm for Tm3+, as well as green emission centered at 550 nm and weak near infrared emission at 755 nm for the Ho3+ ions. In addition, the temperature dependence of the fluorescence intensity ratios of different Stark components was analyzed in the range of 10-300 K. Significant temperature sensitivity was detected for several components, with the largest value of 0.097 K-1 related to the intensity ratio of I-536 and I-772 emissions observed for the Y2O3:Yb,Ho powder. AB  - In this paper we report a systematic study on the properties of irradiated SWCNT films by high power, pulsed visible irradiation of OPO laser in the range from 430 nm to 800 nm. Raman analysis showed that metallic and semiconducting SWCNTs were simultaneously affected by irradiation. It was found that laser irradiation had the most prominent effects on SWCNT films at 430 nm wavelength. To our surprise, semiconducting SWCNTs were more affected by irradiation than metallic and as a result several SWCNTs were isolated partially.

AB  - The effect of visible and infrared irradiation on the structure of single wall carbon nanotubes (SWCNTs) is still an insufficiently resolved subject. In this paper, we report a detailed study of two types of SWCNT thin films treated by pulsed laser irradiation and dynamic and isothermal oxidation, respectively. Raman features of treated films were investigated by 532, 633 and 780?nm excitation lines, respectively. It was established that the 532?nm excitation laser line probes the surface of SWCNT films, while the two others probe the bulk of SWCNT films. It was found that during Raman measurements, one type of SWCNTs warms up significantly while the other type does not change temperature. Raman analysis of SWCNTs probed with 532?nm showed that effects of high power laser irradiation on two types of SWCNTs are completely different. Major effects are decomposition, debundlation and photooxidation. It was found that debundlation and removal of defective carbon are much more pronounced in one type of SWCNTs treated with dynamic oxidation. Dynamic oxidation affects mostly thinner SWCNTs that easily burn in air. The only significant effects of isothermal oxidation are the increase of defective amorphous carbon in the first type of SWCNTs and the decrease of D band and debundlation in the second type. AB  - Fine yttrium oxide powders doped with Yb3+. and co-doped either with Tm3+ or Ho3+. were synthesized via spray pyrolysis at 900 degrees C using 0.1 M nitrate precursor. Synthesized powders were additionally thermally treated at 1100 degrees C for 24 h. The characterization was done through X-ray powder diffraction (XRPD), scanning electron microscopy (SEM) and photoluminescent measurements (PL). Generation of cubic bixbyte-structure with space group Ia-3 is confirmed in all samples. Spherical particles with the mean size of similar to 400 nm, generated through volume precipitation of much smaller nanograins, expose certain degree of porosity which alters particle morphology during additional thermal treatment. Powders optical characterizations include measurement of up-conversion emission spectra after excitation at 978 nm, as well as determination of emission lifetimes and up-converted emission intensity dependence on excitation power. Dominant green (F-5(4), S-5(2) - GT I-5(8)) and blue ((1)G(4) - GT H-3(6)) emission is found for Ho3+ and Tm3+ samples, respectively. The amplified emission intensities and enhanced lifetime in thermally treated samples are correlated with the powders morphological and structural changes. (C) 2012 Elsevier B.V. All rights reserved.

AB  - In this research we prepared Zn2SiO4:Eu3+ phosphor nanopowders using a combination of sol-gel and combustion synthesis with the aim to examine the influence of synthesis conditions on the optical properties of the phosphor. As combustion fuels we used polyethylene glycol (PEG) with different average molecular weights, and the combustion was performed in two ways-in a microwave oven and a conventional furnace. Optical properties were examined by photoluminescence spectroscopy and spectra of all samples showed intense red emission, typical for f-f electronic transitions of the Eu3+ ions. Emission decays exhibited classical one exponential behavior at longer times and nonlinear nature at short times, with average lifetimes varied from 0.49 to 0.71 ms between samples. Judd-Ofelt theory was applied to experimental data for the quantitative determination of optical parameters such as Omega(2,4) Judd-Ofelt parameters, radiative and nonradiative transition rates and emission quantum efficiency. Calculated parameters vary moderately between samples prepared with different PEGs and combusted in different manner. (C) 2011 Elsevier B.V. All rights reserved.
